    Notes: We present a strategy for solvating biomolecules in molecular dynamics or Monte Carlo simulations. The method employs a thin layer (often monomolecular) of explicit water with additional external forces representing the electrostatics, pressure, fluctuations, and dissipations caused by the neglected bulk. Long-range electrostatic corrections are supplied through a set of variable surface charges (polarons) that recreates the mean reaction field (or dielectric properties) of an infinite solvent. We refer to this "fictitious" boundary layer as a "surface of active polarons" (or SOAP). Test simulations of the solvation free energies of 15 amino acid analogs and nine ions are in good agreement with experiment (correlation coefficients: 0.995 and 1.000, respectively) despite the use of unaltered published force-fields with only one adjustable parameter. Dynamical capabilities of SOAP are illustrated by application to a six residue peptide with a stable conformation (SYPFDV), as well as a flexible nine residue HIV-1 gp120 peptide (TLTSCNTSV from PDB 1hhg). Future extensions, calibrations, and applications are discussed briefly. © 2000 American Institute of Physics.

    Notes: Point defects have been observed directly in non-stoichiometric niobium oxides, Nb12O29, by high- resolution electron microscopy at a resolution of 3-4 Å. A proposed model for the point defect is a complex composed of two interstitial metal atoms and two interstitial oxygen atoms. Observations have been made on the annealing out of these defects under electron irradiation. Evidence on the density of these defects offers a semi-quantitative explanation of how the non-stoichiometric oxygen excess in these oxides is accommodated in the matrix structure.

    Notes: With the use of high-resolution electron microscopy, defects have been observed in crystals of Nb22O54: two types of point defects and extended Wadsley-type defects having a structure corresponding to Nb12O29. For one type of point defect a model is proposed which contains two displaced metal atoms and two interstitial oxygen atoms within a 'block'. For the other, one metal atom and three oxygen atoms are considered to be missing from a tetrahedral site. Contributions made by the three types of defects to the non-stoichiometry of the crystals are discussed.

    Notes: The mesogloea and skin of a common edible jellyfish, Stomolophus nomurai, were characterized with respect to amino acid composition and compared with a commercially salted jellyfish. Then the mesogloea was digested with pepsin at 3°C for 48 hr and its solubilized protein was isolated and subjected to biochemical analyses. These composite results have shown that the major edible component of jellyfish was the connective tissue protein, collagen, characterized by its high content of hydroxylysine and its glycosides.

    Notes: Two different types of T cell clones which recognize Mlsb haplotype gene products were established by using mice congenic at the Mls locus. The antigens defined by these T cells showed differences not only in strain distribution but also in the restriction element for antigen recognition by T cells (i.e. Kd or Dd). The antigens appear to be expressed on various types of lymphocytes.

    Notes: Three cases of trichilemmal keratosis (horn) were light microscopically examined and all showed numbers of U- or V-shaped epidermal proliferations which keratinized in a fashion either identical or similar to trichilemmal keratinization. Electron microscopy revealed both uneven and linear borders between the keratinized and the keratinizing cells with a few keratohyalin droplets, remnants of desmosomes, no marginal band in the horny layer, perinuclear vacuolation, few spherical bodies in the intercellular spaces (ICS) of the upper epidermis, and widening of the ICS of the lower epidermis. A number of electron dense spherical particles. 40-50 nm in diameter, were observed in nuclei of the upper epidermis. This suggests that ultrastructure of trichilemmal keratosis is similar rather to viral warts than to trichilemmal cysts, although there are close similarities between trichilemmal keratosis and cyst.

    Notes: A tumor of 60 years' duration overlying the sternum of an 87-year-old woman is described. Histologically, the tumor was composed of lobules and cords of epithelial cells with finely granular, eosinophilic cytoplasm. The epithelial nests were floating in large pools of mucinous stroma, separated by thin fibro-vascular septa. Cytoplasmic argyrophilia was seen by Grimelius silver stain. Positive reactions of the tumor cells were also found for chromogranin and neuron-specific enolase. Ultrastructural study revealed that the cytoplasm of the tumor cells contained two types of granules: neurosecretory and mucinous. From these findings, the present tumor is considered to be a carcinoid tumor with amphicrine cells, and the term ‘mucinous carcinoid’ of the skin seems to be appropriate for the diagnosis of the present case.

    Notes: The body wall collagen of an edible sea cucumber, Stichopus japonicus, was studied with respect to its chemical composition and subunit structure. About 70% of the total body wall protein was accounted for by highly insoluble collagen fibers. The disaggregation with β-mercaptoethanol, 0.1 M NaOH treatment, and limited pepsin digestion of these collagen fibers resulted in complete solubilization. The solubilized collagen was isolated and characterized; it had 2 distinct subunits, αl and α2, which formed (α1)2α2 heterotrimers and was rich in glutamic acid when compared with other fibrillar collagens. The unique textural properties of cooked sea cucumber seem to be due to thermal denaturation of the insoluble collagen fibers.
