    Notes: A spectrometer for inverse photoemission in the vacuum ultraviolet range is described. A spherical grating with an acceptance of f/4 is used in normal incidence. Two position-sensitive detectors allow the registration of spectra covering the whole range of photon energies from 8 to 28 eV in parallel. The optical resolution is 18 A(ring) for the Lyman-α line of hydrogen. A space-charge-limited electron gun with an energy spread of 0.25 eV is used to excite inverse photoemission spectra. The large acceptance angle of the grating allows one to measure spectra with high efficiency and low background level.

    Description / Table of Contents: Summary In autoradiographic studies after the local application of H3-benzpyrene and H3-methylcholanthrene it could be demonstrated that the carcinogens themselves or their labelled breakdown products were bound firmly into the proteins, ribonucleic acids, and deoxyribonucleic acids of the various cells of the mouse epidermis. The pattern oflabelling of the skin studied, which in part was hyperplastic, in part normal, corresponded greatly with the pattern of incorporation which was obtained after the application of labelled amino acids. As the experimental period increased, there was a displacement of the radioactive labelling towards the surface of the stratifield squamous epithelium. Consequently, it seemed most probable that the carcinogens were eliminated by means of the cellular desquamation during the process of physiologic regeneration. Since labelled carcinogens were not demonstrable in the basal cell layers from four to ten days after single applications, it must be assumed that the carcinogenic action of the substances investigated does not depend on a lasting uptake of substances, but on the irreversible changes of certain cellular structures.
    Notes: Zusammenfassung In autoradiographischen Untersuchungen nach lokaler Applikation von 3H-Benzpyren und 3H-Methylcholanthren konnte nachgewiesen werden, daß die Carcinogene selbst oder aber ihre markierten Abbauprodukte fest in Protein, Ribonucleinsäuren und Desoxyribonucleinsäuren der verschiedenen Zellelemente der Mäuseepidermis eingebaut werden. Das Markierungsmuster der untersuchter teils hyperplastischen, teils normalen Mäusehaut entspricht weitgehend dem Incorporationsmuster, welches nach Applikation markierter Aminosäuren erhalten wird. Mit zunehmender Versuchszeit erfolgt im mehrschichtigen Plattenepithel eine Verschiebung der radioaktiven Markierung nach der Gewebsoberfläche hin. Die Eliminierung des Carcinogens erfolgt somit mit größter Wahrscheinlichkeit durch die im Zuge der phyliologischen Regeneration stattfindende Zellabstoßung. Da 4–10 Tage nach einmaliger Applikation auch in den basalen Zellagen keine markierten Carcinogene mehr nachweisbar sind, muß angenommen werden, daß die carcinogene Wirkung der untersuchten Substanzen nicht auf einem dauernden Einbau der Substanzen, sondern auf der irreversiblen Veränderung bestimmter Zellstrukturen beruht.

    Description / Table of Contents: Summary In radioautographic investigations the patterns of distribution and incorporation of non-carcinogenic H3-1,2,3-Dibenzanthracene and H3-1,2-Benzpyrene was compared with that of the carcinogenic H3-3,4-Benzpyrene and H3-20-Methyl-cholanthrene in mouse skin. The non-carcinogenic substances showed much less uptake, their ability to enter the mouse skin was diminished and they were eliminated faster. There were no differences of the pattern of uptake, however. The pattern of distribution of H3-3,4-Benzpyrene in rat skin was about equal to that of mouse skin. The elimination of radioactivity, however, ensued faster, for 24 h after the last application a large amount of radioactivity had disappeared. The epidermis was practically free of labelling 48 h after application, although the amount of labelled carcinogen used and the initial intensity of incorporation were equal in rat and mouse. The inflammatory reactions caused by the toxic effect of carcinogens appear to be responsible for the poorer carcinogenesis by rat skin after local applications. After H3-3,4-Benzpyrene and H3-20-Methylcholanthrene were applied on mouse skin, some radioactivity could be demonstrated in the liver and kidneys up to 4 days. The incorporation-pattern of the investigated carcinogenic hydrocarbons corresponds closely with the incorporation-pattern after application of labelled amino acids.
    Notes: Zusammenfassung In autoradiographischen Untersuchungen an der Mäusehaut wurde das Verteilungs-und Einbaumuster nichtcarcinogenen 3H-1,2-Benzpyrens und 3H-1,2,3,4-Dibenzanthracens mit dem der carcinogenen Kohlenwasserstoffe 3,4,-Benzpyren-H3 und 20-Methylcholanthren-H3 verglichen. Die nichtcarcinogenen Stoffe ergaben eine wesentlich geringere Silberkorndichte im Autoradiogramm und zeigten neben einem geringeren Eindringvermögen in die Mäusehaut eine raschere Eliminierung. Unterschiede im Einbaumuster konnten aber nicht entdeckt werden. Das Verteilungsmuster von 3,4-Brenzpyren-H3 in der Haut der Ratte glich im wesentlichen dem der Maus. Die Eliminierung der Radioaktivität verlief aber rascher, so daß schon 24 Std nach der letzten Tropfung ein großer Teil der Silberkornbelegung verschwunden und die Epidermis nach 48 Std praktisch frei von Markierung war, obwohl die applizierte Radioaktivitätsmenge und die anfängliche Intensität der Schwärzung bei Ratte und Maus die gleiche waren. Die durch die toxische Wirkung des Carcinogens hervorgerufenen entzündlichen Reaktionen scheinen für die schlechte Cancerisierbarkeit der Rattenhaut bei lokaler Applikation mit verantwortlich zu sein. Nach lokaler Applikation von 3,4-Benzpyren-H3 und 20-Methylcholanthren-H3 auf die Mäusehaut konnte ein Teil der Radioaktivität bis 4 Tage nach der letzten Tropfung in der Leber und den Nieren nachgewiesen werden. Das Verteilungs-muster entspricht weitgehend dem Inkorporationsmuster, welches nach Applikation mit markierter Aminosäuren erhalten wird.

    Notes: Abstract. Bronchial provocation studies performed in our research center have consistently demonstrated airway hyperresponsiveness to both inhaled methacholine and histamine in subjects with chronic cervical spinal cord injury (SCI). More recently, we reported that the airways of such subjects maintained on chronic baclofen (γ-aminobutyric acid) therapy were not hyperreactive to inhaled methacholine. In this study we determined whether baclofen also blocks the effects of the bronchoprovocative agent histamine in subjects with cervical SCI. Twenty-four male subjects with cervical SCI participated in this study; 14 were maintained on oral baclofen, and 10 served as age-matched controls. The subjects were challenged with increasing concentrations of aerosolized histamine until either a 20% fall in forced expiratory volume in 1 s (FEV1) from baseline (defined as PC20) was observed, or a maximum of 25 mg/ml histamine was administered. We found that 11 of the 14 baclofen subjects (78.5%) and 8 of the 10 control subjects (80%) responded (PC20 〈 8 mg/ml) to the histamine challenge. Mean PC20 values among responders in the baclofen (PC20= 2.91 ± 2.3) and control (PC20= 2.18 ± 1.9) groups did not differ significantly. Because histamine acts directly on histamine receptors and indirectly on cholinergic pathways, our findings that baclofen blocks bronchoconstriction due to inhaled methacholine, but not that due to histamine, suggests that hyperresponsiveness in subjects with cervical SCI may be secondary to nonspecific airway hyperreactivity.

    Notes: Summary We have carried out a light microscopical study of Müller cells in the retinae of rats with inherited retinal dystrophy (Royal College of Surgeons rats). Isolated retinae of both control and Royal College of Surgeons rats were exposed to a Procion Yellow solution which is taken up selectively into Müller cells. The shape of the cells was then studied by confocal microscopy. Enzymatically isolated Müller cells were studied immunocytochemically with antibodies against glial fibrillary acidic protein, cathepsin D, β-amyloid precursor protein, bcl-2 protooncogene product, and glutamine synthetase. Müller cells from RCS retinae were shorter than those from control retinae, and showed a coarse hypertrophy of their distal (sclerad) processes. In Müller cells isolated from the retinae of Royal College of Surgeon's rats, the expression of glial fibrilliary acidic protein, cathepsin D, β-amyloid precursor protein and bcl-2 protooncogene product was increased, and the expression of glutamine synthetase was reduced. Obviously, loss of neighbouring neurons leads to major alterations of both the shape and metabolism of Müller cells. The expression of enzymes that serve functional glio-neuronal interactions, such as glutamine synthetase, seems to be down-regulated, whereas proteins involved in cell reconstruction (cathepsin D), cell repair (possibly β-amyloid precursor protein), and protection against apoptotic cell death (bcl-2 protooncogene product), are up-regulated, together with the ‘pathological marker’ glial fibrilliary acidic protein.

    Notes: Summary Two fluorescence techniques to study metal-humic interactions are presented. In the first technique, Lanthanide Ion Probe Spectroscopy (LIPS), the humic samples are titrated by Eu3+ ions. The ratio of the intensities of two emission lines of Eu3+, R=I592/I616, is used to estimate the amount of bound and free species of the probe ions. The titration plot is presented as R versus the logarithm of total added Eu3+. In the second technique, fluorescence quenching of the humic material by Cu2+ is used to produce titration curves of intensity versus the logarithm of total added Cu2+. The two techniques are used in conjunction with a model that treats the various ligands in humic substances as continuous distributions of binding sites in which individual ligand concentrations are normally distributed with respect to the individual stability constants for metal binding. The model includes the effects of pH, ionic strength, and competing metal ions. The parameters of the model are estimated by fitting the spectral titration data to the calculated titration plot. Some simulation and experimental data are presented and discussed.

    Description / Table of Contents: The light absorption of the following compounds and mixed phases has been investigated: CuF2, MCuF3, M2CuF4 (M = alkali metal), CsCu2F5, KMg1-xCuxF3, KZn1-xCuxF3, Mg1-xCuxF2, Zn1-xCuxF2. The reflexion spectra show a strong absorption band with two maxima, even if the site symmetry of Cu2+ is cubic, as in KMg1-xCuxF3 and KZn1-xCuxF3 (Jahn-Teller effect). The spectra of some phases show a third band at larger wave length, compatible with the assumption of a tetragonal field around Cu2+. In the system CsF/CuF2 the new compound CsCu2F5 is found.
    Notes: Es wurde die Lichtabsorption des Cu2+ in folgenden Verbindungen bzw. Mischphasen in Remission untersucht: CuF2, MCuF3, M2CuF4 (M = Alkalimetall), CsCu2F5, KMg1-xCuxF3, KZn1-xCuxF3, Mg1-xCuxF2 und Zn1-xCuxF2. Alle Spektren zeigen eine hohe Bande mit zwei Maxima, auch dann, wenn die Site-Symmetrie des Cu2+, wie in den kubischen Mischphasen KMg1-xCuxF3 und KZn1-xCuxF3 (Perowskit-Typus), kubisch ist. Dies ist eine Folge des Jahn-Teller-Effektes. Die Spektren einiger Phasen weisen noch eine dritte, langwellige Bande auf, wie es bei einer tetragonalen Feldsymmetrie am Ort des Cu2+ zu erwarten ist. Im System CsF/CuF2 wurde die neue Verbindung CsCu2F5 aufgefunden.
