    Notes: We present a procedure that locally determines the composition (from a given set of available materials) of a segmented or graded thermoelectric generator leg in order to maximize its output power to a matched load. The method is based on a new interpretation of the electrical output relation, which is organized to resemble a differential output power along the device coordinate. The procedure is applied to calculate the segment length and output power of a segmented bismuth-telluride/iron-disilicide based thermoelectric generator leg. © 1998 American Institute of Physics.

    Notes: Aims  The non-benzodiazepine hypnotics zolpidem and zopiclone, which are indicated for short-term treatment of insomnia, were considered originally by physicians as almost devoid of abuse and dependence potential. Several recent publications, however, have suggested that both agents carry a significant risk of abuse. To substantiate and re-evaluate this situation, the world literature was reviewed for cases of dependence of both agents; these cases were analysed in order to identify certain underlying patterns, if evident.Methods  A systematic review based on a Medline literature search was conducted including the years 1966–2002 to assemble all available clinical case reports that were analysed for typical features of abuse and dependence according to prespecified criteria. Only case reports were of interest, and clinical studies were excluded. No limitations as to language or publication year were applied. The terms ‘zolpidem’, ‘zopiclone’ and ‘abuse’, ‘dependence’, ‘addiction’, ‘withdrawal’ and ‘intoxication’ were used to identify relevant publications. Potentially relevant citations were retrieved and assessed for inclusion independently by two authors.Results  A total of 36 cases for zolpidem were identified, most of them reported in recent years, and 22 cases for zopiclone. Both sexes were involved to a similar extent; and cases were reported in all age groups. In extreme cases, dose increases reached a factor of 30–120 above the recommended doses. The majority of patients had a history of former drug or alcohol abuse and/or other psychiatric conditions.Conclusion  On the basis of world-wide prescription numbers, which are approximately twofold higher for zolpidem (1 338 774 000 tablets from June 2001 to June 2002 in Europe, Japan and United States) than for zopiclone (664 897 000 tablets during the same period in Europe and Japan), the relative incidence of reported dependence similar for both drugs and remarkably lower than that of benzodiazepines used for the treatment of disturbed sleep. The findings offer the conclusion that zolpidem and zopiclone are relatively safe drugs. However, as both drugs are psychotropic drugs, patients with a history of abuse or dependence and those with psychiatric diseases seem to be at increased risk of abuse of these agents.

    Notes: Abstract In a previous study it was demonstrated that a lectin controls cell-cell interaction in the gorgonian Eunicella cavolinii (Koch) as a negative modulator. Now we describe the procedure to purify this lectin to homogeneity; its molecular weight is 23 400. The homologous proteoglycans were identified as positive modulators of cell-cell (and/or cell substrate) interaction. The purified single proteoglycan aggregates were 1200±700 nm long and the distance between the attachment points of the proteoglycan subunits was about 45 nm. The glycosaminoglycan residues of the gorgonian proteoglycans were identified as hyaluronic acid (35.5%), heparan sulfate (47.9%) and dermatan sulfate (14.1%). Binding studies with immobilized homologous proteoglycan preparations revealed that gorgonian cells attach to this substratum presumably via its glycosaminoglycans. Quantitative determinations of these two modulators of cell-cell recognition in the three regions of individual colonies revealed concentration gradients these concentration differences reflect a lower cell motility in the polyp-bearing branches and a higher motility of the cells in the polyp-free stem and in the basis, is discussed.

    Notes: Abstract Induction of benzo (a) pyrene monooxygenase (BPMO) activity occurred in Blennius pavo, a species with a restricted territorial range, in response to exposure to a Diesel 2 oil. A response delay of 14 days was found at a concentration of 170 ppb and of 3 days when the water was saturated with Diesel 2 oil. When induced fish were transferred to clean water, elevated BPMO activity was maintained at a high level for at least a month. A benthic protochordate, Microcosmos sulcatus, showed no increase in BPMO activity when exposed to these concentrations even after 30 days of exposure. Field observations revealed a great variation in the BPMO activity from B. pavo caught at different sites. Fish from contaminated sites had significantly elevated levels of BPMO activity. Sardine schools caught at different sites had different, low levels of BPMO activities. However, specimens from the same school had closely similar levels of enzyme activity. An oil pollution incident (New Year 1977 oil spill in Northern Adriatic Sea) caused an increase in the BPMO activity in the livers of Blenniideae, reaching a peak on the 23rd day (representing an 8.5-fold increase in the background level), followed by a decrease in activity until a new background level, 3 times that of the original background level, was reached on the 45th day. This new background level is constant (through May-5 months after the incident). Measurement of BPMO activity in the livers of nonmigrant fish could serve as a useful biochemical parameter for monitoring and evaluation of acute or long-term oil pollution at a given site.

    Notes: Abstract A hydrolysis procedure along with a high-pressure liquid chromatographic procedure is given enabling simple and reliable thymine determinations in the nanogram range in different fractions of sea-water samples taken from three different locations in the Northern Adriatic Sea. The levels corresponded to 1–3 μg DNA per liter. From total polyanionic thymine, which had been precipitated as the cetyltrimethylammonium salt, the highest percentage was linked to the particulate fraction, with a definite subsurface minimum at 10 to 15 m. There was a corresponding maximum of a high molecular “non-particulate” thymine-containing fraction at the corresponding depth. From the bottom at 30 m upwards to about 20 m, a low molecular thymine-containing material has been found. Remarkably these basic features were common to all three locations, one of which was supposed to be in clean water, one near a thickly settled, touristic area, and the other in front of a large river delta coming from industrial hinterland.

    Notes: Abstract Chronic treatment (2 weeks) with piracetam (500 mg/kg, once daily PO) elevatedm-cholinoceptor density in the frontal cortex of aged (18 months) female mice by about 30–40%, but had no effect onm-cholinoceptor density in the frontal cortex of young (4 weeks) mice. The effect of piracetam onm-cholinoceptor density as determined by the specific binding of tritiated QNB was not affected by concomitant daily treatment with either choline (200 mg/kg) or scopolamine (4 mg/kg). It is concluded that the effect of piracetam onm-cholinoceptor density could explain the positive effects which have been reported for combinations of cholinergic precursor treatment with piracetam on memory and other cognitive functions in aged experimental animals and patients and could also represent part of the possible mechanism of action of piracetam alone.

    Notes: Summary By means of the gel filtration technique, the effect of nine benzo-diazepine derivates on the binding of l-tryptophan to human serum albumin was investigated. Using equimolar tryptophan and benzodiazepine concentrations, all benzodiazepines with binding constants higher than 104 (M−1), displace l-tryptophan from its binding site to a high degree. The mechanism of the displacement was characterized as a competition for a common binding site. Some of the benzodiazepines displace l-tryptophan to a greater extent than salicylic acid. The benzodiazepines and tryptophan are the only substances known with a high degree of stereospecific binding to human serum albumin. This study shows that there is only one binding site on the human serum albumin molecule, which binds tryptophan and the benzodiazepines in a highly stereospecific manner. Therefore it is concluded that the benzodiazepines and l-tryptophan must have similarities in their molecular structure, so that both can bind to the common binding site in such specific manner. These considerations are discussed in regard to the known influence of benzodiazepine derivatives on the l-tryptophan metabolism in brain. A direct involvement of the reported displacement in the pharmacological actions of the drugs seems not to be relevant because of their small therapeutical plasma levels.

    Notes: Summary The interaction of 5 sulfonylurea derivatives with insulin was investigated by means of circular dichroism measurements. It was found that all sulfonylureas investigated decrease the ellipticity band of insulin at 208 nm, whereas the band at 222 nm remains unaffected. It is suggested that these observations are due to a change of the insulin conformation, provoked by the interaction of the drugs with insulin. It is assumed that such an effect on the insulin conformation can influence the binding properties of insulin, e.g. in respect to the insulin aggregation, to the binding to insulin antibodies and to a bound, inactive form of insulin in the plasma. Some other drugs have similar, but mostly smaller effects on the CD spectrum of insulin. Therefore, the importance of these observations for the hypoglycaemic action of the sulfonylureas is not yet clear.
