    Notes: Limiting dilution analysis was performed in the presence of interleukin 2 (IL-2) on lymphocytes isolated from the synovial fluid (SF) and peripheral blood (PB) of patients with rheumatoid arthritis (RA) and PB of normal donors. Clones of theses ‘spontaneously’ IL-2-responsive cells from PB and SF were compared for their reactivity with component of the extracellular matrix (i.e. native or denatured type I or type II Collagen and proteoglycan). It was determined that all clones from both PB and SF were activated to produce interferon (IFN) in the presence of any of the connective tissue components (CTC) Clones derived from normal PH behaved in a similar fashion hut produced lower IFN-γ levels There was a synergy between the CTC and serum or plasma fibronectin, which was more apparent when soluble CTC were used as the stimuli rather than immobilized CTC. The fibronectin alone was unable to induce 1FN-γ production under any of the conditions tested (i.e. soluble or immobilized). These results demonstrate that clones of IL-2-responsivc T cells can be activated by interactions with connective tissue components lo produce IFN

    Notes: Malignant epithelioid schwannoma is a well-defined entity. However epithelioid tumours arising other than in a nerve trunk, in a benign nerve sheath tumour or in neurofibromatosis are problematical. We describe a case of a nodular tumour of the cheek which recurred twice without metastasis. Light microscopy showed an amelanotic epithelioid tumour. Immunohistology showed positive staining for vimentin and S100 protein and electron microscopy showed distinctive appearances suggestive of schwannoma. We discuss the differential diagnosis of the tumour, compare it with reported tumours resembling both melanoma and schwannoma and conclude that this may be a further example of the distinctive tumour reported as ‘malignant epithelioid schwannoma of superficial soft tissues’.

    Notes: Abstract Twelve poor sleepers of mean age 52 years performed 2 h of laboratory tests three times on 1 day during each week of nightly intake of loprazolam 0.5 mg, loprazolam 1 mg, triazolam 0.5 mg, or continued placebos. Only in the mornings did loprazolam 1 mg cause impairment in manual dexterity and card-sorting, and triazolam 0.5 mg in manual dexterity. Using difference from baseline, withdrawal of either drug after 3 weeks was associated with poorer subjective quality of sleep than following continuing placebos. The rate of spontaneous complaints (including accidents) associated with the drugs was as informative as formal testing.

    Notes: Summary We describe the range of phenotypes caused by cell death when larvae of the heat-sensitive cell-lethal mutant,l(1)ts726 ofDrosophila, are subjected to heat treatment at different stages of development. When the treatment extends into the pupal period, certain bristles fail to develop but the disc derivatives are otherwise normal. Earlier treatments cause the replacement of sets of leg and eye-antennal markers by mirror image duplications of neighbouring sets. The results are compared in detail with those expected under a gradient model proposed earlier to account for the phenotype. It is found that although the results for the second leg are in excellent agreement with the predictions of the model, a more elaborate hypothesis is necessary to account for the eye-antennal disc data. Abnormal head patterns fall into several distinct categories, any one of which could be explained by postulating the existence of a gradient, if other categories did not also occur. The markers affected in each case belong to overlapping sets, and each category of pattern can be induced by heat treatments administered throughout the temperature-sensitive period. The statistical distribution of the data is such that only one category of pattern would be detected in a small scale experiment. The possible implications relative to pattern formation in normal development are discussed.

    Notes: Summary Spontaneous mineralization observed in the thalamus, and meninges of the median longitudinal fissure, of several strains of mice was studied by light and electron microscopy. The mineralized deposits were closely associated with blood vessels, were periodic acid-Schiff positive, and contained variable amounts of calcium. The thalamic deposits were found to be in an extra-cellular location, and consisted of a randomly arranged mass of fine fibrils or spicules, with laminations of varying electron density in larger deposits. The deposits appeared to develop on the surface of subendothelial and medial, vascular basement membranes. Energy dispersive, X-ray microanalysis demonstrated the presence of both calcium and phosphorus in small and large deposits.

    Notes: Summary The genetics of peroxidase and leucine-aminopeptidase isozymes was studied utilizing starch gel electrophoresis in the diploidsMedicago sativa L. (M. coerulea Less.) andM. falcata L. Three anodal and one cathodal sets of peroxidase isozymes identify four linked loci. In addition, two anodal sets of leucine-aminopeptidase isozymes identify two loci that may be linked. The allozymes at each of the loci segregated as expected for monomeric enzymes. However in several crosses there were deficiencies in the number of progeny in particular genotypic classes. This could result from the segregation of recessive deleterious genes linked to some of the allozyme alleles. This is the first report of multiple loci and multiple alleles determining isozymes inMedicago.
