    Notes: Abstract In the present study we have investigated the effect of L-nitro arginine mono methyl ester (L-NAME), an inhibitor of nitric oxide (NO) synthase on Sephadex induced inflammation in the rat lung. Instillation of Sephadex into the airways induced an inflammatory reaction characterized by a long-lasting interstitial oedema, measured as an increase in lung weight, and an influx of inflammatory cells into the airways. L-NAME given s.c. prevented the increase in lung weight following Sephadex instillation. The inactive enantiomer D-NAME had no effect, nor did aminoguanidine which indicates that this effect of L-NAME was mediated by inhibition of the constitutive form of NOS. Treatment with L-NAME did not reduce an established oedema. In contrast, L-NAME tended to enhance the influx of oesinophils into the airways of Sephadex-instilled animals. L-NAME did not have any effect on the development of oedema in adrenalectomized rats or in animals where formation of glucocorticosteroids (GCS) was inhibited with metyrapone. L-NAME did not however, increase plasma levels of corticosterone. The present results indicate that, in this model, inhibition of NO-synthesis has marked anti-inflammatory effects. The underlying mechanism is complex but seems not to involve prevention of overproduction of NO.

    Notes: Abstract. Objective and Design: To correlate the changes in periarticular tissue levels of pro-inflammatory cytokines and endothelin-1 (ET-1) to local pathophysiology in rats with antigen-induced arthritis (AIA).¶Materials: The periarticular soft tissue was excised from sixty-six rats at different stages of AIA.¶Methods: The samples were homogenized and the levels of immune like (LI) reactivities were determined. The levels in the arthritic joints were compared to those in non-arthritic tissue. Statistical significance was determined by ANOVA followed by Fisher PLSD.¶Results: Compatible with a role in an early alarm reaction some mediators (tumor necrosis factor α-LI, interleukin-2-LI and interferon-γ-LI) were elevated prior to the vascular inflammatory activity. The curve for ET-1-LI closely followed the intensity of the inflammation whereas these parameters preceeded the elevation of interleukin-1β-LI.¶Conclusions: Transient waves of different mediators appear during the course of the arthritis. This indicates the presence of active downregulatory mechanisms. Here the model could differ from human rheumatoid arthritis.

    Notes: Abstract Aims/hypothesis. The purpose of this study was to examine whether fetal leptin concentration correlates with severity of chronic or subchronic fetal hypoxia as indicated by increased fetal concentrations of erythropoietin in fetuses of mothers with Type I (insulin dependent) diabetes mellitus.¶Methods. We measured leptin and erythropoietin concentrations in cord plasma and amniotic fluid with radioimmunoassay in 25 pregnancies (gestational age 37.2 ± 1.0 weeks). Fetuses with amniotic fluid erythropoietin over 22.5 mU/ml were classified as hypoxic (n = 9) and those with amniotic fluid erythropoietin below 22.5 mU/ml (n = 16) as non-hypoxic.¶Results. The hypoxic fetuses had significantly higher cord leptin concentrations than non-hypoxic fetuses (median 36.8; range, 12.5–135.1 vs median 16.2; range, 3.7–52.2 μg/l), (p = 0.0066). Cord plasma leptin (n = 25) correlated directly with amniotic fluid erythropoietin (r = 0.727, p = 0.0001), with cord plasma erythropoietin (r = 0.644, p = 0.0005) and with the maternal last trimester HbA1C (r = 0.612, p = 0.0019) and negatively with cord artery pO2 (r = –0.440, p = 0.032), and pH (r = –0.414, p = 0.040).¶Conclusion/interpretation. Fetal leptin concentrations increased concomitantly with erythropoietin during chronic or subchronic hypoxia. This phenomenon could indicate a role for leptin in fetal adaptation to hypoxia.     Notes: Abstract Zygotes and young embryos derived from Fucus vesiculosus collected in the archipelgo of Stockholm in 1990, growing at a salinity of 6 to 7‰ S, were cultured under different salinity conditions and in media of different bromine concentrations. Optimum salinity was 10 to 12‰ S for germination (rhizoid initiation) while apical hair formation showed a broader tolerance curve with an optimum at 8 to 14‰ S. Bromine caused inhibition of early development of F. vesiculosus. At 6‰ salinity a 50% reduction in germination took place at 10.0 mM Br and at 1.25 mM Br only 4.7% of the embryos developed apical hairs, as compared to 32.7% in the control. Bromine toxicity decreased at higher salinities. The results indicate that F. vesiculosus in the Baltic Sea has diverged from its Atlantic progenitors and to some extent acclimated to low salinity. Still, the salinity in the normal environment of the tested population is lower than optimum, leading to a lower degree of germination of zygotes, a lower growth rate of young embryos and probably also a higher sensitivity to additional stress factors such as chemical pollution.

    Notes: Abstract Copper is an active ingredient in many antifouling products, and pleasure boats are estimated to be the major single source of copper pollution in Swedish coastal waters. For this reason, the effects of copper were studied on egg volume, fertilization, germination and development of apical hairs of Baltic Sea Fucus vesiculosus L. Germination was the most sensitive stage and was studied at different concentrations of copper, different salinities and different ages of zygotes. Low concentrations of copper, 2.5 μg Cu l-1, added to natural brackish water before fertilization, adversely affected germination at the ambient, suboptimal salinity of 6‰, suggesting that as little as a doubling of the copper levels in the studied area will severely affect the germination frequency of F. vesiculosus. The addition of 20 μg Cu l-1 caused about 70 to 80% decline in germination at 6‰S but also at 20‰S which is higher than optimum. At a salinity close to optimum (14‰S) no negative effect was noticed on germination when 20 μg Cu l-1 was added. The results suggest that the degree of salinity stress acting upon the zygotes is a more important factor for the response to copper than the influence of salinity on metal availability. When 2.5 to 60 μg Cu l-1 was added to the medium 24 h after fertilization, the zygotes were more resistant, resembling the response of adult marine fucoid tissue.

    Notes: Abstract  Spruce and birch seedlings were grown together in boxes filled with unsterile peat. Both seedlings were colonized by the ectomycorrhizal fungus Scleroderma citrinum. The two plants thus shared a common external mycelium. 15N-labelled ammonium was supplied exclusively to the fungus, while the birch or the spruce plant was continuously fed with 13C-labelled CO2 for 72 h. The carbon and nitrogen transfer rates were strikingly different for birch and spruce seedlings. The mycorrhizal mycelium received carbohydrates mainly from the birch plant and the nitrogen transfer by the fungus to the plants was largely directed towards the birch. Carbon assimilates were also transferred in both directions between birch and spruce; however, there was no conclusive evidence for a net transfer of carbon between the plants.

    Notes: Abstract   Objective:To investigate granulocyte activation, as well as hypoxanthine and free radical production in children during the first day after cardiopulmonary bypass. Design: A prospective study of pediatric patients undergoing either cardiac surgery with a cardiopulmonary bypass or thoracotomy and extracardiac vascular surgery not requiring a cardiopulmonary bypass. Setting: Operative and intensive care units, Children’s Hospital, University of Helsinki, Finland. Patients: Seven consecutive patients undergoing elective correction of a ventricular septal defect and six patients undergoing extracardiac surgery for ligation of a patent ductus arteriosus or repair a coarctation of the aorta. Measurements and main results: Plasma concentrations of myeloperoxidase (140–334 μg/l preoperatively, 460–1692 μg/l at 0.2 h after declamping, 471–1386 μg/l at 0.5 h after declamping) and lactoferrin (77–258 μg/l preoperatively, 533–1783 at 0.2 h, 404–1482 μg/l at 0.5 h) as markers of granulocyte activation, and hypoxanthine (0–5.7 μmol/l preoperatively, 4.3–17.0 μmol/l at 0.2 h, 6.5–17.9 μmol/l at 0.5 h) increased in a biphasic manner at 0.2–0.5 h and 6–10 h postoperatively (all p〈0.05). Expired ethane, as an index of free radical activity, increased at 10 h postoperatively (36–119 pmol/kg per min preoperatively, 72–152 pmol/kg per min, p〈0.05). Conclusion: Granulocyte activation, and hypoxanthine and free radical production occur at least 10 h after cardiopulmonary bypass. In children undergoing open heart surgery, attempts to reduce free radical activity should be extended to the postoperative period.
