Zusammenfassung Dimethylnitrosamin (I), Methyl-tert. butylnitrosamin (II), Nitrosopiperidin (III) und Nitrosomorpholin (IV) als ihre 0-Äthyl fluoroborate wurden auf Toxicität und carcinogene Wirkung untersucht. Die akute Giftigkeit war geringer als die des entsprechenden Nitrosamins und der elektrophilen Reaktivität etwa proportional; II war bei weitem die reaktivste und giftigste Verbindung. — Carcinogenese-Versuche wurden mit II und IV bei subcutaner Injektion durchgeführt. Bei II war das Ergebnis negativ, wahrscheinlich weil die verträgliche Dosierung von 2 mg/kg (33% der LD 50) einmal wöchentlich nicht ausreichte. Dagegen erzeugte IV nach wöchentlicher Gabe von 40 mg/kg (10% der LD 50) bei allen 15 behandelten Ratten lokale Sarkome an der Injektionsstelle, und sogar nach einmaliger Dosis von 350 mg/kg bei 5 von 10 Tieren. Nach diesen Ergebnissen ist IV ein „direkt” alkylierendes Carcinogen. Die Freisetzung von Nitrosomorpholin, das besonders schnell Leberkrebs erzeugt, kann daher praktisch ausgeschlossen werden. Die möglichen Reaktionsmechanismen von Äthoxydiazenium-Verbindungen werden dargestellt.

Summary Dimethylnitrosamine (I), methyl-tert.-butylnitrosamine (II), nitrosopiperidine (III) and nitrosomorpholine (IV) as the corresponding 0-ethyl fluoroborates have been tested for toxicity and carcinogenicity. The acute toxicity was lower than that of the respective free nitrosamines, and in direct proportion to the electrophilic reactivity, II being by far the most reactive and toxic compound. — Carcinogenicity tests were performed with II and IV at subcutaneous injection. With II the outcome was negative, probably because the tolerated dosage of 2 mg/kg (33% of LD50) once per week was insufficient. On the other hand, IV at a weekly dosage of 40 mg/kg (10% of LD50) induced local sarcomas at the site of injections in all 15 treated rats, and even after a single dose of 350 mg/kg in 5 out of 10 rats. No tumors in remote organs were observed. From these results it follows, that IV is a “direct” alkylating carcinogen, and that the liberation of nitrosomorpholine, a very potent liver carcinogen, can practically be excluded. The potential reaction mechanisms of ethoxydiazenium compounds are discussed.
