    Notes: Complete separation of the TMS derivatives of histamine, 1-methyl histamine and 3-methyl histamine was achieved in two minutes in a 7 m × 0.25 mm column coated with SP 2250. Using NPD, histamine was detectable at levels as low as 20 pg. This offers a rapid, sensitive procedure with potential for clinical diagnosis and food analysis.

    Notes: This paper describes the results of a study on the deactivation of the surface of glass capillary columns by high temperature silylation (HTS). The different steps, leaching, washing, dehydration and HTS were optimized. A practical procedure yielding a high percentage of very good columns is given. The influence of leaching and HTS on the temperature stability and coating efficiency of capillary columns coated with OV-1 was studied. The inertness of the glass surface after HTS is demonstrated by several examples.

    Notes: This article describes the development of the application of capillary column GLC methods to steroids, prostaglandins and especially drugs and their metabolites in samples of biological origin.     Notes: Two regular GC instruments were modified with injection port liners, split-mode injection systems, pressure regulators, effluent traps and needle valves to produce an all-glass GC-2 system. This simple and inexpensive system allows the quantitative analyses of phenols, hydrocarbons, acids, sterols, terpenoids, and alkaloids with excellent reproducibility and linear response. Even more impressive was its ability to analyze a C45-terpenoid alcohol, solanesol, as its TMS-derivative (M.W. 702).

    Notes: The purpose of the studies being carried out is to find some regularities of a general character which determine the relationship between the structures of the chromatographed substances and the composition of the mobile phase, i.e., between the log of the partition coefficient of the sample (Az), the K1-values which characterize the adsorption equilibrium of component “1” in a given chromatographic system, the structures of the mobile phase components and the structures of the substances being chromatographed. This paper deals with the relationship between the Az and K1 parameters and the structure of naphthalene and a number of its derivatives. It has been shown that there is a close relationship between the kind, size, frequency and site of the substituent on the one hand and the Az and K1 values on the other. In order to eliminate the possibility of unwanted side effects, the investigations were carried exclusively using non-active binary mobile phases of the type N + N or N + /B/. From these investigations, a comparison between the parameter Az and the surface of adsorbed molecules (As) was also possible.

    Notes: Several impurities which were suspected to be present in commercial reagent-grade phenol were separated using high performance liquid chromatography following sample concentration by steam distillation. A reverse-phase, microparticulate column and 5 solvent systems containing 5% to 25% methanol in 0.05 M phosphoric acid were used. A variable wavelength ultraviolet detector and an electrochemical detector were connected in series, in that order, to the column. Capacity factors (ḱ) for pyrogallol, hydroquinone, resorcinol, catechol, orcinol and phenol were determined in the 5 solvent systems and compared to ḱ-values of impurities present in the residue remaining from the steam distillation of phenol. Co-chromatography of standards suspected as contaminants and possessing ḱ-values similar to those of impurities were carried out in 3 solvent systems to confirm identical retention characteristics. Detector response ratios of contaminants and suspected standards, using various wavelengths and voltage potential settings, were utilized as major identification parameters. Two of the several contaminants separated were identified as hydroquinone and catechol.

How to do this with commercially available instruments, injectors and detectors is described in the paper.An attractive feature of the long narrow column approach is that the same HPLC set-up can be used for both fast-low and slow-high efficiency separations.High efficiency is essential for really complex natural mixtures, e.g. the hop and beer bitter acids.     Notes: The influence of chamber saturation in thin-layer chromatography on theoretical and experimental parameters of mixture separations were examined. Five different types of chambers for development of chromatograms were taken into consideration. The relation found experimentally differed widely from that calculated using Ościk's equation for different method of carrying out of the chromatographic process. The investigations presented here show the possibility of choosing chromatographic chambers which are optimum for theoretical studies on the process of thin-layer chromatography.

    Notes: A capillary gas chromatographic method for the quantitative analysis of diethylbenzene fractions is described. Estimation of ortho-, meta- and para-isomers and other C9 and C10 aromatic impurities is covered. The conditions developed involve the use of a capillary column of Carbowax-1540 (300 feet × 0.01 inch) under isothermal operation. The retention index data for a number of aromatics are presented at four temperatures (90, 100, 110 and 120°C). The method offers a good choice for any level of concentration both for isomers and impurities commonly encountered within a reasonable analysis time.

    Notes: Hydrocarbon group-type analysis of gasoline-range materials is a very important method for evaluating feedstocks and products in the petroleum industry. In this paper the method of group-type analysis of gasoline-range materials by high performance liquid chromatography is investigated.The results demonstrte that the separation of saturates and olefins can be accomplished with reactivated and cooled columns. The low-temperature columns and the use of n-hexane as an eluent play a particularly important role in the improvement of the separation.

    Notes: Different methods for producing a pre-assigned gradient in liquid chromatography are presented. A systematic approach to the calculations involved is given. The derived equations are mostly Poisson and Poisson summation distributions which are tabulated in the literature. The incremental method of gradient elution developed by Scott has been modified. In the modified apparatus two mixing chambers are used instead of one. This leads to an appreciable decrease in the number of reservoirs needed for the same precision in fitting a desired gradient. The application of the derived equations, together with other Varigrad modifications, will be given in Part 2 of this paper (to be published shortly in this Journal).

    Notes: A method is presented for the simultaneous determination of a wide range of carboxylic acids and phenols in water. Extractive alkylation is used with the tetrabutylammonium ion as counter ion and pentafluorobenzylbromide as alkylating agent. Extracts are analyzed by glass capillary gas chromatography and electron capture detection. Using one ml of water sample, the detection limit lies in the range of 1-10 μg/l. Application has been made to the analysis of steam condensate and domestic sewage effluent.

    Notes: Unequal elution of sample components of different volatility out of the syringe needle is one of the major causes for discrimination occuring during the injection, i.e. the phenomena which results in peak areas for the high boiling compounds which are too small compared with the volatiles. This problem, associated with all vaporizing injectors that are used with syringes, can be minimized by careful choice of the needle handling technique. Various methods are compared experimentally. The “solvent flush” method is discussed in detail and demonstrated to be ineffective for reducing losses in the syringe needle. The “hot needle” technique, where the empty needle is preheated in the injector before pushing the plunger, was found to be superior or equal to an improved “solvent flush” method (which included preheating the needle and omitting the air plug between sample and flushing solvent. Generally it was found that the discrimination due to the syringe needle was reduced for larger sample volumes, although no further changes were noticed when these exceeded 2.5 to 3μl.

    Notes: A simple but effective GC approach to odour evaluation and fraction collection with capillary columns is described. An allglass device splits the column effluent, with one splitter arm going to an FID. For odour evaluation, the other splitter arm is led via teflon tubing to the nose. Off-line heart-cutting is carried out by collecting the split effluent on a glass capillary microtrap, the inner walls of which are coated with OV-101. This allows recovery of nanogram amounts of material. Multipass operation or repeated collection leads to preparative capillary GC for further spectroscopic investigation. These techniques do not involve sophisticated equipment or valves.     Notes: The “Distribution Function Method”, which makes possible the measurement of systematic variations in the shape of signals, has been experimentally tested on the detection of two visually confounded gas chromatographic peaks, irrespective of the estimation of the resolution or the concentration ratio. Two detection procedures are suggested. The first compares directly, under the same experimental conditions, the shape of a pure peak and that of a peak suspected of contamination. The second makes use of variations of column temperature and gas flow rates to distinguish the behaviour of a pure substance from that of a contaminated substance. A marking procedure for the shape of a pure peak is then set up to dispense with the need of a reference standard. In practice, the two procedures make possible the detection of heptane as an impurity in n-hexane at levels of one percent and under experimental conditions artificially chosen so as to make the resolution inferior to 0.25.

    Notes: Basic expressions are derived for both the retention time and the effective separation factor in serially coupled GC columns. The retention time is determined by two main parameters. The first is the fractional time spent by an unretarded solute in each column which, in turn, is determined by the relative column lengths and flow velocities through each column. The second parameter is the relative mass distribution coefficient of a particular solute in each column; a variable that can be adjusted by changing the relative temperatures of the columns. The expression for the effective separation factor relates the measured separation factor for the series combination to the separation factors on the individual columns, the fractional time spent by an unretarded peak in each column, as well as the relative values of the mass distribution coefficients of a particular solute on the different columns.

    Notes: 1-(o-methoxyphenoxy)-3-isopropylamino-2-propanol hydrochloride (moprolol, OmeralR), is a new β-adrenergic blocking agent. A gas chromatographic method for determining unchanged moprolol in human biological fluids has been developed. After solvent extraction, a bis-trifluoroacetyl derivative was formed and measured by an electron capture detector. The quantification was checked by using an internal standard (pronethalol), which was added to all samples. The electron capture response was linear between 10 and 500 ng/ml. To avoid contamination of the peaks, careful extraction was imperative. The main advantages offered by the described method are: specificity, high sensitivity (10 ng.ml-1) and the small amount of biological fluids (1 ml plasma a. e.) required.

    Notes: Modern liquid chromatographic (LC) conditions which effect a separation of polycyclic aromatic hydrocarbons (PAH) containing up to ten rings are described. The LC conditions include reverse-phase chromatographic columns in combination with a quaternary solvent system. Methanol and water are combined to form solvent system A, while tetrahydrofuran and acetonitrile form solvent system B. Solvent systems A and B are then combined in a linear gradient relationship to effect the separation. The separation of benzo(a)pyrene and perylene along with benzo(ghi)perylene and indeno (1,2,3-cd)pyrene is achieved. In the same chromatogram the PAHs, coronene, dibenzo(a,e), (a,i), (a,h)pyrene and tribenzo(a,e,i)pyrene are also separated. An increase in column temperature from 45°C to 55°C is shown to shorten the elution time and increase chromatographic resolution of many of the PAHs examined. At least 25 PAHs ranging in molecular weight from 178 to 536 are examined for their chromatographic behavior. As examples, the application of this method to samples from a coal liquefication process are included. The samples are examined with both a UV detector and fluouescence detector to demonstrate the utility of dual detectors when analyzing for PAHs in complex matrices. With minor modification of the gradient this method is applicable to PAHs containing ring systems other than those reported here.

    Notes: Simple procedures are presented for separating the enantiomers of α-methyldopa, 5-hydroxytryptophan, tryptophan, triiodothyronine and thyroxine, which require neither special sorbents nor difficult-to-obtain or unstable reagents.The method for α-methyldopa, 5-hydroxytryptophan and tryptophan is based on the use of L-phenylalanine copper complex as the chiral constituent of the mobile phase; LiChrosorb® RP-18 serves as the stationary phase. The procedure for triiodothyronine and thyroxine is grounded on the L-proline copper complex as the chiral reagent and LiChrosorb® Si 60 as the stationary phase.In all observed cases, the D-enantiomer is eluted prior to the respective L-enantiomer. Chirality inversion of the mobile phase (application of the D-phenylalanine copper complex) reverses the order of elution; a racemic eluent (DL-phenylalanine copper complex) leads to no separation. In addition to the enantiomers of α-amino acids, the enantiomers of α-hydroxy acids (mandelic acid) can be separated.

A computer program has been designed for the chromatogram reconstruction of all valid sample runs from a system subsequence on an HP-2648 A graphics CRT terminal.     Notes: A method has been developed for the preparation of modified silica plates for high performance thin-layer chromatography (HPTLC). Some typical organosilanes were thus allowed to react in situ with the silica of Merck HPTLC-plates. This method was found to be highly reproducible, simple and cheap. Non-polar plates were prepared and compared with commercial plates from Merck, Whatman and Macherey-Nagel. Modification with cyanodecyltrichlorosilane resulted in plates that showed good coverage, efficiency and low residual silica activity. Silica modified with a multifunctional silane has different properties in different organic solvents. It will appear to be non-polar in a polar solvent and vice versa. New advantageous separation systems are thus made feasible by the presence of cyano groups on the plate.The utility of modified thin-layer plates is demonstrated by the separation of some homologues of p-hydroxybenzoic acid esters and of some polycyclic aromatic hydrocarbons.

    Notes: Glass capillary gas chromatography/mass spectrometry is used to profile Alaskan crude oil for purposes of origin verification. Alaskan oil, sampled at Valdez, Alaska, and transshipped to the U.S. East Coast, is compared with 21 samples of foreign crude oil using GC/MS techniques in which original data is reconstructed at selected parent and fragment ions to generate a series of chromatographic profiles. Comparison of selected profiles and compositional parameters derived from peak ratios allows distinction of Alaskan oil from many foreign crudes and may be applied to the examination of crude mixtures.
