    Notes: Summary Fluid samples from brain tumour pseudocysts were examined for the presence of Fibroblast Growth Factor (FGF). Fluids were collected from 6 patients with differentiated low grade gliomas (group A), 3 anaplastic gliomas (group B) and 3 metastases (group C). For FGF assays pooled fluids from group A, B, and C were subjected to affinity chromatography on a Heparin-Sepharose column. Each pool contained endothelial cell mitogenic activity which eluted in the 1.2 M NaCl fraction and to a lesser degree in the 0.6 M fraction of Heparin-Sepharose high affinity chromatography. Mitogenic activity in the 1.2 M NaCl fraction of Heparin-Sepharose chromatography suggests the presence of acidic FGF (aFGF).

    Notes: Abstract— γ-Vinyl GABA (4-amino-hex-5-enoic acid, RMI 71754) is a catalytic inhibitor of GABA-T in vitro. When given by a peripheral route to mice, it crosses the blood-brain barrier and induces a long-lasting, dose-dependent, irreversible inhibition of brain GABA transaminase (GABA-T). Glutamate decarboxylase (GAD) is only slightly affected even at the highest doses used. γ -Vinyl GABA has little or no effect on brain succinate semialdehyde dehydrogenase, aspartate transaminase and alanine transaminase activities. GABA-T inhibition is accompanied by a sustained dose-dependent increase of brain GABA concentration. From the rate of accumulation of GABA it was estimated that GABA turnover in brain was at least 6.5 μmol/g/h. Based on recovery of enzyme activity the half-life of GABA-T was found to be 3.4 days, that of GAD was estimated to be about 2.4 days. γ -Vinyl GABA should be valuable for manipulations of brain GABA metabolism.

    Notes: Abstract— 4-Amino hex-5-ynoic acid (γ-acetylenic GABA, γ-ethynyl GABA, RM171.645), a catalytic inhibitor of GABA transaminase in vitro, induces a rapid, long-lasting dose-dependent decrease of GABA transaminase activity and, to a lesser extent, of glutamate decarboxylase activity in the brains of rats and mice when given by a peripheral route. The GABA concentration in whole brain increases up to 6-fold over control values. The action of γ-acetylenic GABA is relatively specific, as no in vivo inhibition of brain aspartate and alanine transaminase activities could be detected. Furthermore, the amount of radioactive drug bound to the protein fraction of brain homogenate is of the same order of magnitude as that of the GABA transaminase present, as calculated from total GABA transaminase activity, molecular weight and specific activity of the pure enzyme. γ-Acetylenic GABA illustrates the usefulness of a catalytic irreversible enzyme inhibitor in altering neurotransmitter metabolism in vivo.

    Description / Table of Contents: Zusammenfassung Eine Methode zur direkten gaschromatographischen Bestimmung kleiner Mengen leichtflüchtiger organischer Lösungsmittel (Hexan) in Blut und Organen wurde entwickelt. Blut konnte direkt, Organe nach Ultraschallhomogenisation in Wasser in den Gaschromatographen eingespritzt werden. Die direkte Einspritzung wurde durch die Verwendung eines speziellen Einlaßsystems mit auswechselbarem Filter, welcher die festen Blut- und Organbestandteile zurückhielt, ermöglicht. Der methodische Fehler wurde abgeschätzt und Reproduzierbarkeit experimentell bestimmt (7–10%).
    Notes: Abstract A method for the direct gas Chromatographic determination of small amounts of volatile organic solvents (hexane) in blood and tissues was developed. Blood was injected directly into a gas chromatograph, tissues were first homogenized in water by an ultrasonic technique. The direct injection was made possible by utilization of a special inlet system with exchangeable filter which retained nonvolatile components of blood and tissues. The methodical accuracy was estimated and the reproducibility was determined experimentally (7 to 10 %).

    Notes: Summary Mosquito trypsin was purified using a combination of ion exchange and affinity chromatography with the ligand soybean trypsin inhibitor. ThreeAedes and threeAnopheles species were tested, all of which are specialized in the digestion of vertebrate blood. Amino-terminal sequences of HPLC-purified trypsins fromAedes aegypti andAnopheles quadrimaculatus revealed homologies of 30–40% with vertebrate and other invertebrate proteases previously identified as serine-proteases. The purified mosquito trypsins have molecular masses between 25 kDa and 36 kDa, as determined by denaturing polyacrylamide electrophoresis, and are heterogeneous in size and number in the various species. The number of SDS-bands varies between 3 and 6 inAedes and between 1 and 3 inAnopheles. The specific activities, determined with the substrate TAME, range from 240 U/mg inAedes aegypti to 1065 U/mg inAnopheles quadrimaculatus. All mosquito trypsins tested have acidic isoelectric points between pH 3.5 and pH 5.4. No alkaline proteases were detected. Polyclonal antisera againstAedes aegypti andAnopheles albimanus trypsin do not cross-react with bovine trypsin. Cross-reactivity of the two sera with trypsin from six mosquito species suggests the presence of at least 2 enzyme families.

    Notes: Abstract In 1986, Brown and Clemmons (Proc. natl Acad. Sci. USA83 (1986) 3321) showed that platelets contain a substance, platelet-derived growth inhibitor (PDGI), that inhibits in vitro endothelial cell replication. Although platelets are rich in transforming grwoth factor β (TGF-β), PDGI was considered not to be related to TGF-β, on the basis of its reported properties (extraction from platelets at neutral pH, binding to heparin-Sepharose). However, we purified PDGI to near homogeneity and showed that on the basis of HPLC retention behavior, in vitro growth inhibitory activities with several cell types, receptor binding, and immunoneutralization of growth inhibitory activity with specific anti-TGF-β type 1 antibodies, PDGI is most probably identical with TGF-β type 1.
