    Notes: The new sequential centrifugal layer chromatography extends the possibilities for preparative separation of furocoumarin isomers. In the present work, isobergapten, pimpinellin, bergapten, isopimpinellin, and sphondin were isolated from a prepurified extract of Heracleum sphondylium roots using two different techniques, namely, centrifugal layer chromatography (CLC) and sequential centrifugal layer chromatography (SCLC). The former technique was carried out with four plates, whereas the latter method required only one plate, using the same mobile phase in each case. A further advantage of the SCLC method was that all completely separated compounds could be quickly and efficiently eluted in small volumes of mehtanol.

    Notes: A modification of a packed GC injector to a capillary injector is presented using a simple device which is connected to the GC column by an adsorption-free connector. It permits injections of large sample volumes up to 500 μl hexane solutions on normal 0.3 mm i.d. capillary columns. No special requirements for the stationary phase are necessary. Involatile residues remain inside the device which can be exchanged easily. High performance of separation and quantification is achieved.

    Notes: The capability of the helium ionization detector (HID) to operate in connection with capillary columns for trace gas analyses has been evaluated. Two different capillary columns were considered: a PLOT fused silica column with molecular sleve and a thick film WCOT glass column with PS-255. The determination of trace impurities in gases can be achieved with evident advantages over classical adsorption columns, even using a split injection system. Direct on-column injections also have been investigated with promising results.

    Notes: In this paper we report a way to evaluate the removal of volatile halogenated hydrocarbons formed after break-point chlorination in the Arno river raw water at the Mantignano Utility, Florence, Italy, following dechlorination by hydrogen peroxide. The removal evaluation is performed by GC-ECD on fused silica capillary columns with split-splitless injection on n-pentane extracts and by other analytical techniques such as UV254 absorption, TOC and TOX. Both laboratory and pilot plant data on Arno river raw water are reported and discussed.

    Notes: It is of fundamental importance to know the true composition of the essential oil produced by the secretory structures of a plant, both from a biogenetic and a chemotaxonomic point of view. This paper considers a method which combines direct sampling with on-column CGC and studies the oils directly from the secretory structures: some examples are given (Humulus lupulus L., Tagetes erecta L., and Tagetes patula L.). For each plant, the composition of the distilled oil is compared to that obtained by direct sampling, and a photograph of the secretory structures taken by electroscanning microscopy is given.

    Notes: The Community Bureau of Reference has organized a collaborative interlaboratory project to improve the analytical protocol for some specific chlorobiphenyls (CBs) within the European Community (EC). A series of test procedures were prescribed to optimize the gas chromatographic conditions for splitless and on-column injection, which substantially improved the quality of data. Important parameters included the initial column and injector temperatures and the choice of the polarity of the stationary phase. In a study with cleaned eel-fat extracts, coefficients of variation for reproducibility CV(R) ranging from 11 to 24% at the 0.04-0.3 mg/kg level per CB congener were obtained. No significant difference could be found between splitless- and on-column injection.

    Notes: The practical applications of open tubes in liquid chromatography as separation columns and as reactors for post column derivatization are discussed. With present technology, the potential efficiency of open tubular columns cannot be fully exploited. As reactors in post column derivatization, these tubes offer many advantages if knitted or stitched to enhance radial mass transfer. The application of open tubes with diameters between 0.12 mm and 0.6 mm as reactors in the selective and sensitive detection of amino acids is demonstrated.

    Notes: The efficiency of FSOT columns coated with thick films of polyphenylmethylsiloxane was theoretically and experimentally compared to thin apolar and medium polar coatings and to thick apolar coatings. Experimental hū and TZū plots clearly demonstrate the enormous loss in chromatographic efficiency of thick films of medium polar siloxane stationary phases. The low diffusion coefficient in the liquid phaese is responsible for this behavior. The influence of the temperature and the nature of the solute on the efficiency was also studied.

    Notes: The Saccharomyces strains uvarum, montuliensis, bayanus, capensis and florentinus were grown in liquid media prepared from a 1% aqueous glucose solution to which 0.05% of ammonium sulfate or urea were added as the only nitrogen supply. The amino acids exuded during incubation were isolated by cation exchange and derivatized as the corresponding N,O-heptafluorobutyryl isopropyl esters. Separation was achieved on a highly deactivated glass capillary column coated with OV-101. A characteristic amino acid profile was obtained for each of the species studied, allowing the construction of a chemotaxonomic tree, on the basis of present or absent acids.

    Notes: Sampling techniques for practical quantitative capillary GC have to meet certain principal requirements. Both the absolute and the relative peak areas (e.g. column loads) must be reproducible with high precision and at high accuracy; discrimination of certain constituents according to their volatility should not take place on sampling. On the basis of systematic studies, the three most reliable sampling techniques used for GC analyses with the aim of achieving precise and accurate quantitative data proved to be the following: On-column, injection, splitless PTV injection, and an optimized version of split sampling called “cooled needle split” injection. The on-column technique can be optimized by using precolumns with wider internal diameters and without stationary phase coatings to overcome the problems of large liquid sampling volumes and for automation. The PTV technique should only be used in the splitless mode because discrimination cannot be suppressed completely with the split mode. All three of the techniques can be operated automatically, either to avoid “human interference”, i.e. to improve precision or for unattended operation to save man-power.

    Notes: The elution profiles observed with overloaded capillary columns are well described by a four-parameter theoretical model derived from the basic mass balance equations which relies on an expansion of the partition isotherm to the second term and takes the sorption effect into account. The reliability of the model is demonstrated by the fact that the peak leaning coefficient obtained from a least squares fit of the model to the experimental peak is in good agreement with that obtained from the variation of peak height with retention time at peak maximum. The coefficeint of apparent axial dispersion is independent of sample size and can be used to characterize the plate height at zero concentration of an overloaded capillary column.

    Notes: Direct conversion of isothermal to temperature programmed indices is not possible. In this work it is shown that linear temperature programmed retention indices can only be calculated from isothermal retention data if the temperature dependence of both the distribution coefficients and the column dead time are taken into account.Procedures are described which allow calculation of retention temperatures and from these, accurate programmed retention indices. Within certain limits the initial oven temperature and programming rate can be chosen freely. The prerequisite for this calculation is the availability of reliable isothermal retention data (retention times, retention factors, relative retention times, or retention indices) at two different temperatures for one column.The use of compiled isothermal retention indices at two different temperatures for the calculation of retention temperatures and thus temperature programmed indices is demonstrated. For the column for which programmed retention indices have to be determined, the isothermal retention times of the n-alkanes and the column dead time as a function of temperature have to be known in addition to the compiled data for a given stationary phase.Once the programmed retention indices have been calculated for a given column the concept allows the calculation of temperature programmed indices for columns with different specifications. The characteristics which can be varied are: column length, column inner diameter, phase-ratio, initial oven temperature, and programming rate.

    Notes: Work-up procedures and HPLC separation systems for determination of taurocholic, glycocholic, chenodeoxycholic, and cholic acids and lysolecithin in artificial and natural gastric juice are described. These compounds are used for testing the binding capacity of antacida to the individual analytes. Work-up is simple, no extraction or filtration being required. The optimized combinations of stationary and mobile phases allow selective and sensitive determination of the respective bile acids in gastric juice. For optimization, the capacity factors of two related bile acids were evaluated for numerous commercial stationary phases. The mechanism of retention is different for free and conjugated bile acids. Special aspects of routine analysis are discussed.

    Notes: A cold on-column injection system for capillary gas chromatography (GC) applications was constructed. It was based upon a conventional split/splitless capillary GC inlet, which in turn was a modification of a conventional packed GC column inlet. The heart of the laboratory constructed cold on-column inlet design was a disposable pyrex micro-sampling pipet, which functioned as a needle guide for sample injection. The sample was injected through a traditional GC septum. Construction of the injection system is described and applications are illustrated by separations of a variety of complex mixtures.

    Notes: The antihypertensive agent urapidil (Ebrantil Byk-Gulden, Konstanz) can be reliably quantitated with three metabolites in human serum using high performance liquid chromatography. Serum is alkalinized and extracted with ethyl acetate. The organic phase is back-extracted with diluted acid. An aliquot is sampled automatically and chromatographed in an optimized combination of mobile and stationary phase. UV-detection at 273 nm allows a quantitation limit of 5 ng/ml for all analytes. Precise handling of exact volumes facilitates external calibration. The coefficient of variation for spiked samples is less than 5% within and less than 7% between studies. Application of the method to experimental and clinical pharmacokinetic studies of urapidil is illustrated.

    Notes: The construction and evaluation of an interface for two-dimensional gas chromatography is described. The interface consists of commercially available components and is attachable to available gas chromatographs without any major modifications. The interface has been constructed so as to permit “heartcutting” as well as solute band concentration. Trapping is performed in a simple cold trap of fused silica. Flow switching is accomplished by Deans switching. Factors of the over-all chromatographic performance are examined.

    Notes: A digital chromatogram simulator has been used to evaluate the performance of data handling systems. The simulator can synthesize chromatograms with any desired shape from a library of peaks based on real peak shapes. Once generated, the chromatograms can be reproduced exactly any number of times.The algorithms used in data handling systems have a significant effect on the results they produce. An object of this work was to examine how well the true size of a peak was reported when the peak was detected overlapping with other peaks. A series of experiments was therefore performed in which peaks were overlapped by different amounts and in different situations. The reported results from the data systems were compared with the results which would have been reported had the peaks been fully resolved. A significant difference was found between the overlap results and the fully resolved results, which did not have a linear relationship with the parameters tested.The chromatogram simulator was found to be a powerful tool in assessing the performance of the data systems. It offers a valuable means of evaluating the quality of the results produced by these systems.

    Notes: Inorganic mercury and some organo-mercury species have been separated as dithizonates by HPTLC and determined in situ at the subnanogram level by densitometric automatic scanning of the plate. The extraction efficiency of the dithizonates was tested at pH 1, 7, and 10. The Rf values for HgD, CH3HgD, C2H5HgD, and C6H5HgD are 0.15, 0.32, 0.35, and 0.28 respectively. The recovery of the tested mercury species in simulated and real samples has been investigated. Conditioning of the plates with NH3 prior to elution eliminates interference by many other metal ions. Some examples of real sample analysis (tap water, sea water, human urine) are reported.

    Notes: The coating speed upon static coating of glass capillary columans was evaluated in terms of inner diameter and length of the column, viscosity and pressure of solvent vapor, etc. From the equation obtained it can be shown that a smaller diameter of a microbore column restricts solvent vapor transfer to the orifice of the column drastically. To compensate for this restriction, a higher pressure at the meniscus is needed. As an alternative to using a higher coating temperature, application of more volatile solvents such as n-butane and isobutane is proposed. Several glass capillary columns (130 μm i.d.) were coated with SE-54 dissolved inpentane-acetone mixed with n-butane or isobutane and the column performances were evaluated. Selection of these solvents permitted free release static coating of ca. 100 μm columns at lower or even ambient temperatures and they were equally suitable as commonly applied solvents (e.g. pentane) to coat highly efficient columns.

    Notes: Reversed-phase liquid chromatography (LC) on C18 stationary phases provides excellent selectivity for the separation of polycyclic aromatic hydrocarbons (PAH). Recent studies have shown that several factors affect selectivity for the LC separation of PAH including phase type (monomeric or polymeric), pore diameter and surface area of the silica substrate, and surface density of the C18 ligands. In this paper the separation of eleven PAH isomers of molecular weight 278 is used to further illustrate the effect of stationary phase characteristics and shape of the solute (length-to-breadth ratio, L/B) on retention and selectivity. Only polymeric C18 phases with a high C18 surface coverage provided separation of all eleven isomers and the elution order of these isomers generally followed increasing L/B values. The effect of solute nonplanarity on reversed-phase LC retention was investigated on both monomeric and polymeric phases using a series of planar and nonplanar PAH pairs. For each solute pair, the nonplanar solute eluted earlier than the planar solute, the largest selectivity factors being observed on the C18 phase with the highest percent carbon load. Based on these studies, a model is proposed to describe the retention of PAH on polymeric C18 phases.

    Notes: A thermostated HPTLC system was assembled to test the influence of gradient chamber saturation upon resolution of polar lipids chromatographed on boric acid-impregnated silica gel plates. Inexpensive mixtures of nonacidic (14 component peaks) and acidic (8 component peaks) lipids were prepared from soya lecithin by a simplified procedure involving DEAE-Sephadex liquid column chromatography to monitor performance of the system using a readily available reference material. The system requires ca. 5 min chamber presaturation after which the polar lipids are chromatographed in less than 30 min. Retention times (tR) as well as peak area % values of all component peaks and measured by scanning photodensitometry to demonstrate the influence of different modes of chamber saturation upon the resolution of polar lopids.

    Notes: A high sensitivity, multichannel fluorescence detector with small volume has been developed for capillary column liquid chromatography. Using an intensified linear photodiode array to monitor fluorescence emission, several important mixtures exhibiting native fluorescence have been examined following high efficiency separation on a capillary column. By correlating mass spectral, fluorescence spectral, and retention time data, information of potential utility in the structural elucidation of aromatic molecules contained in complex mixtures can be obtained. Examples include the separation and spectral examination of the polyaromatic compounds in samples of both biological and environmental interest.

    Notes: A study was carried out on the effects of acid leaching of capillary column wall materials. Leaching experiments with glass and fused silica were performed under static and dynamic conditions; moreover, the influence of the temperature was investigated. The amount of water released during different temperature programs was measured with a microwave plasma detector. Different leaching conditions and column wall materials cause considerable variations in water production. These results probably explain the disappointing deactivation efficiency obtained for fused silica and dynamically leached soft glass if HMDS is used. Evidence was found for an increase in surface area due to acid leaching.

    Notes: Organic compounds adsorbed on fly ash from an incinerator for radioactive waste were investigated. Following Soxhlet extraction and concentration, analysis was performed by GC and GC/MS using high temperature silylated glass capillary columns coated with OV-1 and SE-54. Compound identification was based on retention data, relative detector responses, and mass spectra. Among other polycyclic aromatic compounds, series of gradually chlorinated PAHs containing four to seven aromatic rings were detected. For unequivocal identification, appropriate reference compounds were synthesized by chlorination of the corresponding parent PAHs.

    Notes: Supercritical fluid chromatography (SFC) sometimes exhibits GC-like behavior and sometimes LC-like behavior, depending on conditions. However, it is not always clear whether one of these types of behavior, or a combination, operates for a particular set of conditions for every solute in a mixture. For example, some components may be partitioned mostly by their vapor pressures, while others, in the same mixture, are partitioned predominantly by solvent-like properties of the mobile phase.Plots of retetion (as log of the capacity factor) vs. reciprocal temperature at constant pressure reveal a clear change in the character of the separation of well-behaved solutes. A thermodynamic explanation of the observed behavior is given, based on the assumption that partitioning is controlled by the heats of solution of solute in the mobile and stationary phases. A model of SFC retention as it deviates from pure-GC behavior on the same column is presented.

    Notes: We describe the polarity of selectivity of a GC separation system in terms of Retention Index data. In a series-coupled capillary system having stationary phases of differing polarity even slight (independent!) carrier gas flow changes in one part of the capillary series result in a dramatic change of selectivity. “Dramatic” is a relative term! Using a simple electronically controlled flow changing device we found retention index changes of several hundred units for polar test compounds such as phenols on a SE30/Carbowax tandem. This means: The classical theoretical model for understanding retention (and selectivity) in chromatography must be corrected. We propose a very simple approach involving addition of the expression RESIDENCE TIME to the chromatographic vocabulary. Instead of using flow resistors, one can just add a pressure regulator to the coupling point. A powerful analytical concept is now in easy reach.

    Notes: Two-dimensional chromatography of gasoline by on-line coupled HPLC-HRGC, as described in this paper, allows separate GC analysis of paraffins and aromatics. The GC system contains a retention gap of only 10 m length for introducing HPLC fractions of 100 μl volume. This becomes possible through evaporation of part of the solvent during introduction of the HPLC eluent. This “partially concurrent solvent evaporation” technique allows transfer of large volumes of HPLC eluent into relatively short retention gaps, maintaining the full efficiency of the solvent effects in reconcentrating the bands of the early eluted solutes.

    Notes: A new approach is described for micro-HPLC for generating nearly linear gradients in the 40 to 550 μl volume range. These “linear” gradients have a straight line middle portion with a gentle curved onset of the gradient andd rapid attainment of the final 100% composition. Such “breakthrough-gradients” are produced by the interface between weak and strong eluent when a small (3 × 0.46 cm) packed “gradient-generator” column is switched abruptly from weak to strong eluent. The model system described here, without an analytical column, shows gradients from weak eluent (water) to strong eluent (modele with water plus acetone) using a 12-port high pressure valve to 1) vent the “flush” eluent, and simultaneously 2) begin the gradient and 3) make the sample injection.The volume of the gradient can be changed in two ways. If the breakthrough-generator is packed with very large (300 to 1600 μm nonporous particles, the gradient volume is nearly independent of the flush flow rate. The flush flow is used to quickly move the breakthrough-interface to the head of the micro-HPLC column. However, the gradient volume can be increased by using larger solid particles in the gradient-generator column because of increased eddy diffusion. If the gradient-generator is packed with porous particles (75-150 μm), the gradient volume can be changed over a broad range by simply changing the flush flow rate. This simple control over the gradient volume is due to the increased mass transfer of weak eluent out of the porous particles into the strong eluent at higher flush flows.The breakthrough-interface gradient-generator column approach provides the following advantages for micro-HPLC: - Gradient volumes can be readily varied over the micro-HPLC range 40-550 μl.- Gradient are linear with smooth and rapid onset of intial and final concentrations.- Gradient are produced inexpensively with a single high-pressure precision pump.- Gradient can be automated with a timer and single 12-port valve that provides both column regeneration and sample injection.

    Notes: Androgen 5α-reductase from the foreskin of 48 boys in prepubertal age was analyzed by evaluation of the apparent Km-and Vmax- values after tissue incubation. Reaction rates were calculated after determination of specific radioactivity of distinct metabolites, by comparison to the radioactivity of the precursor. Reaction products were separated by radio gas chromatography on capillary columns.     Notes: Chromatographic separation of crude oil components has been performed on a 50 m Chrompack Sil 5 fused silica capillary column, temperature programmed from 10 to 300°C. Approximately 70 peaks eluting before n-nonane (n-C9) and all normal paraffins to n-C19 were identified by a retention time table. Peak areas were converted to weight % using 1-heptene as internal standard and relative response factors. Components were divided into boiling range groups from n-C6 to n-C19. Paraffinic, naphthenic, and aromatic distributions were obtained for the boiling point fractions up to n-C9.Group average densities and molecular weights were calculated from pure component properties up to n-C9. Knowing the density and molecular weight as functions of boiling range group to n-C9, these functions can be extrapolated to n-C19, based on data and correlations from more than 20 distillations of paraffinic North Sea oils adn condensates. The densities and molecular weights of the C10+ and the C20+ residues were calculated to satisfy a balance of weight, volume, and number of moles for the whole sample.Measurements on the residues were used to cross-evaluate the GC method against distillation. The difference in weight was estimated to be less than ± 0.5% in absolute terms for the C10+ residue. The corresponding figure for C20+ was ± 2%. Relatively, calculated densities are within ± 1%, molecular weights within ± 2% compared to direct measurements. Calculations were performed by a BASIC program in a GC interfaced computer.

    Notes: A new phenyl-cyanopropyl polymethylsiloxane stationary phase, DB-1301, which can be crosslinked and bonded to the fused silica surface, has been synthesized. The phase composition was optimized for the separation of 18 volatile priority pollutants, selected from the so-called “purgables” list because of problems associated with their resolution. The composition of the stationary phase was based on theoretical considerations of binary phase mixtures and window diagram prediction. Columns coated with this stationary phase are compatible with atom selective detectors, such as the electron capture detector. A mixture of 18 volatile priority pollutants and a mixture of chlorinated pesticides were used as test probes on a prototype 30 m × 0.32 mm fused silica column, containing a 1.0 μm film of DB-1301.

    Notes: High performance thin-layer chromatography (HPTLC) was used to determine 12 metabolites of the tryptophan-NAD pathway in rat urine. The method of separation described in the previous report was improved. The metabolites were separated on the cellulose plate by using five multiple developments with four solvent systems. The plate was scanned by the TLC scanner at 254 nm and the amounts of the metabolites were calculated by comparing peak areas of the scanning curves obtained for the urine sample and for the authentic standards. This procedure can be used as a semiquantitative determination method for the metabolites in biological fluids.

    Notes: Co-bonding ligands on the same support to produce “blended” HPLC stationary phases is most commonly carried out in the form of capped reversed-phase materials. Non-additive retention properties were observed over a range of trimethylsilane/dimethyloctylsilane ratios.     Notes: A homologous series of 2,4-dichlorobenzyl alkyl ethers (DCBEs) have been synthesized and purified for use as retention index calibrants and internal standards. They are stable, sensitive to ECD and FID and ideal for GC-MS, with base peaks at m/z 159/161. The retention index calibration against the n-alkanes for a linear temperature program (LTP) series is given and recommendations made for the most suitable members of the DCBE series for use as internal standards in organochlorine (OC) residue analysis.

Biological samples were collected from healthy volunteers after a single topical administration of the drug in gel form.     Notes: The aim of the present study is to develop a method for following the changes in the chemical composition of volatile substances released by plants. Dynamic headspace enrichment, followed by on-line thermal desorption and cryotrapping was coupled with capillary gas chromatography. The technique was optimized by adjusting the sampling and desorption parameters and by designing a special condensing device to avoid the transfer of excessive amounts of water into the analytical column. Details are given for the characterization of volatiles compounds released by mosses.

    Notes: A gas chromatographic/mass spectrometric assay for quantifying phenprocoumon and its 4′-, 6-, 7- and 8-hydroxy metabolites in microsomal preparations is described. This assay which uses deuterium-labeled analogs of the phenprocoumon metabolites as internal standards has a lower limit of quantitation of 20 ng ml-1. Diazomethane is used to derivatize both metabolites and parent compound yielding along with the expected 4-methoxy derivative a minor amount of the 2-methoxychromone. Resolution of the methylated metabolites is accomplished by capillary gas chromatography.
