    Notizen: Seed storage protein content of durum wheat (Triticum turgidum var. durum) has an important effect on nutritional value and pasta-making characteristics. The objective of this study was to determine by association with genetic markers the number, chromosomal location, and magnitude of effect of quantitative trait loci (QTLs) controlling protein concentration in kernels. A set of 65 recombinant inbred lines (RIs) was developed by single seed descent from a cross between cultivated durum wheat cv. ‘Messapia’ (low protein content) and accession MG4343 of the wild tetraploid wheat var. dicoccoides (high protein content). This population was characterized for eight morphological, six storage protein, one isozyme and 124 RFLP loci. Field trials were conducted in one location in 1993 and two locations in 1994. QTLs were mapped by regression analysis on each marker locus for each location and for the average across environments. A total of six putative QTLs were located on chromosome arms 4BS, SAL, 6AS, 6BS and 7BS. The number and size of QTLs detected varied across environments. The marker with the highest r2 value per QTL in each environment and across environments was chosen for a multiple linear regression analysis, which explained 49.2- 56.4% of the phenotypic variation for protein content. Only some of the markers were found to be negatively associated with plant grain yield and/or seed weight in one or two of the environments.

    Notizen: The recessive dwarfing alleles of rye (Secale cereale L.), ct1 and ct2, caused a 35–55% reduction in the length of leaf 2 compared with corresponding tall lines grown at both 10°C and 20°C. The dwarf lines were 45–50% as responsive to applied GA3 as the tall lines at 20°C but the absolute GA-responsiveness of the dwarfs was greater at 10°C than at 20°C. There was no significant difference in the contents of GA19, GA20, GA29, GA1, GA3 and GA8 in the leaf extension zone of tall and dwarf seedlings grown at 20°C. It was concluded that the mechanism whereby GA homeostasis is maintained is functional in both tall and dwarf lines despite marked differences in leaf extension rate. The recessive rye mutations may cause loss of function late in the GA-cell elongation pathway or, alternatively, indirectly affect GA-responsiveness in vegetative tissues. The genetic and physiological evidence indicates that ct1 and ct2 are unrelated to the GA-insensitive Rht genes in hexaploid bread wheat.

    Notizen: Abstract The molecular forms of β-amylase present in developing, mature, germinating and malted grains of barley (Hordeum vulgare L.), and in vegetative tissues, have been studied using Western-blot analyses and isoelectric focusing of isoenzymes. Five isoforms with different relative molecular masses (Mrs) could be recognised. The major isoform present in the mature grain, called isoform B, had an Mr of about 60 000. This was converted on malting or germination to two lower-Mr forms called C and D. Previous work (R. Lundgard and B. Svensson, 1986, Carlsberg Res. Commun. 51, 487–491) has shown that these result from partial proteolysis of isoform B. Isoenzyme analyses showed complex patterns of bands, with pIs between about 5.0 and 6.0. Two allelic types were present in the eight lines. A number of new bands with a range of pIs appeared during germination and malting. An isoform with the same Mr as D and a minor low-Mr isoform (E) were present in young developing whole caryopses (8–12 d after anthesis), but not in older developing endosperms (14–21 d after anthesis). Isoenzyme analyses also showed different patterns of bands in these two tissues, while hybrid-dot analyses indicated the presence of separate populations of mRNAs. It is suggested that the early endosperm isoforms (D and E) are “green” β-amylases present in the pericarp and-or testa of the young caryopses. Roots but not shoots or leaves also contained an isoform with the same Mr as D, although the pattern of isoenzymes differed from that present in the seed tissues. The fifth isoform, A, was a diffuse high-Mr form present in small amounts in all seed and vegetative tissues, and may correspond to a constitutively expressed form. These multiple molecular forms of β-amylase are discussed in relation to the recent report that β-amylase is encoded by two structural loci, with a total copy number of two to three per haploid genome (Kreis et al, 1988, Genet. Res. Camb. 51, 13–16).

    Notizen: Abstract  Quantitative trait loci (QTLs) controlling the morphological differences between pearl millet (Pennisetum glaucum ssp. glaucum) and its wild ancestor (Pennisetum glaucum ssp. monodii, form mollissimum) were investigated in a cultivated/wild F2 population by means of RFLP markers. The most critical adaptive changes resulting from the domestication process involved the spikelet structure: non-shedding seeds with reduced bracts and bristles and long involucral pedicel. Major differences also concerned characters describing the plant architecture, phenology and spike sizes. Many morphological differences could be attributed to the effect of a small number of loci with relatively large effects. These loci are mainly concentrated on four linkage groups (2, 5, 6 and 7). The loss of shedding ability, due to the absence of a functional abscission layer, is controlled by a single locus on linkage group 6 (al6). Genetic control of the other spikelet traits involved factors with large effects which are located in the region of linkage group 6 close to al6 and to an esterase gene, Esterase-E. Moreover, QTLs with large effects on plant and spike morphology traits such as plant height, number of spikes and weight of the spike were also mapped on linkage groups 6 and 7. This strong linkage of factors in the domestication syndrome may be involved in the maintenance of the phenotypic identity of wild and cultivated populations in sympatry. This result also brings new arguments in the understanding of the domestication process of this allogamous crop.

    Notizen: Abstract  Comparative genetic maps were constructed of the pearl millet genome with foxtail millet and used to describe the homoeology between the genomes of pearl millet, foxtail millet and rice. Despite the close taxonomic relationship of pearl and foxtail millet, their genomes were highly, rearranged. A comparison of the millet and rice genomes indicated that most of these rearrangements were likely to have taken place in pearl millet. Two duplications were identified in pearl millet. A duplication between the distal segments of linkage groups 1 and 4 corresponds to the ancient duplication previously identified between rice chromosome arms 11S and 12S and foxtail millet chromosomes VII and VIII. The other putative duplication, also between regions of linkage groups 1 and 4, is likely to be species-specific. The exploitation of the comparative maps in pearl millet research is discussed.

    Notizen: Summary The effects of the ‘Tom Thumb’ Rht3 dwarfing gene in wheat on plant height, yield components and grain quality in field trials of random inbred lines from two ‘Minister Dwarf’ (Rht3) x rht3 crosses are described. Limited genetic increases in Rht3 plant height, up to 70 cm, were achieved by incorporating genes from the extremely tall variety ‘April Bearded’. Amongst lines from the higher yielding cross, the dwarfing gene did not reduce grain yield but was associated with an increase in grain number of over 25%. Although Rht3 reduced grain protein content by up to 0.8%, breadmaking quality, as measured by SDS sedimentation values and baking tests, was not affected. A controlled environment experiment with near-isogenic Rht3 and rht3 lines indicated that reduced grain size in the dwarfs is a competitive response to the increase in fertility rather than a primary effect of the dwarfing gene.

    Notizen: Summary Analysis of grain esterase isozymes in Chinese Spring aneuploid genotypes by IEF confirmed that genes on the long arms of chromosomes 3A, 3B and 3D (Est-5) control the production of 19 isozymes. Allelic variants have been found for the isozyme pattern controlled by each chromosome. Segregational data involving null alleles and complex phenotypic differences indicate that the wheat grain esterases are encoded by three compound and probably homoeoallelic loci, each capable of producing at least six different isozymes. In a sample of 138 hexaploid genotypes, seven alleles were distinguished.

Two genes which control the production of three of these isozymes were located on chromosome arms 1BS and 1DS by nullisomic analysis. These loci probably form part of a homoeoallelic series and have been designated Per-B1 and Per-D1 respectively. Analysis of chromosome 1B short arm terminal deletion stocks indicated that the Per-B1 locus is located between the nucleolar organiser region and another isozyme marker, Hk-B1. Two variant leaf peroxidase phenotypes were distinguished in a small sample of hexaploid wheat varieties. Analysis of wheat-alien addition and substitution lines identified homoeologous loci in rye (Per-R1) and barley (Per-H1).
