    Notizen: Many of the chemicals in common use possess, to some degree, a capacity to cause skin sensitization. Consequently, it is important to conduct a thorough and accurate risk assessment when it can be anticipated that such chemicals are likely to come into contact with human skin. Such risk assessments must consider the nature and extent of dermal exposure together with the sensitizing potency of the chemical. Whilst the exposure elements of the risk assessment process are often well understood, or can at least be reasonably predicted on the basis of the proposed usage of the chemical, the determination of skin sensitizing potency may be problematic. In this paper, we describe an approach to the determination of relative potency of 3 isothiazolinone biocides. Initially, the local lymph node assay (LLNA) provides an estimate of relative sensitizing potency. Where appropriate, this can be followed by confirmatory testing in the human repeat insult patch test (HRIPT). The data generated reveal that (chloro)methyl isothiazolinone (CMI) is a significantly stronger sensitizer than either methyl trimethylene isothiazolinone (MTI) or benzisothiazolinone (BIT). Furthermore, both the LLNA and the HRIPT data show consistently that BIT is a somewhat weaker skin sensitizer than MTI. As an important additional component of the safety evaluation, the ability of these sensitizing isothiazolinones to cross-react has been considered; the data indicate that the more weakly sensitizing isothiazolinones do not elicit allergic reactions in subjects sensitized to MCI. The implications for use of these biocides in various product types are discussed.

    Notizen: The potential antagonistic effects of Ca2+, Cu2+, Fe2+, Mg2+, Mn2+ and Zn2+ on contact allergy to Co2+ and Ni2+ were studied. The immune response was characterized by the Co2+ or Ni2+ mediated cellular [methyl-3H]thymidine uptake in peripheral blood mononuclear cell (PBMC) cultures from 6 subjects contact-allergic to Co2+ and Ni2+ and 6 non-allergic control individuals. Results from the in vitro experiments were further evaluated with Co2+-sensitized guinea pigs according to the modified Freund's complete adjuvant test. Ni2+ and Co2+ (10–50 μM) significantly increased the lymphocyte proliferation in PBMC cultures from contact-allergic subjects in comparison with those from control individuals. Pretreatment of the PBMCs with Ca2+, Fe2+, Mg2+ (10–100 μM) or Mn2+ (1–10 μM) did not influence, while Zn2+ (100 μM) enhanced, and Cu2+ (5 and 10 μM) markedly reduced the Ni2+ and Co2+ mediated cellular [methyl-3H]thymidine uptake. The inhibition of the Ni2+- and Co2+-induced cell proliferation by Cu2+ in vitro was probably related to toxicity, since the viability of the cells was significantly reduced by applied combinations of Ni2+ or Co2+ with Cu2+. Topical pretreatment of Co2+-sensitized guinea pigs with maximum non-irritating doses of CuCl2· 2H2O (0.8%) did not affect the challenge testing to CoCl2· 6H2O (0.1 and 0.3%). In conclusion, our combined in vitro and in vivo results indicate that Ca2+, Cu2+, Fe2+, Mg2+, Mn2+ and Zn2+ are not able to antagonise the formation of Ni2+ and Co2+ antigens.

    Notizen: SUMMARY.— In eczema patients with nickel allergy, immunogtobulin E (IgE) and the threshold of sensitivity were determined by means of patch testing with dilutions of nickel sulphate (NiSO4). In 4 of the 47 patients investigated the IgE value was raised. Two of these were women in whom, at the age of 20, the onset of nickel eczema took place on the ear lobes. The occurrence of atopic diseases in the patients was of the same frequency (9%) as in other investigations, whereas the frequency among relatives was higher (38% as against 19–22%). The mean threshold of sensitivity in 53 patients was 0.43% (distilled water) and 0.51% NiSO4 (petrolatum). Ten patients reacted to concentrations below 0.039% in distilled water. The lowest thresholds were obtained in patients who were simultaneously sensitive to both nickel and cobalt. No connection was observed between the severity of the skin disease and the threshold of sensitivity. With higher concentrations of nickel in 3 non-atopic patients, pustular test-reactions were observed, which disappeared at lower concentrations. Clinically, nickel eczema seems to have changed its character compared with earlier reports: 16 patients reported that the onset had been on their hands; and at their first consultation 25 patients had hand eczema; in another 16 patients other parts of the body were also affected.

    Notizen: In 25 consecutive cases of chromium contact allergy we have made four parallel serial dilution tests and irradiated three of the test rows, two with solar-spectriim-like radiation from a 150 W Xenon lamp (4/5 MED and 4 MED) and one row with long-wave ultraviolet radiation (blacklight) alone.Comparing an unirradiated row (control) and the row irradiated with 4/5 MED, the reactions were significantly more intense in 48% of the cases, i.e. the patients reacted at lower chromium concentrations.In a corresponding comparison with 4 MED a significantly more intense reaction was obtained in 36% of the cases, whereas blacklight showed no significant difference compared with the control row.The most intense reactions were obtained in the cases which were anamnestically photosensitive and had low MED values.

    Notizen: During 1976–77 a field investigation was carried out into skin diseases among house painters in Stockholm, Sweden. The study was divided into three stages: I. questionnaire investigation, II. examination of painters who reported skin diseases, and III. patch testing and final classification. Of the 2,622 painters registered on 30 November 1976, 2239 (85.4 %) answered the questionnaire. Current dermatoses were reported by 373 painters (16.7 %) and of these, 227 were examined by us, 40 had recently been examined, and 106 did not attend. Occupational contact eczemas were diagnosed in 87 cases, and it was doubtful if the dermatoses were occupational in an additional 56 cases. A prevalence of 3.9 % was suggested, but this represents a minimum figure. Chloracetamide was found to be an important cause of occupational contact eczema. A personal and/or family history of atopy was common among the affected painters. Solvents were widely used for skin cleansing; there is a great need for continuous information on protective measurements, and on skin cleansing and care. It is concluded that field investigations give higher and probably more reliable prevalence figures for occupational dermatoses than data compiled from outpatient clinics.
