    Notes: Screening for B-cell-specific antibodies in unabsorbed pregnancy sera preselected for weak reactivity in regular HLA-A, -B, and -C screening yielded a relatively high proportion (35/81) of B-cell-specific antibodies. Most B-cell-specific antibodies react broadly, showing inclusion phenomena suggesting ‘cross-reactivity’ analogous to that observed in HLA-A and -B serology. In control experiments with T-cell-enriched suspensions three antisera reacted with T cells and not with B cells. These antisera are highly associated with HLA-A2 in the unrelated population and segregate with HLA haplotypes in families and with HLA-A in a family with HLA-A, B recombination. Thus it appears that the human equivalents of la antigens may include—in analogy to the murine Ia antigens—B-cell- as well as T-cell-specific alloantigens.

    Notes: The association of HLA-A and -B antigens with Bf alleles was investigated in 200 parents from 100 unrelated families. There were significant associations between HLA-A3 and Bf-F, B7 and Bf-S, B8 and Bf-S, B12 and Bf-F, and BW35 and Bf-F. Three-point HLA-A,B,Bf haplotype frequencies, linkage disequilibrium parameters, and chi-square values were determined both from the genotype and from the pheno-type data. Although the HLA-B,Bf associations involve antigens that are also present in the highly associated A,B and B,D haplotypes of the Caucasian population, there was—with the possible exception of HLA-A3,B7,Bf-S–no significant three-point association for HLA-A,B,Bf.

    Description / Table of Contents: Zusammenfassung Säurefeste Spindelkörperchen wurden bisher nur in Lymphknoten beobachtet. Ihre Natur und ihre Ätiologie blieben unklar. Wir fanden sie auch in der menschlichen Milz. Ihr färberisches Verhalten und die Beziehungen zwischen ihrem Auftreten und dem Lebensalter, dem Geschlecht und dem Milzgewicht sowie zu bestimmten Krankheitsgruppen und Hämosiderinablagerungen wurden untersucht. Sie waren in 50% der Milzen bei hereditärer Sphärocytose und in 41,7 % der Milzen mit traumatischer Ruptur nachweisbar, dagegen nur in 2,3% aller anderen untersuchten Fälle verschiedener anderer Erkrankungen. Aus unseren Ergebnissen können wir schließen, 1. daß die säurefesten Spindelkörperchen aus Ceroid bestehen, 2. daß sie keine Erreger darstellen und somit keine Rolle für die Ätiologie der Sarkoidose spielen, 3. daß sie bei verstärktem Erythrocytenabbau gebildet werden können, 4. daß sie wahrscheinlich durch oxydative Polymerisation von Membranlipiden entstehen, 5. daß sie auch ohne Fettstoffwechselstörungen auftreten können.
    Notes: Summary Spindle-shaped acid-fast bodies have been identified only in lymph nodes till now. Their nature and their origin remained unclear. We have also found these bodies in the human spleen. Their staining reactions and their correlations to age, sex, weight of spleen, and to hemosiderin deposits were examined. They have been encountered in 50% of cases of hereditary spherocytosis and in 41.7% of cases of traumatic rupture of the spleen—but only in 2.3% of all other cases of groups of various other disorders. We conclude from our results that: 1. The spindle-shaped acid-fast bodies are made of ceroid. 2. They are not causative organisms and therefore cannot be of importance in the etiology of sarcoidosis. 3. They derive from increased destruction of erythrocytes. 4. They originate due to oxidative polymerization of membrane lipids. 5. They may also be found in absence of any fat-metabolism disturbance.

    Description / Table of Contents: Zusammenfassung Es wurde eine Mikromethode zur Bestimmung der Fettsäurezusammensetzung der myelintypischen Glykosphingolipide (Cerebroside und Sulfatide) entwickelt, welche für entsprechende Untersuchungen bei verschiedenen neurologischen Erkrankungen, insbesondere Entmarkungskrankheiten, eingesetzt werden kann. Ausgehend von 1 g Gewebsmaterial werden die Lipide extrahiert, die Phospholipide an aktiviertem Florisil abgetrennt und die Glykolipide dünnschichtchromatographisch in je 2 Cerebrosid- und Sulfatidfraktionen aufgetrennt. Diese können dann einzeln untersucht werden. Die Ergebnisse der Fettsäureanalysen von 13 autoptisch gewonnenen normalen Gehirnproben erwachsener Personen besitzen nur eine geringe Streubreite, was besonders für die langkettigen Fettsäuren gilt. Die Befunde mit unserer Mikromethode stehen in guter Übereinstimmung mit den Werten anderer Autoren.
    Notes: Summary A micromethod for the investigation of the fatty acid composition of myelin glycosphingolipids (cerebrosides and sulfatides) suitable for general application in the investigation of neurological disorders, especially demyelinating diseases, is presented. Using the lipids extracted from 1 g of material these are freed of phospholipids by Florisil column chromatography and separated by thin-layer chromatography into 2 cerebroside and sulfatide fractions which are analyzed individually. The results obtained from the white matter of 13 normal adult brains are distributed within a narrow range which is most pronounced for the group of long chain fatty acids. Our results also agree with those quoted from literature.

In der Marksubstanz von 17 normalen erwachsenen Vergleichshirnen sind die wichtigsten Fettsäuren C 18:0 und C 24:1. Unsere Ergebnisse stimmen gut mit den Ergebnissen anderer Autoren überein. Bei Kindern findet sich eine relative Vermehrung kurzkettiger Fettsäuren, diese Verschiebung ist typisch für „unreifes“ Myelin. Ähnliche Veränderungen werden hier auch bei Greisen und Fällen mit unspezifischem Hirnschaden (Autolyse), chronischer Urämie und juveniler Chorea Huntington (alle in Verbindung mit diffuser Entmarkung) beschrieben. Man kann davon ausgehen, daß die Fettsäurezusammensetzung von Sphingomyelin einen empfindlichen Maßstab sowohl für eine gestörte Myelinisierung wie auch für Demyelinisierung darstellt.
    Notes: Summary A micromethod for the investigation of the fatty acid composition of sphingomyelin is presented. In the cerebral white matter of 17 normal adult brains, analyzed for reference, the predominant fatty acids are C 18:0 and C 24:1. Our results are in agreement with those of other authors. Short chained fatty acids are relatively increased in young children; this shift is typical of “immature” myelin. Similar changes are described here in old persons and cases of non-specific brain damage associated with demyelination (autolysis, chronic uremia, juvenile chorea). Sphingomyelin fatty acid composition can be considered a sensitive measure of both disturbed myelination and demyelination.

    Notes: Abstract Cadmium uptake has been studied in starved and fed Mytilus edulis L. It is suggested that fairly elevated cadmium contents in fed mussels are not due to contaminated food, but to increased pumping rate when food is available. Highest concentration and main body burden are found in the mid-gut gland. Transport via haemolymph, and selective discrimination at the basement lamina of the mid-gut gland tubuli are regarded as mainly responsible for accumulation. Mercury seems to be processed in a similar way as cadmium. In the tubuli, both metals are immobilized in membrane-bound vesicles, which are finally defaecated.

    Notes: Summary The plasma level of free fatty acids (FFA) in adrenalectomized rats increases by 50% after treatment with aldosterone (2 μg/100 g rat). Lipolytic activity in peripheral fat tissue is lowered after adrenalectomy and doubles after in vivo administration of aldosterone to adrenalectomized rats (measured as free fatty acid release in vitro from epididymal fat tissue). Lypolysis of adipose tissue stimulated by the in vitro presence of ACTH also increases after in vivo administration of aldosterone. Incorporation of intravenously administered label from U14C-palmitate into total extractable lipid of renal tissue is augmented 3 h after aldosterone administration to adrenalectomized rats, while no increase of the radioactivity is observed in total lipid from liver tissue. Treatment with aldosterone does not affect the total lipid content of kidney or liver in adrenalectomized rats. The oxygen consumption rate of kidney cortex slices with lactate, β-hydroxybuterate or acetoacetate as substrates is lowered after in vivo administration of aldosterone to adrenalectomized rats. With succinate, however, the respiratory rate of kidney slices increases after aldosterone treatment of adrenalectomized rats, the ouabain-sensitive respiration being more affected than the ouabain-insensitive respiration. An interpretation of the O2 consumption data implicating competition of lipid metabolism for CoA-SH is discussed.

    Notes: Summary The effects of theophylline (3×10−5 M–5×10−3 M), isoprenaline (10−6 M), excess calcium (7.2 mM) and ouabain (3×10−7 M) on force of contraction and c-AMP content were studied in electrically driven (frequency 3 Hz) left auricles isolated from guinea pigs pretreated with reserpine. 1. Theophylline (2×10−3 M) increased c-AMP consistently. The theophylline-induced increase in c-AMP proceeded very rapidly. It became significant after an incubation period of 5 s, reached its peak after 5 min and then remained stable. The positive inotropic effect of theophylline also developed very rapidly, reached its peak after 1 min and then declined. The increase in force of contraction corresponded to the increase in c-AMP, but the decline of the positive inotropic effect occurred while c-AMP remained elevated. 2. The concentration-response curves for the theophylline-induced increases in c-AMP and force of contraction were virtually parallel at concentrations up to 2×10−3 M. 3. Washout of the effects of theophylline on c-AMP and force of contraction was also similar. Complete reversal occurred within 15 min. 4. The effects of theophylline on c-AMP and force of contraction were not affected by 10−7 M propranolol added 30 min before theophylline. 5. Isoprenaline (10−6 M) increased c-AMP and force of contraction to approximately the same extent upon incubation for up to 30 min. 6. Ouabain (3×10−7 M) or the elevation of the extracellular calcium concentration to 7.2 mM increased the force of contraction without producing a detectable increase in c-AMP. 7. It is concluded that the theophylline-produced increase in force of contraction may be the result of an increase in c-AMP and a subsequent increase in slow inward current. This effect may be counteracted by a direct, c-AMP independent effect of theophylline on intracellular calcium stores which could account for the decline of the positive inotropic effect of theophylline during prolonged exposure and at high concentrations. 8. The present data also support the view that the positive inotropic effect of isoprenaline is mediated by c-AMP, whereas the effects of ouabain and excess calcium on force of contraction appear to be independent of the c-AMP system.
