    Notes: Summary The influence that changes in proton distribution have on the fluorescence of acridine orange was examined using negatively charged liposomes. Our results indicate that at least two mechanisms are involved: distribution of the probe between the internal aqueous phase of the liposomes and the outside medium, and binding of the probe to the liposome membranes.

    Notes: Abstract In a model for mucosal mast cells (RBL-2H3 cells) a set H1-antagonist derived anti-allergic drugs containing a diphenylmethyl piperazinyl moiety was examined for their ability to inhibit release of the mediatorβ-hexosaminidase. Cells were activated with antigen or the calcium ionophore A23187, whether or not in combination with the phorbol ester 12-O-tetradecanoylphorbol-13-acetate (TPA). Oxatomide, hydroxyzine and cetirizine inhibit the antigen inducedβ-hexosaminidase release. The release triggered by A23187, whether or not in combination with TPA is hardly influenced by the compounds. A biphasic dependence of the inhibition of exocytosis in RBL cells on lipophilicity is observed with the optimum at log P is 5–6. The extremely lipophilic compounds meclozine and buclizine are not active in this model. pH dependence of the effect of the drugs shows that especially the uncharged species are active in inhibiting exocytosis. The investigated compounds show an effect on phase transitions in L-α-phosphatidylcholine dipalmitoyl liposomes as assayed with differential scanning calorimetry (DSC). For the less extremely lipophilic compounds the induced changes in the phospholipid membranes increased with lipophilicity. The relation between structural features of the drug and the interaction with phospholipids is discussed in view of the DSC results. We conclude that location of the active drugs at the membrane or the membrane/protein interface is important for the inhibiting activity on exocytosis. This could affect several membrane related processes, which are abundant in the early phases of the IgE-mediated signal transduction process.

    Notes: Abstract The antigen induced stimulation of mast cells by aggregation of Fc∈RI receptors activates a signal transduction cascade leading to release of mediators of inflammation like histamine, arachidonic acid metabolites and cytokines. In this study we investigated a series of structurally related anti-allergic drugs, containing a common lipophilic diphenylmethyl piperazinyl tail and head groups that differ in lipophilicity. Effects of these drugs on various steps of the signal transduction cascade was investigated to gain insight into the mechanism of action of these drugs. It appeared that addition of the drugs to resting cells induced changes in the tyrosine phosphorylation of cellular proteins. The most active anti-allergics in inhibiting exocytosis, AL3264 and oxatomide, also induced the largest changes in phosphorylation. The effects of the drugs on tyrosine phosphorylation after cell activation was complex. Additionally, Ca2+ fluxes were investigated. Ca2+ efflux from the cells was negligibly influenced by the active drugs. However, the drugs inhibited influx from extracellular Ca2+, which was correlated with the effects of the drugs on inhibition of exocytosis and on membrane stabilization induced by the drugs, measured as haemolysis of erythrocytes. It is concluded that inhibition of Ca2+ influx is the major mechanism with which these drugs inhibit exocytosis and that for this effect drug-membrane interactions, possibly affecting the function of membrane embedded proteins, are of importance. Possible mechanisms including drug-membrane interactions, phosphorylation and inhibition of Ca2+ influx are discussed.

    Description / Table of Contents: Zusammenfassung Das Kalorimeter LKB 2277 kann verwendet werden, um den Einfluss kationischer, anionischer und neutraler Tenside auf die Stabilität einer Serie von n-Alkylacetaten zu untersuchen. Thermodynamische Parameter und Geschwindigkeitskonstanten werden berechnet. Folgende Schlussfolgerungen werden gezogen: 1, Die Stabilität der Ester steigt mit zunehmender Tensidkonzentration. 2. In Gegenwart von Tensiden steigt die Stabilität mit zunehmender Kettenlänge des Esters, der Beitrag einer CH2-Gruppe zur steigenden Stabilität des Esters scheint konstant zu sein.

    Notes: Abstract This work reports on the possibility of using the LKB 2277 thermal activity monitor to study the influence of anionic, cationic and neutral surfactants on the stability of a series of an-alkyl acetates. Thermodynamic parameters as well as rate constants could be calculated. Two conclusions can be drawn: 1. The stability of esters increases with increasing concentrations of surfactants. 2. The stability of esters in the presence of surfactants increases with the chain length of the esters; the contribution of a CH2 group to a higher stability of the esters in the presence of surfactants seems to be constant.

    Description / Table of Contents: Zusammenfassung Es wird gezeigt, dass mit dem kalorimetrischen Messgerät LKB 2277 Untersuchungen zur Bestimmung der Parameter chemischer Reaktionen sowohl in Ampullen wie auch im Durchflussbetrieb möglich sind. Theoretische Beziehungen für Reaktionen 1. Ordnung werden abgeleitet und mit solchen aus der Literatur verglichen. Vorgeschlagen werden ferner mathematische Beziehungen, die sich auf Reaktionen 2. Ordnung anwenden lassen. Die experimentellen Ergebnisse bestätigen die Gültigkeit der theoretischen Beziehungen und zeigen, dass die Durchfluss-Mikrokalorimetrie eine Schnellmethode für kinetische Untersuchungen ist.
    Abstract: РЕЗЮМЕ Используя ЛКБ 2277 термом онитор, было показано, что оба метода, как амп ул, так и проточный, могут быт ь использованы для регулирования реакц ионных параметров хи мических реакций. Предложены т еоретические соотно шения, применимые к реакция м первого порядка и которые сопоставле ны с приведенными в ли тературе. Предложены также мат ематические соотнош ения, применимые к реакция м второго порядка. Экспериментальные р езультаты подтверди ли справедливость теор етических соотношен ий и показали, что калориметрия в по токе является быстрым метором кине тических исследован ий.
    Notes: Abstract By using the LKB 2277 thermal activity monitor, it is demonstrated that both ampoule and flow-through modes can be used to monitor the reaction parameters for chemical reactions. Theoretical relationships applicable to first-order reactions are proposed and compared with those reported in the literature. Mathematical relationships applicable to second-order reactions are also proposed. The experimental results confirmed the validity of the theoretical relationships and showed that flow microcalorimetry is a rapid technique in kinetic studies.
