    Notes: Abstract With the aid of column chromatography, solvent fractionation and toxicity assays we made the following observations. (1) The defensive skin secretion of the Red Sea trunkfish Ostracion cubicus consists, on a dry weight basis, of about 15% proteins, of 5 to 70 kDa molecular weight. (2) The proteins serve as an integral part of the chemical defense mechanism and fulfill two essential roles: (a) as ichthyotoxins which affect fish through external application into the surrounding water; and (b) as modifiers/regulators of the action of the well-known active ingredient of trunkfish secretion, the cationic, quaternary-ammonium, surfactant pahutoxin (PHN). The latter, when isolated from other components, plays only a limited role in ichthyotoxicity. (3) The regulatory role is performed by nontoxic, and as yet unidentified, proteins which potentiate PHN ichthyotoxicity. Potentiation is presumably preceded and mediated by the protein–PHN association, suggesting a guiding “pharmacokinetic” role of the regulatory proteins. (4) Finally, it may be concluded that the entire secretion process functions with unique pharmacological complexity due to the multiplicity of active substances and their interdependency.

    Notes: Summary Weight loss is the main symptom of so-called tumor cachexia. The pathogenetic mechanisms underlying cachexia are poorly understood; however, it appears that enhanced formation of cytokines such as interferon-γ and tumor necrosis factor-α are involved. In 94 patients suffering from hematological neoplasias we compared body weight changes with serum neopterin, tryptophan, and kynurenine. Biochemical changes, the formation of neopterin, the degradation of tryptophan are closely related to interferon-γ activity. The majority of our patients had increased neopterin and decreased tryptophan concentrations. Weight loss was seen particularly in patients with higher neopterin and lower tryptophan values. An association between higher neopterin levels and greater weight loss was apparent at study entry and during the follow-up of patients. Our data support the concept that weight loss is closely linked to endogenous interferon-γ activity.

    Description / Table of Contents: Zusammenfassung Submerskulturen von Streptomyces rimosus scheiden auf glucosehaltigen Medien in Gegenwart von Casaminoazids Pyruvat und α-Ketoglutarat aus, deren Konzentration durch Zugabe von puffernden Carbonaten erheblich erhöht werden kann. N-freie Versuchsansätze enthalten geringere Ketosäuremengen, zeigen jedoch innerhalb der späteren Entwicklungsabschnitte ein antagonistisches Verhalten von Pyruvat und α-Ketoglutarat. Von der 24. Std bildet sich α-Ketoglutarat unter Abnahme des ausgeschiedenen Pyruvates. Die Umbildung erfolgt stöchiometrisch. Zusätze von Succinat oder α-Ketoglutarat verzögern lediglich die Wiederaufnahme der extracellularen Brenztraubensäure, sie beeinflussen nicht die Anstiegswerte der Pyruvatbildung. Versuchsansätze mit Glycerin anstelle von Glucose zeigen, daß die Pyruvatstauung keinem Glucoseeffekt im Sinne spezieller Enzymrepressionen zugeordnet werden kann. Aus den Ergebnissen der NH4 .-Versuche geht hervor, daß Wachstumsvorgänge und Pyruvatstauung Antagonisten sind. Eine NH4 .-bedingte Pyruvatanhäufung erfolgt nur bei Anwesenheit von Succinat. Die Ketosäureausscheidung der Streptomyceten wird auf zwei ungleiche Stoffwechselstörungen zurückgeführt: die anfängliche gehemmte Pyruvatoxydation und die anschließende Konkurrenzbeziehung der Pyruvat-bzw. α-Ketoglutaratoxydation auf der Basis eines früher formulierten Kontrollprinzipes.
    Notes: Summary Submerged cultures of Str. rimosus excrete pyruvic and α-oxoglutaric acid in glucose-containing mediums in the presence of casaminoacids. After buffering with carbonates the amounts of keto acids increase about threefold. N-free cultures show smaller amounts of keto acids, but after 24 h we can see an antagonistic behaviour of both curves: pyruvic acid is converted stoichiometrically to oxoglutaric acid. Succinate and α-oxoglutarate delay the secondary reception of pyruvate without changes in primary excretion. Replacement of glucose by glycerine-containing cultures shows that pyruvate excretion is not due to a glucose-effect in the sense of enzyme-repression. NH4-ions depress, pyruvate-excretion and induce a very good growth. A NH4-dependent pyruvate peak appears only in the presence of succinate. The excretion of keto acids by Streptomycetes is caused by two unequal metabolic disturbances: the initially inhibited pyruvate-oxidation with failed pyruvate dehydrogenase, and the associated competition from pyruvate and/or α-oxoglutarate oxidation based on a previously formulated regulation principle.

    Notes: Abstract The clinical findings and pathological changes in a 14 year old boy who died from severe malabsorption are presented. Early signs of the illness were attacks of abdominal pain, diarrhoea, and intermittent periods of constipation. Later, severe malabsorption together with chronic ileus were apparent but no organic obstruction was found at several laparotomies. X-ray examination revealed extremly distended intestinal loops and a very slow transit time of barium. Malnutrition could not be improved by dietary, medical, or surgical therapy. Microscopic examination of the tissues obtained at autopsy showed changes in the small vessels of the lungs, heart, kidneys, and intestine. Increased fibrous tissue combined with atrophy of the muscular coat was observed in the wall of the entire intestine. Although cutaneous changes of scleroderma were not evident the deposition of collagenous material and the vascular changes seem typical of systemic sclerosis.

    Notes: Abstract The laminins comprise of a family of heterotrimeric proteins of the extracellular matrix. The cross-shaped proteins consist of a heavy α-chain and two light chains, called β and γ. Each group of chains, classified on their sequence identity and domain organization, include different isoforms. A deficiency of the α2 chain of laminin-2, previously termed merosin or M component, was shown to be responsible for one form of congenital muscular dystrophy (CMD). We investigated muscle biopsies of 20 patients with the clinical diagnosis of CMD and histological evidence of muscular dystrophy for the expression of different laminin chains. Patients with evidence of pachygyria/lissencephaly of the CNS were excluded from this series. The immunohistochemical analysis was correlated to clinical findings and MRI data of the brain. Of 20 patients, 11 (55%) revealed complete or near-complete deficiency of the α2 chain in their skeletal muscle specimens. So far none of these patients became ambulant. Of 20 patients 2 showed partial but clear-cut α2 chain-deficiency. These two patients became ambulant at 18 months and 3 years. All 13 patients with complete or partial α2 chain-deficiency demonstrated cerebral white matter changes on MRI. In contrast, 6/7 CMD patients with normal α2 chain expression became ambulant and none of the 6/7 tested showed evidence of cerebral abnormal T2 sequence signal of the myelin on MRI.

    Notes: A 205–210 bases long, small RNA (MP200RNA) of Mycoplasma pneumoniae encodes an open reading frame (ORF pmp200) that has the potential to be translated into a 29 amino acids long peptide with nine cysteines. The expression of this peptide in M. pneumoniae was proven indirectly by constructing a gene fusion between the ORF pmp200 and mrfp1, the gene encoding the monomeric red fluorescent protein. The fusion construct was translated in M. pneumoniae. The corresponding fusion protein, with a molecular mass of approximately 35,000 Da, was isolated and the correct sequence was proven by Edman degradation and by mass spectrometry
