Notes: Basic aspects f fast atom bombardment mass spectrometry such as matrix effects, discriminatory effects between positive and negative ion formation, sensitivity, and isotope dilution have been investigated using oligopeptides and amino acid derivatives as model compounds. The matrix selected was found to have a decisive influence on the sensitivity of fast atom bombvardment. Acidic and basic properties of the samples could be correlated with different ionization efficiencies in positive or negative ion formation. With respect to sensitivity, a roughly linear relation between signal intensity and sample amount was observed in the range of 1 ng to 10 μg of sample. Nanogram amounts of samples could be detected by fast atom bombardment mass spectrometry. Finally, the suitability of fast atom bombardment for quantitative investigations is proven by the investigation of isotopically labelled mixtures.

Notes: The behaviour of substituted cyclopropenes under electron impact is to a large extent determined by the presence of the unsaturated three-membered ring which is capable of efficient delocalization of the positive charge. The loss of one of the substituents at the C(3) position of the small ring is characteristic for the fragmentation of cyclopropenes; the loss of the substituent which is less electron donating occurs preferentially. The presence of substituents with heteroatoms on the three-membered ring may lead to changes in the fragmentation scheme characteristic for the specific set of substituents.

Notes: Dinucleoside monophosphates bearing usual protecting groups at the exocyclic amino functions of the heterocyclic bases, the 5′-hydroxy group, the phosphate moiety and the O-6-position of the guanine ring have been analysed by FAB mass specrometry in the negative ion mode. These non-charged key intermediates for oligodeoxynucleotide synthesis exhibit very simple spectra in the mass range of m/z 400-1500 using tetra-ethylene glycol as matrix. Most of the observed ions could easily be assigned. Apart from deprotonated molecular ions sequence specific ions arising from C-O-P bond cleavage and ions derived from loss of protecting groups were observed. The simplicity of spectra interpretation makes FAB mass spectrometry to a rapid and valuable tool for unambiguous identification of dimeric synthons for oligonucleotide synthesis.

Description / Table of Contents: Arc-PVD-Coating of Cemented Carbides and Analysis of their Functional Behaviour in Interrupted Cut MachiningInterrupted machining of quenched and tempered steel demands the application of tools with high resistance against mechanically and tribologically induced wear. These demands can not be fully satisfied by uncoated carbides. Also CVD-coatings give only some improvement due to a certain embrittlement of the substrate during the coating process. PVD-coatings deposited at low temperatures without affection of the substrate lower the influence of the relevant wear mechanisms preferably at moderate cutting speeds. The combination of tough substrates with structural and compositional optimized coatings results in a multiplication of tool life within the whole range of low and medium cutting speeds. Coatings with homogenious structure, compressive residual stresses and high content of metalloides ensure excellent resistance against wear.Titanium-carbonitride deposited on carbide was found to be superiour compared with other carbonitride, nitride and carbide coatings within the system Ti—Zr—C—N due to excellent tribological properties. It was optimal suited for the interrupted cut application.

Notes: The properties of hexakis(3-O-acetyl-2,6-di-O-pentyl)-α-cyclodextrin as a chiral stationary phase for capillary gas chromatography are described. For the first time the enantiomers of a series of different lactones are separated and their order of elution is assigned. Moreover, the enantiomers of trifluoroacetylated aldols and amino alcohols, the cyclic carbonates of 1,2-diols, 1,3-diols, O-alkylated glycerols, and some chiral pharmaceuticals are also separated on the new chiral phase. The modified α-cyclodextrin is stable above 200°C.

Notes: The reaction products of trifluoroacetic anhydride with peptide amides are identified as stable heterocyclic imidazolin-5(4)-ones, which can be analysed by gas chromatography mass spectrometry. The mass spectra show intense molecular ions and allow the assignment of the amino acid sequence.
