    Notes: Calmodulin levels in cultured skin fibroblasts from patients with progressive systemic sclerosis (PSS) and healthy controls were measured by their ability to activate cyclic AMP-phosphodiesterase. Calmodulin levels were significantly increased in PSS fibroblasts compared with normal control fibroblasts. The changes in calmodulin content of PSS fibroblasts were also assessed by a radioimmunoassay. These findings suggest that an elevated of calmodulin may play a role in the pathogenesis of PSS.

    Notes: Summary Single-unit activities were recorded from floccular Purkinje cells in 29 alert pigmented rabbits. The floccular areas specifically related to horizontal eye movement (H-zone) were identified by the effects of local stimulation and later confirmed histologically. Most of the 53 H-zone Purkinje cells responded to both vestibular stimulation with turntable oscillation (5° peak-to-peak, 0.1 Hz) in darkness and optokinetic stimulation with dot screen oscillation (2.5°, 0.33 Hz), often showing a preference either for turntable oscillation or screen oscillation. Since these responses were not correlated with the velocity of evoked eye movements or eye position shift, it is concluded that the major inputs to flocculus H-zone Purkinje cells are head velocity or retinal slip signals rather than eye velocity or position signals.

    Notes: Abstract Four patients with pontine gliomas were treated with radiation and intra-arterial chemotherapy using ACNU, cisplatin, and/or carboplatin. All underwent biopsy and were histologically proven to have gliomas. Even though three were benign histologically, all appeared malignant on neuroimaging. Two patients had complete remissions; the others had partial responses; three patients died, 36, 14, and 15 months after the primary diagnosis. One patient, now in complete remission, has been alive for 24 months. There was no neurotoxicity except for transient nausea and vomiting. Intra-arterial chemotherapy is effective against brain stem gliomas, producing remission with minimum side effects.

    Notes: Summary Intracellular Ca2+ concentrations ([Ca2+]i) in cultured skin fibroblasts from normal subjects and progressive systemic sclerosis (PSS) patients were determined by using Fura-2 and fluorescent videomicroscopy. With the exception of fibroblasts from one PSS patient showing a higher [Ca2+]i, no significant difference was observed in resting [Ca2+]i between the two groups of fibroblasts. Bradykinin (BK) (10 ΜM) induced a transient [Ca2+]i increase in normal fibroblasts, whereas the BK-induced [Ca2+]i increase was reduced or not detectable in PSS fibroblasts. Removal of extracellular Ca2+ did not eliminate the BK-induced [Ca2+]i increase in normal fibroblasts. These findings suggest that BK stimulates Ca2+ release from intracellular stores in human fibroblasts, and also that the BK-mediated Ca2+ release is impaired in PSS fibroblasts.
