Notes: Abstract The relevance of tumor necrosis factor-α (TNF-α) inducing early inflammatory reactions in the liver after hemorrhagic shock, for example, leukocyte adhesion, has been well described. This study evaluated the anti-inflammatory effects of a monoclonal antibody against TNF-α (TN3.19.12) in terms of the time of application, namely, prior to shock induction, at the time of resuscitation, and after resuscitation. The hepatic micro-circulation was investigated by intravital fluorescence microscopy in female Sprague-Dawley rats undergoing severe hemorrhagic shock for 60 min and subsequent resuscitation. TN3.19.12 or placebo was given in a randomized and blinded manner either 60 min prior to shock induction, l min prior to resuscitation, or 15 min after the onset of resuscitation. The number of firmly adherent leukocytes in the livers of treated animals depended on the time of application of TN3.19.12. Leukocyte adhesion was significantly reduced when TN3.19.12 was given prior to shock induction or at the time of resuscitation and was less effective when administered after the onset of resuscitation. The results further confirm that TNF-α initiates very early pathological leukocyte adhesion in the liver 5 h following shock. Inhibition of leukocyte adhesion after shock, however, depends strongly on the time of TNF-α blocking. While TN3.19.12 prior to shock induction resulted in most effective attenuation, only very early treatment allowed limitation of posttraumatically increased leukocyte adhesion.

Notes: Abstract The indole alkaloid ibogaine (NIH 10567, Endabuse) is currently being examined for its potential utility in the treatment of cocaine and opioid addiction. However, a clearly defined molecular mechanism of action for ibogaine's putative anti-addictive properties has not been delineated. Radioligand binding assays targeting over 50 distinct neurotransmitter receptors, ion channels, and select second messenger systems were employed to establish a broad in vitro pharmacological profile for ibogaine. These studies revealed that ibogaine interacted with a wide variety of receptors at concentrations of 1–100 µM. These included the mu, delta, kappa, opiate, 5HT2, 5HT3, and muscarinic1 and 2 receptors, and the dopamine, norepinephrine, and serotonin uptake sites. In addition, ibogaine interacted withN-methyl-d-aspartic acid (NMDA) associated ion and sodium ion channels as determined by the inhibition of [3H]MK-801 and [3H]bactrachotoxin A 20-α-benzoate binding (BTX-B), respectively. This broad spectrum of activity may in part be responsible for ibogaine's putative anti-addictive activity.

Notes: Abstract: Based on a microscopic density functional theory we calculate the internal structure of the three-phase contact line between liquid, vapor, and a confining wall as well as the morphology of liquid wetting films on a substrate exhibiting a chemical step. We present a refined numerical analysis of the nonlocal density functional which describes the interface morphologies and the corresponding line tensions. These results are compared with those predicted by a more simple phenomenological interface displacement model. Except for the case that the interface exhibits large curvatures, we find that the interface displacement model provides a quantitatively reliable description of the interfacial structures.

Notes: Abstract The behavior of a water soluble calixarene ligand has been investigated in an (NH4)2SO4/polyethylene glycol-2000 (PEG) aqueous biphasic system. Spectroscopic measurements indicate that 4-sulfonic-calix-6-arene partitions preferentially to the polymer-rich phase in this system. The utility of this ligand as a metal ion extractant was also investigated. The addition of the calixarene (10−2M in initial PEG-rich stock solution) did not significantly enhance the partitioning of Na+, Cs+, Sr2+, Ba2+, Co2+, Cd2+, or UO 2 2+ from the (NH4)2SO4-rich to the PEG-rich phase. The partitioning behavior of these metal ions is dependent, rather, on their Gibbs free energy of hydration (ΔGhyd *).

Notes: Abstract Two surface antigens ofEimeria nieschulzi merozoites were characterized by monoclonal antibodies (Mcab) as follows: an antigen of 31 (reducing conditions), 25 and 31 kDa (nonreducing conditions), by Mcab A1F6; and an antigen of 52 and 30 kDa (reducing conditions) or 42 kDa (nonreducing conditions), by Mcab H7. The localization of the antigens in fluorescence and electron micrographs is presented. Despite being accessible to antibodies on live (nonpermeabilized) parasites, neither antigens were significantly labelled using lactoperoxidase radioiodination.
