Summary The rate of destruction of normal donor lymphocytes in the peripheral blood was determined in 12 cases after in vitro labeling with Na 2 51 CrO4 by means of a new method. After red cell sedimentation with dextran (3 %; Mol. Weight 250.000) leukocytes from 500 ml blood were concentrated, labeled and reinjected. The first blood sample after retransfusion was drawn at the 13th day. At this time there is no activity of granulocytes or platelets in the peripheral blood. The red cells were hemolyzed in ammonium chloride. The average activity of lymphocytes on the 13th day p.i. was 0.53 per cent of the total radioactivity transfused and decreased to 0.12 per cent to the 48th day. The slope of the activity curve is neither linear nor definitely exponential. The parameter for the rate of destruction of lymphocytes was the half-life of lymphocyte-activity determined from the 13th day onward. The half life was 13±2.6 days. The reinjected cells were morphologically normal which was shown by electronic microscopy. The method is available for a comparison of the destruction of lymphocytes between normals and patients with deseases of the lymphatic system.

Zusammenfassung Bei zwölf freiwilligen, gesunden Spendern wurde der in vivo-Abbau von Lymphocyten nach in vitro-Markierung mit Na 2 51 CrO4 mittels einer neuen Methode quantitativ bestimmt. Dabei wurden die Leukocyten mit Dextran (3%, Mol.-Gew. 250000) aus 500 ml Vollblut angereichert, markiert und reinjiziert. Die erste Blutabnahme nach der Retransfusion erfolgte am 13. Tag, da zu diesem Zeitpunkt weder Granulocyten- noch Thrombocytenaktivität im Blut mehr vorhanden ist. Die Erythrocyten wurden durch Ammoniumchlorid hämolysiert. Die mittlere Lymphocytenaktivität betrug am 13. Tag p.i. 0,53 % des Sollwertes und fiel bis zum 48. Tag auf 0,12 % ab. Der Verlauf der Lymphocytenaktivitätskurve ist weder linear noch sicher exponentiell. Als Parameter für die Abbaurate der Lymphocyten wählten wir die Halbwertszeit der Lymphocytenaktivitätskurve, die vom 13. Tag an bestimmt wurde. Die Halbwertszeit betrug 13±2,6 Tage. Die reinjizierten Zellen zeigten nach den in vitro-Manipulationen selbst elektronenmikroskopisch keine morphologischen Veränderungen. Die Methode ist zu vergleichenden Untersuchungen über den Abbau von Lymphocyten zwischen Normalpersonen und Patienten mit Erkrankungen des lymphatischen Systems geeignet.
