    Notes: Abstract Objective and Design: Relevance of the preclinical pharmacodynamic, toxicity and pharmacokinetic parameters predicting the clinical potency of nonsteroidal antiinflammatory drugs (NSAIDs) was evaluated. Material: Data for oral potencies of 24 NSAIDs in rats were collected from the literature and from New Drug Applications with respect to the following parameters: antiinflammatory, analgesic, antipyretic, acute ulcerogenic activities, acute toxicity, in vitro inhibition of prostaglandin synthesis, acid dissociation constant (pKa), octanolwater partition coefficient and elimination half-life. Treatment: Data for most of the in vivo parameters in rats were collected following single dose administration with the exception of adjuvant arthritis. Single and daily clinical doses were considered. All of these NSAIDs have been approved for marketing although not all have been sold in the USA. Methods: The preclinical data were compared to human dose (unit or daily doses) using single and multiple stepwise regression analyses. Results: Analyses suggest that NSAIDs are effective in all models of preclinical tests for fever, pain and inflammation, however, carrageenin-induced rat paw edema model is clearly the best predictor of human dose. Rank order of preclinical models for predicting human dose is carrageenin 〉yeast induced fever〉pressure induced pain=adjuvant arthritis in rats. The analysis suggested that the pain and adjuvant arthritis models in rats may also involve a prostaglandin independent mechanism. Of the two physicochemical factors tested, pKa contributed best to the carrageenin model towards predicting the clinical potency of NSAIDs. Mathematical relationships between human dose, carrageenin ED50 and pKa were established that may assist in the future clinical development of NSAIDs. Conclusions: Carrageenin-induced paw edema model in rats is the most robust predictor of the clinical potency of NSAIDs. Acid dissociation constant (pKa) appears to be a secondary contributor to the potency of NSAIDs. The relevance of the data analyses for developing cyclooxygenase-2 (COX-2) selective NSAIDs is discussed.

    Notes: Abstract. This investigation shows the membrane stabilizing effect of α-tocopherol against the damaging action of viper venom phospholipase A2 (PLA2). Liver lysosomal membranes from rats fed 100 mg and 200 mg α-tocopherol acetate per kilogram of diet were more resistant to damage by viper venom PLA2 compared with vitamin E-deficient rats. The membrane stabilizing effect of vitamin E is proposed to be due to the formation of a complex with the phospholipid hydrolysis products of the membrane.

    Notes: Direct observation of superplastic deformation in shear in a scanning electron microscope in Pb-62% Sn eutectic alloy at different microstructural levels were performed. The sliding of grains as an entity was observed. Such cooperative grain-boundary sliding proceeds by means of the sequential shear of grains along shear surfaces, and is accompanied by cooperative grain-boundary migration. The pattern of shear surfaces at the level of the entire deformed volume is close to one that is predicted by slip-lines field theory. The processes to accommodate deformation of grain-boundary sliding include the operation of grain-boundary migration, intragranular deformation, and grain rotation. A critical analysis of the proposed geometrical models for superplastic flow is given. The model of sequential shear of grains as a result of the movement of cellular dislocations is extended to the case of the two-phase material, having structure that is typical for superplastic eutectics.

    Notes: A high speed computer interface and closed-loop feedback control system has been integrated into an existing, mechanical lever-arm, constant load, high temperature creep testing machine to shed load as a function of decreasing cross-sectional area such that a state of constant stress can be obtained throughout the test duration. This allows a computer interface and the custom designed mechanical load shedding apparatus to precisely control the applied stress in response to any changes in the specimen's measured resistance to deformation during a material evaluation test. A background analysis of typical mechanical creep machines and the design concepts and technical innovations related to the novel apparatus presented in this work will be discussed in detail. © 1997 American Institute of Physics.

    Notes: Distributed Bragg reflection from integrated polymethylmethacrylate waveguides fabricated on corrugated spin-on-glass substrate is reported. Utilizing third-order Bragg resonance, a maximum reflection of 92% from the grating filter is measured at a wavelength of 423.6 nm. A maximum bandwidth of the Bragg reflection is found to be ∼0.5 nm for a 6 mm long filter. The broad bandwidth and the significant transmission loss (∼90%) away from resonance are believed to be related to the radiation coupling by the grating. It is shown that the radiation loss can be minimized without sacrificing high reflectivity by limiting the waveguide length to 0.5 mm, the saturation length of the Bragg reflector. © 1995 American Institute of Physics.

    Notes: Two-photon absorption cross sections of two dye molecules, doped in thin films of polymethylmethacrylate, are measured by comparing fluorescence intensities induced by single- and two-photon excitations. Ratio of the single- and two-photon absorption cross sections is related to the ratio of the single- and two-photon pumped fluorescence intensities. Using this sensitive and background-free detection technique, dispersion of the nonlinearity is measured over a wavelength range of 700–800 nm. © 1997 American Institute of Physics.

    Notes: A novel process for the direct laser writing of thin-film high Tc patterns is demonstrated. The process consists of deposition of a high quality film (308 nm laser ablation from a YBa2Cu3Ox target with a 750 °C substrate temperature and a 150 mTorr O2 ambient), annealing in an inert atmosphere (Ar at 400 °C for 5–20 min) to reduce the oxygen content and depress or eliminate the superconducting transition temperature, and direct-write laser heating (1.06 μm at ≈0.5 kW/cm2 for ≈5 min) in an oxygen atmosphere at ≈590 Torr to selectively regenerate the high Tc material. rf eddy current and four-point resistivity probe results confirm this process for both SrTiO3 and LaAlO3 substrates. Scanning electron micrographs indicate that this is a very mild processing sequence with no observable changes in film morphology.

    Notes: Three antituberculous drug regimens have been employed to study the therapeutic response in 90 patients with any one of the commonly encountered paucibacillary forms of skin tuberculosis, namely lupus vulgaris, tuberculosis verrucosa cutis and scrofuloderma. The first two regimens contained rifampicin, isoniazid and either pyrazinamide or thiacetazone, and the third regimen had rifampicin and isoniazid only. The disease was clinically defined as localized when confined to one area and widespread when the lesions were disseminated. The observations revealed that the response of lupus vulgaris and tuberculosis verrucosa cutis was alike in all the three regimens, with the localized lesions subsiding completely after 4 months of therapy and the more extensive forms taking 5 months. Patients with scrofuloderma responded similarly to both the triple drug regimens. The discharge, sinuses and ulcers cleared in 6 months but the lymph nodes took longer to regress, up to 7 months in localized and 9 months in more widespread scrofuloderma. To obtain the same results with rifampicin and isoniazid, all patients with widespread scrofuloderma and one-third of those with localized forms had to be treated for 10 and 9 months, respectively. No serious drug side-effects, apart from giddiness with rifampicin and acneiform eruptions with thiacetazone, were encountered. No instances of relapse were noted in the 50% of patients who were followed-up for 31/2 years after therapy. Single-drug therapy with isoniazid for lupus vulgaris, as given in the past, is to be discouraged as it may promote the emergence of drug-resistant bacilli in those with an undetected focus of infection. An additional object of defining effective mutliple drug regimens is to improve patient compliance by reducing the duration of therapy and to avoid confusion by adhering uniformly to the recommended regimen in places where tuberculosis is prevalent.
