Notes: [Auszug] Stem cell fate is influenced by specialized microenvironments that remain poorly defined in mammals. To explore the possibility that haematopoietic stem cells derive regulatory information from bone, accounting for the localization of haematopoiesis in bone marrow, we assessed mice that were ...

Notes: Meitauza, an indigenous Chinese food made from fermented okara (soymilk residue), was used as a model system for monitoring solid substrate fermentations. The Instron Universal Testing machine and Chatillon hand operated force gauge were used to measure the force to penetrate meitauza as an estimation of mycelial binding. Increased firmness during fermentation was caused by fungal growth and not by water loss and surface crusting. Puncture force readings with the Instron were werll correlated (r = 0.86) with Chatillon measurements. The results of this study indicate that textural measurements using puncture force may have valuable application in monitoring the degree of mycelial growth or fermentation in a solid substrate.

Notes: Abstract. The automatic retrieval of indexing information from colored paper documents is a challenging problem. In order to build up bibliographic databases, editing by humans is usually necessary to provide information about title, authors and keywords. For automating the indexing process, the identification of text elements is essential. In this article an approach to automatic text extraction from colored book and journal covers is proposed. Two methods have been developed for extracting text hypotheses. The results of both methods are combined to robustly distinguish between text and non-text elements.

Notes: Summary The use of peripheral blood feeder layers as a source of stimulus for colony formation by human granulocytic progenitor cells in semi-solid agar cell cultures was examined. Comparison with various conditioned media demonstrated that cultures stimulated by peripheral blood feeder layers produced the greatest number and largest colonies. The cell concentration in the feeder layers was more important than the total cell number. Feeder layer plates containing 2 × 105 cells at a concentration of 1 × 106 cells/ml proved to be just as potent as the conventional feeder layer plates containing 1 × 106 cells/ml in 1 ml. Thus feeder layer plates can be more economical in terms of cell numbers than has previously been reported. By using a known panel of donors for the leukocytes, the number of sub-optimal batches of feeder layers was reduced. Addition of 1 μM adenosine 3′ : 5′-cyclic-monophosphate or 10 μg/ml Li2CO3 to poor feeder layers enhanced their colony-stimulating ability.

Notes: Summary This study examines the role of plasma lactoferrin in the assessment of neutropenia. In particular, we have studied lactoferrin as an inhibitor of granulopoiesis and as an indicator of the size of the total blood granulocyte pool (TBGP). Plasma lactoferrin concentration was determined in a heterogeneous group of 30 patients with neutropenia. Serial plasma lactoferrin levels in a patient with cyclic neutropenia correlated with the cycles of the neutrophil count. Patients with splenomegaly had a grossly elevated lactoferrin: neutrophil ratio. Most chronic idiopathic neutropenia patients had no real clinical problems and a normal plasma lactoferrin level. The results provide further evidence to support the concept that plasma lactoferrin indicates the size of the TBGP and the lactoferrin: neutrophil ratio indicates the degree of granulocyte margination. There was no evidence to suggest that lactoferrin acting as a feedback inhibitor of granulopoiesis caused neutropenia in these patients.

Notes: Abstract The human gene for parathyroid hormone (PTH) was chromosomally mapped using human-rodent hybrids and Southern filter hybridization of cell hybrid DNA. A recombinant DNA probe containing human PTHcDNA insert (pPTHm122) hybridized to a 3.7-kb fragment in human DNA cleaved with the restriction enzyme EcoRI. By correlating the presence of this fragment in somatic cell hybrid DNA with the human chromosomal content of the hybrid cells, the PTHgene was mapped to the short arm of the chromosome 11.
