    Notes: Abstract The [15N]methacetin urine test was used to study human O-demethylase activities to characterize the maturation of hepatic detoxification capacity. The study involved 43 healthy subjects aged 1 day−47 years. The urinary15N elimination rates were measured following oral administrations of an aqueous [15N]methacetin solution. Age-dependent normal values of hepatic drug elimination capacity were established. Parameters were the15N elimination half-life and cumulative elimination of the15N dose as a percentage over a 9 h period. The maximum elimination rate (% dose/h) and peak time can give additional information. The15N method is a simple, noninvasive and non-radioactive liver function test avoiding disadvantages of14C and13C breath tests. The [15N]methacetin test is suitable and useful in studying the hepatic development at birth and pathological changes of the microsomal detoxification capacity in early childhood.

    Notes: Abstract To establish nutritional management of low birthweight infants according to their individual metabolic situation, hepatocellular partial function was studied in 13 appropriate (AGA) and 11 small-for-gestational-age (SGA) low birthweight (LBW) infants during the first weeks of postnatal life. The concentrations of total bile acids and of alpha-amino-nitrogen in serum, the renal excretion of urea and ammonia and the renal excretion of15N after enteral administration of 3 mg15N-labeled methacetin/kg were measured. In comparison to AGA infants, SGA infants had elevated serum concentrations of total bile acids and of alpha-amino-nitrogen, decreased excretion of urea, increased excretion of ammonia in urine, and lower urinary15N-excretion after enteral administration of15N-labeled methacetin. The data suggest that hepato-cellular functions are influenced by intrauterine growth retardation resulting in a reduced metabolic capacity in SGA infants. The metabolic differences between SGA and AGA infants should be considered in the nutritional management of LBW infants.

    Notes: Abstract Stable isotopes, such as 13C, should be substituted for the long-lived radionuclide 14C in the 14C breath test when doing liver function tests in children and pregnant women. For comparison 13C, 15N-methacetin and 14C-methacetin were synthesized as suitable diagnostic agents. Methods are described for the measurement of 14C and 13C in the breath. After oral administration of labelled methacetin to healthy subjects and patients with liver diseases a good correspondence between 13C- and 14C-measurements in the same subject on the one hand, and a good discrimination between controls and patients on the other hand were shown. Findings with regard to 14C measurements in urine are discussed supporting the supposed advantage of 13C-methacetin over 14C-methacetin application.

    Notes: Abstract A simple, non-invasive, non-radioactive liverfunction test is proposed. After an oral dose of 3 mg 15N-methacetin per kilogram body mass, the kinetics of 15N excretion via urine were characterized by the quotient of the amounts of 15N excreted in two successive urine samples (Q value). The stable nitrogen isotope 15N was found to be an excellent and easily detectable indicator of the sum of all methacetin metabolites present in urine. Alterations in the nature or ratio of methacetin metabolites due to liver diseases could not be found. From the investigation of 11 men, 3 pregnant women and 15 children, a clear difference was observed in Q values of healthy persons and patients suffering from liver-cell-activity diseases. The discriminating power of our new liver-function test is shown to be equivalent to that of the 14CO2 breath test.

Aus den maximalen Autoxydationsgeschwindigkeiten wurden folgende primäre kinetische Brutto-Isotopieeffekte bei 61 °C bestimmt: Es wird versucht, den Gang der Isotopieeffekte innerhalb der quasihomologen Reihe mit der Annahme unterschiedlicher energetischer Verhältnisse im Übergangszustand der Reaktionen zu deuten.

    Notes: Durch die Bestimmung der Deuterium-Isotopieeffekte bei der Oxydation von Adipinsäure-di-n-amylester und der in der Alkoholkomponente D-substituierten Analoga Adipinsäure-di-n-amylester-[α-D2],-[β-D2],-[γ-D2] und -[α,β-D4] wurde gefunden, daß sich die Reaktivitäten der Methylengruppen der Alkoholkomponente gegenüber einem oxydativen Angriff nicht beträchtlich unterscheiden und in der Reihe α 〉 β 〉 γ abnehmen.Durch eine Abschätzung der Isotopieeffekte bei der thermischen Zersetzungsreaktion aus den Zersetzungstemperaturen der Verbindung konnte nachgewiesen werden, daß der rein thermische Angriff im Gegensatz zum oxydativen Angriff praktisch nur an der Methylengruppe in β-Stellung der Alkoholkomponente erfolgt. Dies steht in Übereinstimmung mit dem nach anderen Methoden bestimmten Reaktionsmechanismus der thermischen Zersetzung von Esterölen.
