    Notes: Abstract Forty bacterial isolates from the effluents of a gelatin factory (Jabalpur, India) were screened for protease activity and the two most potent producers were identified as Bacillus laterosporus and a Flavobacterium sp. The enzymes of both isolates were optimal at pH 8 and 60°C, with maximum activity after 90 min. The enzyme activity of B. laterosporus was suppressed by Fe2+, Mg2+, Mn2+ and Zn2+ ions but was enhanced by Ba2+ and Ca2+. That of Flavobacterium sp. was suppressed by Mg2+ and Mn2+ ions but enhanced by Ba2+, Ca2+ and Fe2+. The enzyme activity of the former was strongly inhibited by KCN, whereas that of the latter was only slightly inhibited by 8-hydroxyquinoline.

    Notes: Abstract Twenty-six different species of keratinophilic fungi were examined to determine their ability to utilize free cystine. Of the fungi tested, the majority metabolized free L-cystine in a glucose-peptone culture medium. Cystine was used as source of sulfur, and carbon and nitrogen as well. Excess sulfur was excreted into the culture fluid, as thiosulfate and sulfate, following oxidation. The rate of cystine oxidation varied with the different fungal strains, but was maximal for Graphium penicilloideus (88.5%). Low quantities of thiols were found in the medium. Cystine oxidation and inorganic thiosulfate excretion were found to correlate significantly (r = 0.94).

    Notes: Summary Nitrogen and sulphur requirements ofColletotrichum inamdarii Lal isolated from the leaves ofCarissa carandas L. have been studied. DL-serine, L-asparagine and L-phenylalanine have been found to be of good nitrogen source followed by potassium nitrate, calcium nitrate, magnesium nitrate, DL-alanine, ammonium nitrate, glutamic acid, ammonium sulphate, DL-valine, aspartic acid, ammonium chloride, ammonium hydrogencarbonate, L-histidine and potassium nitrite. There was no growth in the absence of nitrogen. Sporulation was excellent on calcium nitrate and sodium nitrate, Very good on DL-serine, potassium nitrate, and magnesium nitrate. Good on L-asparagine, L-phenylalanine and ammonium oxalate. Fair on DL-alanine, DL-leucine, ammonium sulphate, DL-valine, ammonium chloride and L-histidine whereas poor on glutamic acid, aspartic acid, ammonium tartarate and ammonium nitrate. Few spores were observed on ammonium hydrogencarbonate but potassium nitrite did not show any sporulation. Amongst the sulphur compounds sodium bisulphate gave the best growth and good sporulation, followed by sodium thiosulphate, magnesium sulphate, ammonium sulphate and potassium sulphate. Thiourea gave negligible growth whereas it failed to grow on zinc sulphate and potassium persulphate.

Glucose was the predominant sugar in all the cases.

    Notes: Abstract The ability of five keratinophilic fungi, i.e., Chrysosporium indicum, Geotrichum candidum, Gymnoascoideus petalosporus, Scopulariopsis brevicaulis, and Talaromyces trachyspermus, to digest human hair keratin in stationary culture has been studied. Degradation of human scalp hair was studied by determination of cysteine, cystine, inorganic sulfate, thiosulfate, total protein, keratinase and change in alkalinity of culture filtrate. Gymnoascoideus petalosporus showed maximum degradation as compared to remaining isolates when grown on human scalp hair as the sole source of nutrients ‘in vitro’.
