Fourier transform Raman spectra were obtained from viable colonies of bacteria. Despite the highly coloured nature of the samples, the spectra were of sufficient quality to allow comprehensive assignments consistent with the vibrational modes. The spectra showed no clear evidence for the presence of phospholipids, indicating that penetration of the exciting infrared radiation was limited to the outermost components of the bacterial cell walls. The data describing the molecular and conformational state of the bacterial cell wall may aid in probing the modes of action of a wide range of antimicrobial agent.
