    Notes: Recombinant E. coli clones expressing a 50-kDa poly-histidine tail tagged antibody fragment against botulinum toxin (bt-Fab) were initially screened for yield and binding affinity. One clone was selected for bioprocess development. The selected bt-Fab vector was induced by addition of IPTG and the protein was targeted to the periplasm by inclusion of a pelB leader sequence. A histidine6 affinity ligand at the heavy chain C-terminus facilitated single-step purification by immobilized metal-affinity chromatography (IMAC). Notably, the effects of post-induction temperature on bt-Fab expression and downstream purification were evaluated. Our results demonstrated that fermentation conditions interfered with purification on the IMAC column at 37°C. Protease analysis by gelatin polyacrylamide gel electrophoresis (GPAGE) indicated the presence of a membrane-bound ∼39 kDa protease activity shortly after induction. The appearance of the protease activity was inversely correlated with the bt-Fab yield. The protease was purified and was shown to degrade bt-Fab. A simple kinetic model was developed describing temporal regulation of protease and bt-Fab degradation. Partially degraded bt-Fab was unrecoverable by IMAC, presumably due to the loss of the His6 affinity ligand. The amount of purified bt-Fab obtained per liter of fermentation broth was typically ∼1 mg.

    Notes: Summary Preclinical evidence suggests that the D-1 dopamine receptor contributes to the generation of behaviors used as models for human extrapyramidal disorders. To evaluate the potential of D-1 receptor stimulation in neurologic disease, SKF 38393, a selective D-1 dopamine receptor agonist, was administered to seven patients with idiopathic Parkinson's disease in a double-blind, placebo controlled study. SKF 38393 was found to be rapidly absorbed when administered orally, and to occur in micromolar concentrations in spinal fluid. No change in scores of parkinsonian severity were noted when SKF 38393 was administered alone, or when the drug was combined with intravenous levodopa. The results support the view that the pathophysiology of Parkinson's disease may relate exclusively to the D-2 subclass of dopamine receptors.

    Notes: Summary In animal models of Parkinson's disease (PD), glutamate antagonists diminish levodopa (LD)-associated motor fluctuations and dyskinesias. We sought to investigate if these preclinical observations can be extended to the human disease, by evaluating the effects of three non-competitive NMDA antagonists (dextrorphan, dextromethorphan and amantadine) on the motor response to LD in patients with advanced PD. In four separate trials, adjuvant therapy with these drugs reduced LD-induced dyskinesias and motor fluctuations. These findings support the view that drugs acting to inhibit glutamatergic transmission at the NMDA receptor can ameliorate LD associated motor response complications.

    Notes: Abstract The cellobiohydrolase II (CBHII) of Microbispora bispora, originally cloned in Escherichia coli, was subcloned into Streptomyces lividans using shuttle vectors pSKN 01 and pSKN 02. The enzyme was secreted from Streptomyces, whereas it was intracellular in E. coli. The yields of CBHII produced by S. lividans transformants were 15–20-fold higher than those produced by E. coli transformants. The optimal pH of M. bispora native cellobiohydrolase and the cloned enzyme from S. lividans is 6.5. The thermal and pH stability of CHBII produced in M. bispora, E. coli and S. lividans were compared. Enzyme produced in E. coli was inactivated more rapidly (k = 0.252 min−1 at 90° C; 90% inactivation after 10 min vs. 0.119 min−1 for the others). CBHII was monitored following electrophoretic separation by reaction with a monoclonal antibody. The apparent molecular mass of the protein produced from the S. lividans clone was 93 kDa, the same as that of the native enzyme, but that of the enzyme produced in E. coli was smaller (82 kDa).

    Notes: Abstract Zymomonas mobilis is a potential candidate for fuel ethanol production because of its high ethanol productivity. A key enzyme in ethanol production is alcohol dehydrogenase (ADH). Z. mobilis possesses two isozymes, ZADH-1 and ZADH-2. To clarify their physiological roles, mutants with modified ADH were isolated by selection based on resistance to allyl alcohol. From the physiological characteristics of a ZADH-2-negative mutant, it is suggested that ZADH-1 functions as the major ADH, while ZADH-2 could become functional in the latter stages of fermentation.

    Notes: Abstract Monterey Fan is the largest modern fan off the California shore. Two main submarine canyon systems feed it via a complex pattern of fan valleys and channels. The northern Ascension Canyon system is relatively inactive during high sea-level periods. In contrast, Monterey Canyon and its tributaries to the south cut across the shelf and remain active during high sea level. Deposition on the upper fan is controlled primarily by the relative activity within these two canyon systems. Deposition over the rest of the fan is controlled by the oceanic crust topography, resulting in an irregular fan shape and periodic major shifts in the locus of deposition.

    Notes: Abstract This ten-year general circulation model experiment compared a simulation where land surface boundary conditions were represented by observed, present day land cover to a simulation where the surface was represented by natural, potential land cover conditions. As a result of these estimated changes in historical land cover, significant temperature and hydrology changes affected tropical land surfaces, where some of the largest historical disruptions in total vegetation biomass have occurred. Also of considerable interest because of their broad scope and magnitude were changes in high-latitude Northern Hemisphere winter climate which resulted from changes in tropical convection, upper-level tropical outflow, and the generation of low-frequency tropical waves which propagated to the extratropics. These effects combined to move the Northern Hemisphere zonally averaged westerly jet to higher latitudes, broaden it, and reduce its maximum intensity. Low-level easterlies were also reduced over much of the tropical Pacific basin while positive anomalies in convective precipitation occurred in the central Pacific. Globally averaged changes were small. Comparisons of recent, observed trends in tropical and Northern Hemisphere, mid-latitude climate with these simulations suggests an interaction between the climatic effects of historical land cover changes and other modes of climate variability.
