    Description / Table of Contents: Summary The effect of Aldadiene (-K, -Na) was studied in the isolated cat papillary muscle preparation. At doses of 500–2000 µg/ml, significant increases are observed in the extent of shortening, in the velocity of isotonic shortening and relaxation and in the rate of isometric tension development. Force-velocity relations are shifted upwards and to the right with increases of both the preload velocity and the maximum isometric tension. The maximum increases of cardiac mechanics are observed 3–4 min after Aldadiene incubation. In most of the experiments there is a late decrease of cardiac mechanics compared with the controls, approximately 20 to 30 min after Aldadiene incubation, which is most probably due to the elevation of extracellular potassium concentration associated with Aldadiene-K incubation. Using Aldadiene-Na, this late decrease of cardiac mechanics was largely reduced. The results demonstrate a direct positive inotropic action of Aldadiene (-K, -Na) on the isolated ventricular myocardium.
    Notes: Zusammenfassung In isotonischen Unterstützungskontraktionen wurde der Einfluß von Aldadiene auf die Kontraktilität des isolierten Ventrikelmyokards untersucht. In Konzentrationen zwischen 0,1–100 µg/ml werden nichtsignifikante Zunahmen wesentlicher herzmechanischer Größen beobachtet. In Konzentrationen zwischen 500–2000 µg/ml sind signifikante Zunahmen der Muskelverkürzung, der Verkürzungs-und Erschlaffungsgeschwindigkeit sowie der Geschwindigkeit des isometrischen Spannungsanstieges zwischen 20–60% der Ausgangswerte nachweisbar. Simultan ermittelte Kraft-Geschwindigkeits-Beziehungen zeigen eine Rechtsverlagerung mit Zunahmen der isotonischen Verkürzungsgeschwindigkeit und der maximalen isometrischen Muskelspannung. Das Wirkungsmaximum ist nach etwa 3–4 min erreicht. Eine nach Abklingen der positiv inotropen Aldadienewirkung cinsetzende Reduktion der mechanischen Größen unter die entsprechenden Ausgangswerte läßt sich auf die Erhöhung der extracellulären Kaliumkonzentration zurückführen, die mit der Inkubation von Aldadiene verbunden ist. Unter der entsprechenden Natriumverbindung (Na-Aldadiene) ist diese späte Abnahme der Herzmechanik weitgehend aufgehoben. Die Befunde zeigen, daß Aldactone (Na-Aldadiene, K-Aldadiene) am isolierten Ventrikelmyokard positiv inotrop wirkt.

    Notes: The thin-film growth of Co oxides and layered Bi–Sr–Co–O compounds was investigated in detail under an ozone ambient. All samples were prepared using a molecular-beam-epitaxy machine modified for the growth of Bi-based high-temperature superconductors. The temperature versus pressure phase diagram of Co oxidation was determined for growth on Nd:YAlO3 substrates. According to the phase diagram, Bi2Sr2CoO6+α and Bi2Sr3Co2O9+β were grown on Nd:YAlO3 (001). The boundary between the stable regions of them is basically the same as that between CoO and Co3O4. Bi2Sr2CoO6+α was an insulator with an activation energy of approximately 0.20 ± 0.01 eV. Bi2Sr3Co2O9+β shows metallic behavior down to 80 K, and the carriers are revealed to be holes by Hall-effect measurement. © 1996 American Institute of Physics.

    Notes: Several sets of data indicate the possibility that carbohydrate moieties on the target cell are important structures in natural killer (NK) cell-mediated lysis. Strkining changes in the NK susceptibility of targets can be induced in several systems involving in vitro differentiation of tumour cell lines. The effect on target cells of the glycosylation inhibitor tunicamycin, which acts by blocking the dolichol-dependent asparagine-linked glvcosvlation pathway was investigated. Using several different tumour cell lines we can conclude that; (a) asparagine-linked carbohydrate chains do not contribute directly to NK susceptibility, (b) induced differentiation may or may not be linked with a change in NK susceptibility, and (c) secondary changes caused by tunicamycin treatment may lead to alterations in the gangliosides, a finding that is positvely correlated with decreased NK susceptibility.

    Notes: A large granular lymphocyte (LGL) leukaemia cell line from the Fisher/F344 rat strain called RNK-16 has been established in vitro, maintaining the same surface markers as the tumour cell growing in vivo. The tumour has also maintained its specificity pattern and cytotoxic reactivity and serves as a suitable source of natural killer (NK)-like effector cells in vitro. The cells show no evidence of dependency on, or production of, interleukin 2 or interferons, not is the cytotoxic capacity influenced by treatment with mitogens. The in vitro line does not produce natural killer cytotoxic factor (NKCF) in a constitutive manner, hut can be induced to do so via coculture with tumour target cells. When the fine specificity patterns were analysed, the RNK-16 cells express species-preferential lysis of susceptible target cells and a highly discriminatory power to kill only 1 out of 5 rat erythroleukaemia cell lines. When testing normal target susceptibility patterns. RNK-16 kills lymphoblasts of B type better than T blasts, which is well in line with previous findings on normal NK cell specificity patterns.

    Notes: Summary The phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (IBMX) mimicked the effects of isoprenaline on the force of contraction, the cAMP content and the slow Ca++ inward current (Isi) in isolated guinea pig papillary muscles. The results support the hypothesis that phosphodiesterase inhibitors and β-adrenoceptor agonists exert their positive inotropic effects by increasing Isi via the common mediator cAMP.

    Notes: Abstract A 68-year-old woman with early rheumatoid arthritis (RA) was admitted to the hospital because of tender and swollen knee joints. We performed a targeted synovial biopsy under arthroscopy to examine the histopathological characteristics 1 month after clinical onset. The synovia showed the typical histopathology of RA. Although the inflammatory changes were predominantly limited to the surface area of the synovia, associated with neovascularization and cell infiltrates composed mainly of T cells, plasma cells, and macrophages, lesions with fibrin deposition, mesenchymoid transformation and/or immature lymphoid follicles were also observed in part, indicating that this case was in the progression phase of RA. What we regularly call "early" might be "too late" even if it is within 1 month of clinical onset.
