    Notes: We have isolated a hybrid gene, composed of the first 455 nucleotides of hisP and nucieotides 275–1107 of malK, the genes coding for the nucleotide-binding components of the high-affinity transport systems for histidine and maltose in Salmonella typhimurium, respectively. The fusion had occurred by recombination within 11 homologous base pairs located between the two DNA fragments. In the chimeric protein peptidic motifs A and B, proposed to be part of the nucleotide-binding fold, originate from HisP and MalK, respectively. Plasmid pES42-39, harbouring the hybrid gene, was shown to complement only a malK mutation but failed to complement a hisP deletion mutation. The chimeric protein was identified by immunoblotting as a protein with an apparent molecular mass of 49kDa. Removal of the C-terminal 77 amino acid residues from the chimeric protein resulted in the loss of function in transport. In contrast, 51 amino acid residues could be removed from the C-terminus of wild-type MalK without any effect. Upon overproduction the chimeric protein, as wild-type MalK, inhibited expression of the malB regulon. However, both truncated proteins, when overproduced, did not exhibit this activity. Based on these results, a tentative model of the functional domains of MalK is presented.

    Notes: In the framework of the European project aimed at the sequencing of the Bacillus subtilis genome the DNA region located between gerB (314°) and sacXV (333°) was assigned to the Institut Pasteur. In this paper we describe the cloning and sequencing of a segment of 97 kb of contiguous DNA. Ninety-two open reading frames were predicted to encode putative proteins among which only forty-two were found to display significant similarities to known proteins present in databanks, e.g. amino acid permeases, proteins involved in cell wall or antibiotic biosynthesis, various regulatory proteins, proteins of several dehydrogenase families and enzymes II of the phosphotransferase system involved in sugar transport. Additional experiments led to the identification of the products of new B. subtilis genes, e.g. galactokinase and an operon involved in thiamine biosynthesis.

    Notes: Abstract The problem of detection and sizing of fatigue cracks that are closed due to the reverse plastic deformation is addressed in this paper. In particular, it is suggested that the zone of plastic deformation can be identified by measuring the changes in the wave speeds in the plastically deformed material. This is supported through experimental results, first by determining the changes in wave speed with monotonic plastic deformation and then by investigating the plastic zones near fatigue cracks. The characteristic changes in the wave speeds are interpreted in terms of the theory of wave propagation in nonlinearly deformed media and elastic plastic fracture mechanics.

Therapeutische Vitamin C-Zufuhr ruft, wenn auch nicht regelmäßig, eine Abnahme des Globulin-N und eine Zunahme des Albumin-N hervor, wobei gleichzeitig eine Herabsetzung der Blutsenkungsgeschwindigkeit stattfindet. 2. Der Vitamin C-Gehalt des Blutserums scheint in der Jugend höher zu liegen als im Alter. Hypovitaminosen sind zur Zeit mit Hilfe von Serumbestimmungen noch nicht sicher erfaßbar. 3. Gleichzeitige Zufuhr von thyreotropem Hormon und Vitamin C ruft eine Vergrößerung und Gewichtszunahme der Nebennieren hervor, die, im Gegensatz zur alleinigen Hormonzufuhr, nicht auf Zunahme der Rinden-, sondern der Marksubstanz beruht.

    Notes: Zusammenfassung Vitamin A wird vom normalen Organismus im Harn nicht ausgeschieden. Selbst stärkste Belastung bedingt hierin keine Änderung. Dagegen wird die Vitamin A-Ausscheidung zu einem auffällig hohen Prozentsatz unter Krebskranken sowie bei den an Tuberkulose oder Allgemeininfektion Erkrankten gefunden. Als Ursache wird neben der Leberschädigung eine Veränderung der Nierenpermeabilität angesehen. Nach Menge und Dauer der Ausscheidung ist der entstehende Vitamin A-Verlust für die Ausbildung einer Hypovitaminose in Betracht zu ziehen. Es läßt sich eine Lebenskurve des A-Vitamins im Serum festlegen, der gegenüber das Vorliegen eines Hypovitaminismus bestimmt werden kann. 4. Der bei Ausschluß von Mangelernährung feststellbare Hypovitaminismus ist die Folge einer funktionellen Leberschädigung. 5. Gemessen an der Bildung des Methylglyoxals, führt Carotinzusatz eine Steigerung der Glykolyse herbei. Carotin und Vitamin A rufen eine kräftige Steigerung hervor, während sich ein Einfluß auf die Glykolyse durch die Ascorbinsäure nicht nachweisen läßt.

    Notes: Summary To determine the reproducibility of the first phase insulin response to intravenous glucose, ten normal subjects underwent two intravenous glucose tolerance tests separated by at least two weeks. Intravenous dextrose (0.3 g/kg) was administered over 2 min by continuous infusion and arterialised-venous samples were taken from a retrogradely cannulated hand vein in the opposite arm. Within subjects, median coefficient of variation for the 3 min insulin was 4.0% (range 1.2–24.3%) and median coefficient of variation for the 0–10 min area was 6.7% (range 1.7–18.8%). These coefficients of variation are close to those of the assay itself (〈 10%). Despite this, between subject responses varied by greater than sixfold. In conclusion, contrary to previous reports the intravenous glucose tolerance test is highly reproducible. This makes it a very valuable tool for further studies of the pathogenesis of diabetes.
