    Description / Table of Contents: Summary The effect of Levamisole on the human granulopoiesis was studied in patients randomized to receive, in addition to adjuvant chemotherapy for primary breast cancer, either no other treatment or additional unspecific immune therapy with Levamisole. The reaction of granulopoiesis to the cytostatic drugs, as characterized by changes of peripheral blood polymorphonuclear neutrophils (PMN), functional bone marrow granulocyte reserve, serial bone marrow cytology, and granulopoietic stem cells (CFU-C) in marrow and blood, was not affected by administration of Levamisole. The data support the concept that Levamisole has no direct effect on human bone marrow granulopoiesis, but that an allergic mechanism is involved in the pathogenesis of Levamisole-induced agranulocytosis. The expectation that Levamisole exerts a beneficial effect by stimulation of the granulopoiesis, as previously suggested for BCG and Corynebacterium parvum, could not be substantiated in our studies.
    Notes: Zusammenfassung Die Wirkung von Laevamisol auf die menschliche Granulopoese wurde bei Patientinnen untersucht, die nach operativer Behandlung eines primären Mammakarzinoms adjuvant chemotherapiert und bei denen ein Teil zusätzlich mit Laevamisol als einem unspezifischen Immunstimulans behandelt wurden. Die Reaktion der Granulopoese auf die zytostatische Therapie,- Veränderungen des peripheren Granulozytenpools, der funktionellen Granulozytenreserve des Markes, der Knochenmarkszytologie und der granulopoetischen Stammzellen (CFU-C) in Mark und peripherem Blut, — wurden durch die Gabe von Laevamisol nicht beeinflu\t. Diese Untersuchungen stehen mit der Vorstellung im Einklang, da\ Laevamisol keine direkte Schädigung der Granulopoese verursacht, sondern da\ ein allergischer Mechanismus der Laevamisol-Agranulocytose zugrunde liegt. Eine günstige Wirkung durch Stimulation der Granulopoese, wie sie für BCG und Corynebacterium parvum angenommen wurde, konnte für Laevamisol unter den Bedingungen dieser Studie nicht gefunden werden.

    Notes: Summary The antiproliferative effect of fibroblast interferon (IF-β) on the colony tumor growth of 42 miscellaneous carcinomas (15 gastric, 11 breast, eight colon, five ovarian, three bronchial) and two malignant melanomas were assayed in a semisolid, double-layered agar culture system. Of the 44 evaluable experiments only four tumor specimens (three breast-, one gastric carcinoma) showed at least a 30% inhibition of colony formation due to application of interferon at concentration of 1,000 IU/ml or less. In these cases, a dose response relationship of the interferon effect was obvious. Yet, in the majority of carcinomas investigated, interferon had no antiproliferative influence on colony tumor growth and was markedly inferior to a simultaneously tested cytostatic agent (adriamycin). Our in vitro results contradict a general direct cytotoxic action of interferon on human solid carcinomas.
