    Notes: Summary Recent evidence indicates an important role for cell-surface mediated signal transduction in embryonic induction. We, therefore, started a systematic search to identify signal transduction pathways which are activated during embryonic induction and specifically during neural induction. We showed previously that the protein kinase C and cAMP pathways mediate neural induction inXenopus laevis. Here, we investigated whether cGMP is also involved in the early development of the nervous system. We measured the cGMP content of whole embryos at embryonic stages which mark important events in the early development of the nervous system, as well as in the developing neural tissue itself, after this was induced from ectoderm by dorsal mesoderm. No changes in cGMP content were found, either in whole embryos at different developmental stages, or in developing neural tissue from these stages. We also found no evidence for the presence of nitroprusside stimulatable guanylate cyclase in these developmental stages. A cGMP analogue, 8-Br-cGMP, was not able to induce neural tissue, either alone or in combination with known neural inducers, the phorbol ester TPA and 8-Br-cAMP. 8-Br-cGMP also had no negative influence on the neural inducing ability of dorsal mesoderm or TPA, alone or in combination with 8-Br-cAMP. We conclude that cGMP has no role in the early development of the central nervous system inXenopus laevis. This conclusion underlines the specificity of the signal transduction pathways (PKC and cAMP pathways) that do mediate neural induction.

    Description / Table of Contents: Zusammenfassung Am Anwendungsbeispiel von Patienten, die einer Pankreasfunktionsprüfung unterzogen wurden, wird die computergerechte Dokumentationqualitativer Daten (Anamnese, Symptome, körperliche Untersuchungsbefunde, Art und Ergebnis von Röntgen-, Isotopen- und endoskopischer Diagnostik) sowie vonquantitativen Informationen (Alter, Gewicht sowie Werte von Laboruntersuchungen) für ein Krankengut mit begrenzter Patientenzahl beschrieben. Die Informationen werden zunächst ohne Verschlüsselung in einem Datenheft gesammelt und anschließend mit Port-a-Punch-Karten abgelocht. Nach ausgiebiger Fehlerkontrolle durch die EDV werden die Daten in einer Datenbank auf Magnetplatte gespeichert. Das Informationssystem ISIS und speziell entwickelte Programme ermöglichen den Zugriff auf die Daten in jeder beliebigen Kombination. Es wird dadurch sowohl eine patienten-als auch eine statistikbezogene Auswertung wesentlich erleichtert.
    Notes: Summary Computer-oriented documentation of quantitative and qualitative data obtained from patients with gastroenterological diseases is described as an example of a method when considering a relatively small number of patients. All qualitative and quantitative features are entered uncoded in a data booklet and subsequently on port-a-punch cards. After extensive computerized error check — and correction, as need arises, — the data are transferred to a data base and stored on magnetic disk. The ISIS information system, in conjunction with special routines provides access to the data in any combination desired. Analysis of the data, both on an individual and a statistical basis is thus facilitated.

Pankreatitis wurde dieβ 2m- und CEA-Konzentration im Serum bestimmt. 11 Patienten mit Pankreascarcinom (n=22) zeigen gleichzeitig eine Erhöhung derβ 2m- und CEA-Konzentration und grenzen sich signifikant von Patienten mit chron. Pankreatitis und anderen malignen Tumoren ab. Keiner der Patienten mit chron. Pankreatitis zeigte eine Erhöhung der Serum-β 2m-Konzentration und nur 3 Patienten mit Pankreascarcinom wiesen bzgl. beider Parameter Werte im Referenzbereich auf. Die gleichzeitige Bestimmung vonβ 2-Mikroglobulin und CEA kann deshalb bei klinischem Verdacht auf ein Pankreascarcinom einen zusätzlichen diagnostischen Hinweis geben.
    Notes: Summary The concentration ofβ 2-microglobulin (β 2m) and carcinoembryonic antigen (CEA) was measured radioimmunologically in the sera of 79 patients with malignant disorders and 15 patients with chronic pancreatitis. Elevated levels ofβ 2m and CEA were found in 11 out of 22 patients with carcinoma of the pancreas, which sets off this malignancy from chronic pancreatitis and other malignant tumors. Only 3 patients with carcinoma of the pancreas exhibited serum levels within the normal range for both parameters and none of the patients with chronic pancreatitis was shown to have elevated levels ofβ 2m. The simultaneous determination ofβ 2m and CEA suggests itself for the diagnosis of pancreatic malignancy especially in the case of a tentative diagnosis of a pancreatic tumor.

    Description / Table of Contents: Zusammenfassung 31 Patienten mit operativ gesicherter chronischer Pankreatitis, 26 Patienten mit chronisch calcifizierender Pankreatitis und 16 Fälle von Pankreascarcinom wurden einer Pankreasfunktionsprüfung unterzogen. Zur Stimulation der Pankreassekretion wurden je 1 E/kg KG Cholecystokinin (CCK) und Secretin (Boots) i.v. verwandt und 60 min später dieselben Hormondosen kombiniert mit der Gabe von 6 g Rindergalle intraduodenal (Kombinationsreiz). Die in 60 min sezernierte Trypsinmenge war der zuverlässigste diagnostische Parameter, die Aussagekraft des Kombinationsreizes übertraf die des CCK-Secretintests. Der Anteil der normalen Testergebnisse bei den Patienten mit gesicherter Pankreaserkrankung sank bei alleiniger Berücksichtigung der Trypsinmenge von 24,6% nach Injektion von CCK und Secretin auf 10,9% nach dem Kombinationsreiz. Die paarweise lineare Varianzanalyse ermöglichte eine weitere Verbesserung der Diagnostik. Mit dem Kombinationsreiz ist es außerdem möglich, funktionelle, auf mangelndem Gallefluß beruhende Pankreasfunktionsstörungen von organischbedingten abzugrenzen.
    Notes: Summary Pancreatic function was measured in 29 normals and in 73 patients with proved pancreatic diseases (chronic pancreatitis, chronic calcifying pancreatitis and carcinoma of the pancreas). Pancreatic secretion was stimulated by simultaneous injection of 1 U/kg cholecystokinin (CCK) and 1 U/kg secretin (Boots) followed by intraduodenal instillation of 6 g cattle bile combined with the injection of 1 U/kg CCK and 1 U/kg secretin (combined test) 60 min later. Trypsin secretion was the best single parameter for the diagnosis of an impaired pancreatic function. The diagnostic value of the combined test exceeded the cholecystokinin-secretin test. Normal values of trypsin were observed in 24.6% in patients with proved pancreatic diseases after stimulation with CCK and secretin whereas this figure decreased to 10.9% after the combined test. Further improvement of diagnostic procedure can be achieved by the use of calculus of variations. Furthermore, the combined test allows differentiation between functionally disturbe 1 pancreatic secretion as a result of diminished endogenous bile flow and reduced pancreatic secretion as a consequence of pancreatic diseases.

    Description / Table of Contents: Zusammenfassung Im Abstand von 60 min wurden 15 Personen 1 Ivy-Hundeeinheit/kg Cholecystokinin (CCK) und 1 CHR-Einheit/kg Secretin zweimal nacheinander i.v. injiziert. Die Volumen- und Bicarbonatsekretion des Pankreas war nach beiden Injektionen fast identisch. Nach der zweiten Gabe der Hormone lagen die Bilirubin- und Enzymmengen deutlich niedriger. Durch intraduodenale Gabe von 6 g getrockneter Rindergalle gelöst in 20 ml Wasser vor dem zweiten Reiz wurde bei 29 Personen anstelle eines Abfalls der Enzymsekretion eine signifikante Steigerung erzielt. Die Sekretionswerte nach dieser Kombination von intraduodenaler Gallegabe und Injektion von CCK und Secretin (Kombinationsreiz) sind ebenso reproduzierbar wie nach alleiniger Verabreichung der Hormone. Unter Berücksichtigung der log-normalen Verteilung wurden Normbereiche für die 20, 40 und 60 min Sekretionsperioden nach dem CCK-Secretinreiz und dem Kombinationsreiz errechnet.
    Notes: Summary 15 normal individuals received twice at intervals of 60 min 1 Ivy dog U/kg body weight cholecystokinin (CCK) combined with 1 CHR U/kg secretin (Boots) intravenously. The volume and the bicarbonate secretion of the pancreas were almost identical following the initial and repeated injection. The secretion of bilirubin and enzymes were decreased after the second stimulation. However significant increase of enzyme secretion was seen, when 6 g of dried cattle bile dissolved in water were given by intraduodenal instillation immediately before the second injection of CCK and secretin. The results were reproducible after both the administration of the two hormones alone and following the combined stimulation with bile, CCK and secretin. The known log-normal distribution of pancreatic secretion was confirmed. Normal ranges were calculated for periods of 20, 40 and 60 min both after stimulation with CCK and secretin as well as after the combined stimulation.
