    Notes: A multidomain boundary element formulation for the analysis of general two-dimensional plane strain/stress crack problems is presented. The numerical results were accurate and efficient. The analyses were performed using traction singular quater-point boundary elements on each side of the crack tip(s) with and without transition elements. Traction singular quarter-point boundary elements contain the correct √r displacement and 1/√r traction variations at the crack tip. Transition elements are appended to the traction singular elements to model the √r displacement variation. The 1/√r traction singularity is not represented with these elements. Current research studies for the crack propagation analysis of quasi-static and fatigue fracture problems are discussed.

    Notes: In this note, we give the basis functions (shape functions) for Hsieh-Clough-Tocher triangles, complete or reduced. In order to take the best advantage of symmetry and for simplicity, we derive these functions for a general triangle by using simultaneously the ‘area co-ordinates’ and the so-called ‘eccentricity parameters’. We conclude with some remarks concerning the implementation.

    Notes: This paper considers some consequences of the common implementation of the finite element method in which element integrals are evaluated numerically by Gaussian quadrature. It shows that if the order of the Gauss scheme is less than the number of basis functions on an element, then the numerically evaluated capacity matrix for that element is singular. The capacity matrix of an assembly of such elements is also considered and is shown to be singular under certain conditions.

    Notes: This paper is concerned with the large-deflection (bending and free vibration)analysis of thin elastic curved beams by conventional and mixed finite element methods. The conventional finite element method is based on the total potential energy expression, whereas the mixed method is based on a Reissner-type variational statement and involves the bending moments and deflections as primary dependent variables. Incremental and direct formulations are presented for both methods. The nonlinearity is included in the numerical method via an iterative procedure (i.e. the in-plane force due to the large deflection is not treated as constant but included under the integral in the energy expression). In the case of vibrations, the transverse shear and rotary inertia effects are also included. A number of numerical examples of beams with various edge conditions are analysed for deflections and natural frequencies, and the results are compared with those reported in the literature.

    Notes: The stability of plane Poiseuille flow and circular Couette flow are examined with respect to linear azimuthally periodic disturbances by the finite element method. In the case of Couette motion, solutions are obtained for a narrow gap, a wide gap and a dilute polymer solution with an elongational viscosity in the narrow gap limit when both cylinders rotate at almost equal speed in the same direction. Results are in good agreement with previous calculations by other numerical methods.

    Notes: The use of the finite element concept in the time domain has resulted in a reinterpretation of many previously recorded recurrence formulae and the discovery of new ones. An important new feature common to all such schemes is the fact that a weighted forcing term is used. This precludes the application of unrealistic forcing terms resulting in a direct finite difference derivation and ‘smooths’ oscillation due to sudden changes of boundary condition. This problem is discussed in the context of various two- and three-level time schemes.

    Notes: A finite element formulation for the solution of time-dependent inelastic deformation problems for metallic structures, in the presence of transient thermal stresses, is presented in this paper. A rate formulation of the equations is used and any of a number of recently proposed combined creep-plasticity constitutive models with state variables can be adopted to describe material behaviour. The computer program developed can solve planar (plane strain and stress) and axisymmetric problems. Using one of the above-mentioned constitutive models, numerical results are presented for several illustrative problems, and comparisons of results, using either the quasi-steady or the unsteady diffusion equation for the determination of the temperature field, are carried out.

It is a generalization of the algorithm given by Taylor. 1Theory is developed for elements which are loaded under plane stress, plane strain or axisymmetric conditions, and two FORTRAN IV subroutines are included. These subroutines, which are presented in a modular form, are applicable to any type of serendipity, Lagrangian or triangular element, and may be incorporated into a user's library.     Notes: The problem of harmonic waves in layered and fibre-reinforced composites is solved by a method of finite elements. Piecewise linear approximating functions are used for the displacement and stress fields in a mixed variational formulation, recently proposed by one of the writers in the form of a new quotient. Numerical computations are made for approximate phase-velocities of harmonic waves in layered composites, where asymmetric and symmetric triangular meshes, and square meshes with interior nodes, are used. To illustrate the accuracy and effectiveness of the method, these approximate solutions are compared with those given by means of exponential test functions, and the exact ones by Rytov, etc. Calculations are also performed for harmonic waves in fibre-reinforced composites. Dispersion curves for these waves are obtained and displayed graphically.

    Notes: This paper shows that mixed interpolation is required because of the nature of the pressure terms in the equations. These lead to finite element equations which in many circumstances do not uniquely determine the pressure, if the same interpolations are used for pressure and velocities.Several new combinations of pressure and velocity interpolation are analysed with the aid of a novel diagrammatic technique. In particular we consider some very interesting combinations in which, over nearly all the flow, the pressure and velocity are approximated on elements by biquadratic polynomials which are continuous across element boundaries.The theory of this paper is shown to be in complete agreement with numerical experiment.

    Notes: A finite element method with transmitting boundary is developed for time-harmonic elastodynamics on infinite domains. Only a finite inner domain needs to be discretized into conventional finite elements, while the effect of the exterior domain is simulated by introducing the transmitting boundary stiffness matrix on the degrees-of-freedom of the transmitting boundary nodes. In order to incorporate the Sommerfeld radiation condition, the reciprocity relation, on the transmitting boundary for the radiating elastodynamic state under consideration and the one which is induced by a finite number of point source is used to calculate the transmitting boundary stiffness matrix. The effectiveness of the transmitting boundary technique has been demonstrated by solving the stress concentration problems of a homogeneous isotropic full space with a circular cylindrical cavity and of the one with a spherical cavity, which are subjected to plane incident waves, and the results are compared with the ones of the conventional FEM and the exact data.

    Notes: The transient thermal analysis of large aerospace vehicles such as the space shuttle requires models with thousands of degrees-of-freedom. The design of thermal protection systems for such vehicles requires the repetitive thermal analysis of the vehicle and the calculation of sensitivities - derivatives of the transient response with respect to design parameters. When the number of design parameters is large, the cost of the sensitivity calculations may become prohibitive. The present work is concerned with reducing the computational cost and errors in sensitivity calculations with one of the simplest available methods - the finite difference approach. A technique for calculating simultaneously the nominal and perturbed solutions required for the finite difference derivative is proposed. An expression for the optimum value of the perturbation parameter is obtained which permits reducing the computational error. An insulated cylinder is used as an example which demonstrates that significant improvements in accuracy and efficiency may be obtained.

    Notes: A square tensile sheet with symmetric edge cracks is analysed by the finite element method to demonstrate the accuracy obtained with polynomial elements when the mesh patterns are chosen on the basis of the element's interpolation functions. Numerical results are provided for the accuracy of the total strain energy, the stress intensity factor, and the numerical condition of the global stiffness matrix. The stress intensity factor is determined to within 1.0 per cent accuracy using quadratic (linear strain) triangular elements with only 138 degrees-of-freedom.

    Notes: Two alternative hybrid-stress based three-dimensional elements are presented. These elements are intended for use in the analysis of thick plates for which the through-thickness variation of displacement and stress may be of high order, and in which a single element is to be used through the thickness of the plate. For both elements, the displacement assumption allows for cubic through-thickness variation in the inplane displacements, and quadratic through-thickness variation for the transverse displacement. The elements differ in the stress assumption used. In the first element, the z variations in the stress components are chosen to be consistent with the z variations of the strain distribution calculated from the displacement assumption, and the traction boundary conditions at the upper and lower surfaces are satisfied in the weighted integral sense. In the second element, the z variations in the stress components are chosen to achieve consistency with respect to the equilibrium equations, and the traction-free conditions at the upper and lower surface of the plate are satisfied exactly. The example problem of a semi-infinite, simply-supported thick plate under transverse sinusoidal loading is considered. Results obtained by using each of these elements are compared with available elasticity and approximate solutions, and an assessment of the relative merits and range of applicability of each element is made.

    Notes: A new method of solving the two-dimensional Navier-Stokes equations by using the finite element method is proposed. The flow is represented by the stream function-vorticity formulation and the no-slip boundary conditions are explicitly introduced in the nonlinear equations. This formulation coupled with the Newton-Raphson method enables the study of stationary flows for high Reynolds number, without any convergence problem. A number of flow problems are analysed in order to demonstrate the efficiency of the present formulation.

    Notes: Some applications require the repeated calculation of numerical values of a polyhedral function at points specified by their co-ordinates. The function is usually defined by some simple interpolation over each of the finite elements of a given mesh. A necessary step for the calculation of the value of the function at a given point is to determine to which element the point belongs. Such a determination can be efficiently accomplished by establishing a correspondence between the cells of some suitably defined regular mesh and the finite elements. Then, for each given point, the cell to which the point belongs is determined and the elements associated with that cell are inspected. This method is described and discussed in the text, and compared with two other less efficient methods with the help of numerical examples.

    Notes: The dynamic stress field and its concentrations around holes of arbitrary shape in infinitely extended bodies under plane stress or plane strain conditions are numerically determined. The material may be linear elastic or viscoelastic, while the dynamic load consists of plane compressional waves of harmonic or general transient nature. The method consists of applying the Laplace transform with respect to time to the governing equations of motion and formulating and solving the problem numerically in the transfomed domain by the boundary integral equation method. The stress field can then be obtaind by a numerical inversion of the trasformed solution. The correspondence principle is invoked for the case of viscoelastic material behavious. The method is simplified for the case of harmonic waves where no numerical inversion is involved.

    Notes: The boundary integral method is formulated and applied using cubic spline interpolation along the boundary for both the geometry and the primary variables. The cubic spline interpolation has continuous first and second derivatives between elements, thus allowing the accurate calculation of derivative dependent functions (on the boundary) such as velocity in potential flow. The spline functions also smooth the geometry and can represent curved sections with fewer nodes.The results of numerical experiments indicate that the accuracy of the boundary integral equation method is improved for a given number of elements by using cubic spline interpolation. It is, however, necessary to use numerical quadrature. The quadrature slows calculation and/or degrades the accuracy. The numerical experiments indicate that most problems run faster for a given accuracy using linear interpolation. There seems to be a class of problems, however, which requires higher order interpolation and/or continuous derivatives for which the cubic spline interpolation works much better than linear interpolation.

    Notes: A series of algorithms, to solve the nonlinear algebraic equations resulting from nonlinear finite element models, are proposed. These algorithms involve a selective relaxation of the corrections defined by the modified Newton method. The resulting techniques accelerate the convergence of the modified Newton method. The improved convergence rate is obtained at the expense of a small increase in storage requirements. The results of numerical experiments, on certain nonlinear elasticity problems, are presented to verify the developed techniques.

    Notes: A curved triangular element for plate bending is developed, using the displacement method of formulation. The element has one outwardly curved side and conforms on an arbitrary quadratic boundary.To the user, the element appears as a single curved triangle. Internally, however, the element is subdivided into a straight-sided triangle and a ‘curved segment’. The straight-sided triangle is extensively documented in the literature2-5 and is referred to in the paper as the Cowper triangle. The curved segment is a two-noded element with 12 degrees-of-freedom. It has one straight side and one curved side.Independent expansions are assumed in each domain and explicit shape functions derived. The theoretical solution to a few practical problems is contained in each of the two expansions. In such cases, the curved triangle produces perfect results. Otherwise, the accuracy is of the same order as that obtained in similar problems with straight boundaries.The method is easily extended to elements of higher order.

    Notes: Finite element methods are formulated and investigated for the effectiveness factor problem for heat and mass transfer with chemical reactions in catalyst pellet models. A Galerkin finite element method is compared with a previous C1 collocation method7. A scheme that is conceptually intermediate between these two methods and accordingly has been termed collocation-Galerkin is formulated and numerical experiments considered. Of particular interest here are superconvergence results at the Gauss and Jacobi points, respectively. Numerical studies of superconvergence in the presence of a nonlinear reaction-rate term are presented. An integral formula is devised and used to compute the flux at the pellet surface to optimal accuracy. Numerical experiments are conducted to demonstrate the improvement in computed fluxes.

    Notes: A numerical analysis of the critical conditions of thermal ignition for a two-dimensional cylindrical reactor with flow field occurrence was carried out. The critical conditions of thermal ignition were defined as bifurcation points of the limit type. The dependence of the bifurcation points on aerodynamics was analysed by a sequential variation of the Peclet number. The flow field in the reactor was generated previously by a numerical solution of the Navier-Stokes equations.The nonlinear parabolic equation of heat balance and linear eigenvalue boundary problems occurring in the proposed non-stationary method for calculating the bifurcation points was solved numerically using the finite difference technique.It was shown that the critical points of thermal ignition form a surface bounded by the line of ‘limit of criticality’ in the parameter space. For large values of the Peclet number, the dependence of the critical parameter of ignition on Peclet number becomes linear.

    Notes: The upwinded finite element method was introduced for the solution of conduction-convection problems by considering the model problem, u″-Ku′=0, K〉0, subject to the boundary conditions u(0)=1, u(1)=0. In this paper we consider the solution of this problem by the collocation method using piecewise cubic Hermite polynomials and a uniform grid. We consider the collocation points to be arbitrary parameters which enable us to express the collocation points in terms of the upwinding parameter of the upwinded finite element method. Various choices of the collocation points, including values which lead to complete accuracy in the nodal function values as well as the nodal values of the derivatives are discussed. Finally, we discuss the application of these points to slightly more general problems. Numerical results illustrating the behaviour of these points are given.

    Notes: A numerical procedure developed for solving the two-dimensional elastic contact problems with friction is presented. This is a generalization of a procedure developed by Francavilla and Zienkiewicz to include frictional effects under proportionate loading. The method uses the flexibility matrix obtained by inversion of condensed stiffness matrix formed by eliminating all the nodes except those where contact is likely to take place and those with external forces. Compatibility of displacements for both normal and tangential directions is applied to those nodes which do not slip. However, for the nodes which slip, compatibility of displacements is applied for normal direction only and slip condition is applied in the tangential direction. The technique has been applied to several problems and very good results have been obtained. The number of iterations needed are very small.

Computations for a single element are explicitly carried out using a symbolic algebraic manipulation program and in addition to the desired square root singularity, additional singularities are revealed. Cubic transitional elements were used for a double-edge crack problem and only marginal improvement is found for a well-laid out grid.

    Notes: An easy to use stability test is presented for finite elements which are under- or selectively-integrated. This test is reminiscent of the patch test in that it must be applied only to one prototype element, and it is a sufficient test for the nonsingularity of the global coefficient matrix.     Notes: Global and element residuals are introduced to determine a posteriori, computable, error bounds for finite element computations on a given mesh. The element residuals provide a criterion for determining where a finite element mesh requires refinement. This indicator is implemented in an algorithm in a finite element research program. There it is utilized to automatically refine the mesh for sample two-point problems exhibiting boundary layer and interior layer solutions. Results for both linear and nonlinear problems are presented. An important aspect of this investigation concerns the use of adaptive refinement in conjunction with iterative methods for system solution. As the mesh is being enriched through the refinement process, the solution on a given mesh provides an accurate starting iterate for the next mesh, and so on. A wide range of iterative methods are examined in a feasibility study and strategies for interweaving refinement and iteration are compared.

    Notes: For plate bending analysis, both discrete and non-discrete mixed methods are presented by using spline functions in the Hellinger-Reissner variational principle. Piecewise spline interpolation with local supports and overall spline interpolation with boundary knots are applied to each of two mixed functionals. The prescribed boundary conditions can be considered by multiple boundary knots. The convergence behaviour of the mixed methods presented is studied. Numerical examples show that a high degree of accuracy can be obtained due to continuous properties in the high derivatives of the employed shape functions. The proposed mixed methods are found to be very competitive with other numerical methods for plate bending problems.

    Notes: Candida Krusei has a optimum growth temperature of 37°C on SASOL ethanol-isopropanol mixture. The organism was unable to grow on isopropanol, but oxidized it partially to acetone in the presence and absence of ethanol. Growth at 40°C in the alcohol mixture was slightly faster than at 30°C over an ethanol concentration range of 0.43 to 3.6% (v/v), although at both temperatures the growth rate declined continuously with increasing concentration. At an ethanol concentration greater than 3.6% (v/v), the mixture was much more inhibitory to growth at 40 and 30°C. The inhibitory effect was due to the ethanol rather than the isopropanol. Metabolites such as acetate, acetaldehyde, and ethyl acetate accumulated in the medium, but the degree of accumulation depended upon the temperature and alcohol mixture concentration. At 40°C, acetaldehyde and acetate accumulated to a greater extent than 30°C on a 4.0% (v/v) synthetic alcohol mixture and this may also cause the greater inhibition at this temperature. The alcohol mixture is unsuitable for single cell protein (SCP) production in batch culture because of the low cell densities observed at all alcohol concentrations.

    Notes: It has been shown both theoretically and experimentally that interphase nitrogen transport may have a significant influence on the rate of interphase oxygen transport, and thereby also on the value of the volumetric mass transfer coefficient of oxygen, kla, determined in mechanically agitated bubble fermentors using the variants of dynamic method presented in the literature. The experiments were carried out in 1M KCI solution at five stirrer frequencies and two gas inlet levels. The gas interchanges were performed either without interrupting the aeration and agitation of the charge (A) or with the aeration and agitation of the charge turned on at the same time (B). The applied variants of the interchange were N2→ O2→, O2→ N2, N2→ air, air→ N2, O→ O2, and O→ air. In the two last variants the oxygen dissolved in the charge was removed by reacting with sulfite ions. The kla values calculated by allowing for the nitrogen transport for procedure A were approximately equal to the values obtained by disregarding the nitrogen transport, whereas those for procedure B were higher (up to 40%), than the values obtained disregarding the nitrogen transport.

    Notes: Raw cassava root starch was transformed into ethanol in a one-step process of fermentation, in which are combined the conventional processes of liquefaction, saccharification, and fermentation to alcohol. Aspergillus awamori NRRL 3112 and Aspergillus niger were cultivated on wheat bran and used as Koji enzymes. Commercial A. niger amyloglucosidase was also used in this experiment. A raw cassava root homogenate-enzymes-yeast mixture fermented optimally at pH 3.5 and 30°C, for five days and produced ethanol. Alcohol yields from raw cassava roots were between 82.3 and 99.6%. Fungal Koji enzymes effectively decreased the viscosity of cassava root fermentation mashes during incubation. Commercial A. niger amyloglucosidase decreased the viscosity slightly. Reduction of viscosity of fermentation mashes was 40, 84, and 93% by commercial amyloglucosidase, A. awamori, and A. niger enzymes, respectively. The reduction of viscosity of fermentation mashes is probably due to the hydrolysis of pentosans by Koji enzymes.

    Notes: The filamentous fungus, Rhizopus nigricans, was immobilized in polyacrylamide, alginate, and agar gels and its ability to 11α-hydroxylate progesterone was examined. No activity was detected using polyacrylamide gel but both agar and alginate gels have proved capable of hydroxylation. Agar gels displayed faster rates and higher yields. It was possible to induce hydroxylase synthesis within agar and alginate gels, and microscopical examination provided evidence for hyphal growth within these gels. The concept of increased biomass was used to explain the observed increase in the rates of hydroxylase activity of the immobilized cells. Conversely, hyphal overcrowding was postulated for the rapid inactivation observed under some operating conditions.

    Notes: A fermentation system with a plug scheme unit has been developed, offering a variety of solutions to measurement, control, and operational problems. By means of the program unit, e.g., automatic pH control assigned to the dynamic of batch cultures and the feed of different ingredients controlled by a time program or a given variable have been solved. The continuous culture volume was controlled by a level controlled by a level controller equipped with a photosensor. A method was developed for variable control that provide information on the activity of the culture, and allows direct measurement of the different rate values, e.g., generation time or specific product formation rate. Applicability of the direct measurement of generation time is presented in the qualification of molasses and in a static off-line optimization process.

    Notes: The effect of immobilization with various glass-forming monomers on the stability of PS II activity of spinach chloroplast was investigated. PS II activity (O2 evolution due to the Hill reaction) was reduced very slightly by the addition of monomers including polyethyleneglycol (PEG). Immobilization of chloroplast was done with hydrophobic monomer as well as hydrophilic monomer and activity of immobilized chloroplast increased with decreasing monomer concentration as far as the polymerization was possible. The activity of immobilized chloroplast was very high and it decayed far more gradually with the storage time in comparison with the decay of unimmobilized chloroplast and was retained more than 30 days. The optimum monomer concentration for immobilization was about 10%. Thermostability of chloroplast also increased greatly by immobilization with these monomers, especially hydrophilic monomers.

    Notes: A range of cellulosic raw materials in the form of agricultural crop residue was analyzed for chemical composition and assessed for potential yields of sugars through chemical pretreatment and enzymatic hydrolysis of these materials. Corn stover was used as a representative raw material for a preliminary process design and economic assessment of the production of sugars and ethanol. With the process as presently developed, 24 gal ethanol can be obtained per ton of corn stover at a processing cost of about $1.80/gal exclusive of by-product credits. The analysis shows the cost of ethanol to be highly dependent upon: (1) the cost of the biomass, (2) the extent of conversion to glucose, (3) enzyme recovery and production cost, and (4) potential utilization of xylose. Significant cost reduction appears possible through further research in these directions.

    Notes: The cyanobacterium Anabaena cylindrica B629 was suspended in small glass beds and incubated in a gas-tight glass vessel outdoors under a gas atmosphere comprising carbon monoxide (0.2%), acetylene (5%), oxygen (6.5%), and nitrogen. The solution phase initially contained sodium bicarbonate (10mM) at pH 7. Under these conditions the organism continuously produced hydrogen gas for over three weeks. The temperature of the culture was maintained below 30°C and minimum night temperatures were recorded. The vessel was covered by a shadecloth, which reduced the natural illumination by approximately 70%. The system is an alternative to those requiring the strict absence of oxygen and little nitrogen, and requires virtually no attention during the incubation period.

    Notes: An enzyme which catalyzes the synthesis of cephalexin fromD-α phenylglycinemethylester (PGM) and 7-amino-3-desacetoxy-cephalosporanic acid (7-ADCA) was prepared from Xanthomonas citri (IFO 3835) and partially purified 30-fold by ammonium sulfate fractionation, DEAE-cellulose, and Sepharose-4B column chromatography. The Km values for 7-ADCA, PGM, and cephalexin were determined as 11.1, 2.1, and 1.61 mM, respectively. The enzymatic cephalexin synthesis follows the reversible bi-uni reaction kinetics. The equilibrium constant is influenced by the initial mole ratios of 7-ADCA and PGM. The cephalexin hydrolysis is catalyzed by the same cephalexin synthesizing enzyme, but methanol does not participate in the hydrolytic reaction. The amount of enzyme in the reaction mixture affects the initial rate but does not influence the equilibrium product concentration. This cephalexin-synthesizing enzyme was immobilized onto several adsorbents. Among these, Kaolin and bentonite showed a higher retention of enzyme activity and stability for reuse. The immobilized-enzyme reaction kinetics were investigated and compared with those of the soluble enzyme. A rate expression for the enzymatic synthesis of cephalexin was derived. The results of computer simulation showed good agreement with the experimental results.

    Notes: A simple method is presented that establishes intrinsic rate parameters when slow pore diffusion of substrate limits immobilized enzyme reactions that obey Michaelis-Menten kinetics. The Aris-Bischoff modulus is employed. Data at high substrate concentrations, where the enzyme would be saturated in the absence of diffusion limitation, and at low substrate concentrations, where effectiveness factors are inversely proportional to reaction modulus, are used to determine maximum rate and Michaelis constant, respectively. Because Michaelis-Menten and Langmuir-Hinshelwood kinetics are formally identical, this method may be used to estimate intrinsic rate parameters of many heterogeneous catalysts. The technique is demonstrated using experimental data from the hydrolysis of maize dextrin with diffusion-limited immobilized glucoamylase. This system yields a Michaelis constant of 0.14%, compared to 0.11% for soluble glucoamylase and 0.24% for immobilized glucoamylase free of diffusional effects.

    Notes: This paper describes the scale-up from 0.1 to 100 liter of the unit process based on 3-mm-diameter glass spheres for the growth of BHK monolayer cells. The production of four strains of FMD virus at the 0.1-, 10-, and 100-liter scales was examined. Cell growth was estimated from measurements of the concentration of glucose in the growth medium, while the release of virus was inferred from measuring the concentration of LDH in the culture supernatant fluid. The yields of virus at 0.1-, 1-, and 10-liter scales were similar but that from the 100-liter version was somewhat lower. The reason for this lower yield and the method used to overcome it over outlined.

    Notes: A culture of R. rubrum cells apparently contaminated with K. pneumoniae were immobilized by entrapment in agar. This system was used as model for hydrogen production by photometabolic means. Observed results indicated that the contaminant exerted a major influence on the observed results. This preparation, when immobilized and used in a specifically designed reactor with glucose substrate, showed operational half-lives of approximately 1000 hr. The feasibility of using this “mixed” culture for producing hydrogen from acid hydrolyzed cellulose and wood sawdust was also examined.

    Notes: Nickel-NiO-BSA-chymotrypsin has been used to hydrolyze casein in both batch and continuous stirred tank reactors. The kinetics of hydrolysis in both types of reactor were considered. Some operational problems encountered using fine powder catalysts in batch stirred tank reactors are discussed. High gradient magnetic separation was found to be a powerful catalysts retention method for magnetic support particles, particularly when using a ferromagnetic collection matrix. Nickel particles of diameter 3-7 μm were easily separated from water at a processing velocity of 39 mm/sec. Nickel powder and precipitated Fe3O4 were also separated satisfactorily using a rotating drum magnetic separator.

    Notes: There is now growing evidence that the production of many secondary metabolic by microorganisms is subjected to carbon-catabolite regulation. Even though the exact mode of this regulation is not yet clear, an engineering analysis of the production process is still possible based upon a suitable hypothesis. By way of simulation of penicillin fermentation data obtained from the literature, a mechanistic model involving a substrate inhibition kinetics of product formation has been verified in this paper. Such a model has been found successful not only in predicting simple sugar-feeding strategy, but also a complicated computer guided strategy based upon controlling biomass growth rates in the tropo and idiophases. Using this model, for strategies for sugar feeding into penicillin fermentation have been investigated. These results show that similar penicillin productivities can be obtained using any of these strategies provided fermentations are carried out under optimal conditions corresponding to the strategy chosen. Effect of maximum oxygen transfer capacity of the fermentor under the conditions of fungal growth has been incorporated using an upper limit of biomass concentration on achievement of which the fermentations must be stopped due to serious oxygen limitations. Results of model simulations with such limits throw light upon the way in which different fermentors may behave with respect to product formation.

    Notes: In order to reduce the severe flux losses encountered during ultrafiltration of protein solutions, proteases were immobilized on Ultrafiltration membranes to hydrolyze the deposited solute molecules. Over a standard 22 hr run 25 to 78% improvement in cumulative permeate yield was obtained when processing 0.5% albumin or hemoglobin. It was also demonstrated that the flux enhancements were due to the biochemical action of the absorbed protease and not to its physical effect as a prefilter coat. with the aid of a model retardation of gel formation mechanism was demonstrated. Economics of the system were shown to be favorable, improving the rate of return on capital investment up to 50% by reduction of the total membrane area of the plant.

    Notes: This paper describes the implementation of multilevel techniques using a microprocessor to control multistage continuous culture systems. A system which produces gramicidin S is taken as an example. The single level technique using the conjugate gradient method is applied to solve the two-stage and the three-stage continuous culture and is compared with the multilevel one. The results show that the application of multilevel techniques is more advantageous and suitable for this system than any other method which has been utilized so far. The advantages of using a microprocessor will be stated.

    Notes: The growth of E. coli W in a bench scale fermentor to high cell concentration is described. The method involves growth-linked introduction of ammonia to the culture, sparging the culture with oxygen, and maintenance of aerobic conditions during the final growth phase by gradually and automatically decreasing the concentration of the carbon source, sucrose, in the culture. Thus, the oxygen demand is kept within the limits of the supply capacity, and a linear growth rate during the final phase of growth is obtained. A concentration of 42 g dry cell per liter was obtained. The yield constants for nitrogen and phosphorous were determined and were compared with those obtained using the temperature variation method.

    Notes: Rates of oxygen absorption into glucose solutions were measured using an immobilized-enzyme reactor, in which magnetite-containing beads of immobilized glucose oxidase were moved by a revolving magnetic field to reduce the mass transfer resistances at the gas-liquid interface and around the bead. Data were also obtained for oxygen absorption into glucose solutions containing soluble or immobilized glucose oxidase (without magnetite), as well as for physical absorption of oxygen. The rates of physical absorption for the runs with the magnetite-containing beads increased because of mechanical stirring caused by spinning of the beads at the gas-liquid interface. In this case the experimental enhancement factors were found to be larger than those predicted on the basis of the film theory for gas absorption with a pseudo-first order reaction.
