    Notes: Summary The pigment present in melanosis cerebelli where it is deposited in the dentate nucleus and within the cerebellar granular layer is similar to that of S. nigra as judged from the typical infrared absorption bands near wave numbers 3.450 cm−1 and 1.650 cm−1, respectively. According to electron microscopic findings, this type of melanin arises within the astrocytes of the dentate nucleus. The astrocytic melanosomes exhibit a relatively monotonous substructure reaching diameters of 30 μ and more, and they differ in this respect from the triadic neuronal melanosomes of S. nigra that are strongly reminiscent of lipofuscin granules. These differences may be due to additional components of melanosomes, apparently specific of a particular cell type, and they are in turn accounted for by the dissimilar cellular metabolic make-up of nigral cells and astrocytes, respectively. This does not dismiss the possibility, however, that the melanin pigments of either type of melanosomes are the same or closely related to each other as far as their basic chemical composition is concerned. The cause of the melanosis of the dentate nucleus is not known. Possibly involutional tissue changes governed by endocrine factors among others may play an important part.

    Notes: Summary This is a report of the second known case of a primary malignant granular-cell tumor of the cerebral hemispherical white matter. Two cell types may be distinguished apparently representing different developmental stages of otherwise identical tumor cells. Quantitative histochemical and biochemical studies have shown that tumor cells were containing markedly elevated levels of DNA and RNA. Only few ribosomes and polysomes could be detected, however, by electron microscopy. While cytophotometry disclosed only slightly elevated cytoplasmic proteins, there was a distinct increase in total protein concentration of tumor tissue per unit weight, as measured by conventional techniques. It is suggested that this increase is mainly attributable to structural proteins of cellular and subcellular elements of multiplicating malignant cells. A ratio of RNA to DNA in excess of 1 was found for white matter derived from the central tumor, while the ratios of control tissue were lowered to values far below one due to postmortem changes greatly reducing tissue concentrations of RNA. No detailed characterization of RNA was attempted. The nuclear DNA content of tumor cells reached values equivalent to chromosomal hexadekaploidy. This was in sharp contrast to control data from an Abrikossoff tumor of the oral cavity and from a neurohypophyseal tumorette (“choristoma”), respectively, displaying diploidy only associated with a much lesser increase of cytoplasmic RNA and proteins. Qualitative lipid studies were consistent with a marked active demyelination of the tumor centre as indicated by a severe reduction of cerebrosides and sulfatides and the presence of cholesterol esters. In addition there was a striking loss of phosphatidylethanolamine and a lesser one of sphingomyelin of white matter of both the tumor-stricken and the contralateral unaffected hemispherical regions, possibly suggesting a generally disturbed metabolism of myelin. It is not clear whether these general changes were resulting from the presence of the unilateral tumor or from precocious cerebral involution.

    Notes: Summary Melanosomes and isolated melanosomal fragments (melanin particles) originating from gangliocytes (substantia nigra), astroglia (melanosis cerebelli), and melanocytes (melanotic meningeoma; metastases of melanoblastoma; melanosis thalami of the goat) were compared with synthetic melanins prepared from dopamine and serotonin, respectively. Samples were examined by electron microscopy, X-ray diffraction analysis according to Debye-Scherrer and by infrared spectrophotometry, and the results were evaluated with regard to characteristic features as they may relate to specific cell types or chemical structures. On electron microscopy all three types of melanosomes could be differentiated unequivocally as could the two synthetic melanins. Thus, there were similarities between synthetic melanin from dopamine and the gliogenic melanins of the cerebellum; the synthetic melanin from serotonin resembled melanin of melanocytes. X-ray diffraction analysis yielded 2-4 Debye diffraction rings with all human and synthetic samples, suggesting short range orders between 3.8 to 5 Å the sample obtained from a goat with thalamic melanosis showed a specific reflex pattern. While diffraction patterns of some melanins were partially identical, in particular that of melanin from dopamine and melanin of substantia nigra and dentate nucleus, respectively, they were different for the various melanocytic melanins. Further investigations are required to determine whether these differences are due to disparities in basic chemical structures or conformations or else, to particular compositional features of the various types of melanocytes as they arise from benign or malignant tumors or a specific species. Infrared spectrophotometry at higher wave numbers revealed the well known patterns of melanins, which are not, however, very suitable, for their further differentiation. At lower wave numbers (‘fingerprinting’) melanin of substantia nigra and the glial melanin in melanosis cerebelli yielded additional absorption bands of identical configuration. In contrast to melanin from dopamine, melanin from serotonin exhibited a closely similar absorption pattern in this spectral range, suggesting that the neuroectodermal melanins may contain a component possibly arising from serotonin.

    Notes: Summary Alcohol was administed in a vitaminsupplemented liquid diet (Stardit) to Wistar rats 4 weeks before mating and during pregnancy. Of the daily calories 38–40% were supplied by alcohol and by isocaloric sucrose in the controls. Offspring of alcohol drinking dams showed a reduced birth weight and gained weight less rapidly than the controls. On day 3, 4, 7, 12, 17 and 21, experimental animals and controls were perfused for histological and electron microscopic investigations. Morphometric analysis of the cerebella (Fol. 5 and Fol. 8) showed no difference in the number of Purkinje cells. However, on day 7 the Purkinje cell nuclei of experimental pups were significantly smaller. This difference disappeared at day 12. Electron microscopic investigations in the 4, 7, and 12 days old experimental animals revealed a delayed cytoplasmic maturation of Purkinje cells which mainly involved the rough endoplasmic reticulum. At 17 days, there was no difference between the two groups. These data are discussed in relation to alcohol-induced metabolic changes in the brain.

    Notes: Summary Sixty tumors of the central and peripheral nervous system and seven brain metastases of extracranial carcinomas were examined using the peroxidase-antiperoxidase method to study the expression of myelin basic protein (MBP), myelin-associated glycoprotein (MAG) and the HNK-1/Leu-7 epitope. The immunocytochemical findings were compared with and correlated to Western blot results. None of the tumor types, including oligodendrogliomas, neurinomas and neurofibromas, expressed MAG and MBP, whereas myelin sheaths and their remnants within the tumors yielded specific immunoreactions. In contrast, the HNK-1/Leu-7 antibodies labelled the majority of the tumors tested including oligodendrogliomas and Schwann cell tumors. As demonstrated by Western blot experiments the HNK-1/anti-Leu-7 antibodies exhibited positive reactions with diverse polypeptides both in tumors and in non-neoplastic brain tissue at positions not corresponding to MAG. This suggests that the epitope recognized by HNK-1/Leu-7 antibodies is shared by a variety of unrelated proteins in normal and neoplastic tissues. Our results strongly indicate the absence of detectable amounts of MBP and MAG in oligodendrogliomas and Schwann cell tumors. The immunomorphological and immunochemical findings clearly showed the wide distribution of the HNK-1 epitope within different tumor types of the central and peripheral nervous system. In conclusion, the data demonstrate that specific cell markers for human oligodendrogliomas and Schwann cell tumors are still lacking.

    Notes: Summary The influence of moderately reduced arterial oxygen tension (aPO2 of about 45 Torr) on the metabolism and the blood flow of the brain was tested in 20 anaesthetized, artificially ventilated normotensive, normocapnic beagle dogs. It is demonstrated that the decrease in systemic oxygen delivery to the brain is countered by an appropriate increase in flow (CBF being 60.3 ml/100 g min at normoxia and 84.5 mg/100 g min m hypoxaemia) which maintained the cerebral oxygen consumption unchanged (CMRO2 3.80 versus 3.32 ml/100 g min). The cortical tissue content of energy-rich phosphates such as ATP, ADP, AMP, and phosphocreatine was also found to be unaltered. Neuropathological examinations excluded any hypoxic cell damage. This reactive vasodilatory reaction of the cerebral vessels is apparently a sensitive regulatory process which protects the brain against marked oxygen lack. However, a normal carbohydrate metabolism is not restored by this cerebrovascular mechanism. For, significantly increased CMRlactate (0.32 versus 1.46 ml/100 g min) indicated raised cerebral glycolysis, and the tissue metabolites of glucose suggested an increased glycolytic flux in the brain. It is concluded that in moderate arterial hypoxaemia, which is not uncommon in clinical practice, cerebral blood flow plays an effective homeostatic role in preventing a disturbance of the energy metabolism of the brain.

    Notes: The hypoxic tolerance and the cerebral metabolic rates (CMR) of young adult mice (20 to 25 g, 4 to 5 weeks old) and adult mice (30 g and above, 6 to 7 weeks old), respectively, were determined and their interrelationship was evaluated. CMRs increased from 25 mmol - P/kg.min to 38 mmol/kg.min as the animals grew older from young to full adulthood. Concurrently the tolerance to aerogcnic hypoxia (5% O2-95%j N2) declined. The effects of hypoxia on the cerebral energy metabolism were greater in adult than in young adult animals. It is concluded that the full metabolic maturation of the brain is reached in adult animals only. They become more dependent on an adequate oxygen supply as the aerobic activity of the energy metabolism of the brain is further increasing.Hypoxic gasping occurred while the pool of cerebral energy reserves was still far from being depleted. A failure to utilize energy reserves rather than their exhaustion is suggested as the ultimate cause of death from hypoxia. An acid-soluble form of glycogen or related polyglucan was found in addition to the usual amounts of insoluble glycogen. It was utilizcd rapidly during hypoxia and ischaemic anoxia and it may, therefore, constitute an additional source of carbohydrate substrates in thc brain.

Angiotensin potenziert den Calciumwert, erhöht das Magnesium und lÄ\t Kalium nahezu unverÄndert. Die im Bereich der linksventrikulÄren Innenschichten ausgebildeten „Catecholaminnekrosen“ finden sich nach DHT-PrÄmedikation ubiquitÄr. Strophanthin verhindert den massiven, durch DHT- und Angiotensinmedikation induzierten Calciumstieg, wÄhrend es bei der kombinierten Gabe von DHT und Noradrenalin keine Wirkung entfaltet. Myofibrillen und Mitochondrien sind verÄndert, Calciumablagerungen sind nachweisbar, charakteristische Unterschiede fehlen.
    Notes: Summary Changes in certain tissue electrolytes brought about by injecting rats with noradrenalin (NA), angiotensin or dihydrotachysterol (DHT) were compared with the concomitant ultrastructural alterations of cardiac muscle. High doses of NA or angiotensin given singly or together with ouabain were found to lower the overall myocardial calcium concentration without accompanying structural lesions. When calcium was increased more than tenfold by the administration of DHT, electronmicroscopy revealed altered myofibrils and mitochondria. Giving DHT along with NA or angiotensin not only preserved their calcium-lowering effect, but resulted in a massive increase in tissue calcium, with magnesium levels also rising, whilst the tissue content of potassium remained almost unchanged. Following NA administration the necrotic areas, known to be confined to the inner endocardial layer after catecholamine administration, pervaded the entire cardiac muscle mass. Ouabain prevented the massive rises of tissue calcium induced by DHT given together with angiotensin but was ineffective when added to the combination of DHT and NA. In either case, both myofibrils and mitochondria displayed ultrastructural alterations although distinctive features were lacking. The data suggests that DHT acts on cellular calcium, not only by mobilising calcium from osseous structures but also by interacting with intracellular calcium homeostasis. In addition to overstimulating contractile elements, calcium is thought to exert its noxious effect in a more generall way, e.g. by binding to surface membranes, thus affecting the normal functioning and eventually the structure of mitochondria and other organelles besides myofibrils.

    Notes: Zusammenfassung Bei Mäusen wurde die Verteilung von Kreatin-1-14C zwischen Plasma und Gehirn nach einer einmaligen i.v. Gabe von Kreatin-1-14C untersucht. Ein Fliessgleichgewicht stellte sich 36–48 h nach Verabreichung der Substanz ein, wobei Kreatin gegen ein Konzentrationsgefälle aus dem Plasma in das Gehirn übertritt. Während dieser Zeit war die Radioaktivität im Gehirngewebe um das 20fache höher als im Plasma.
