    Notes: Intracytoplasmic μ-positive pre-B cells (μ+VH315− cells) transformed with Abelson virus continuously produced cells (μ+VH315+ cells) that were double-stained by anti-μ and anti-VH315 antibodies which specifically recognized the immunoglobulin heavy chain variable region identical or closely related to that of MOPC315 myeloma protein. Southern blot analysis indicated that the μ+VH315+ cells were generated from the μ+VH315- cells by the VH315·D·JH2 to a germline JH3 joining on the non-productive VH315·D·JH2 allele, which resulted in the production of μ-chains with variable region detectable by anti-VH315 antibodies. Therefore, it is strongly indicated that VHDJH to JH joining is not blocked in μ-chain-producing pre-B cells, and thus is free from allelic exclusion machinery.

    Notes: Rearrangements of immunoglobulin genes are mediated by highly conserved heptamer and nonamer recombinational signal sequence. Using a protein-blotting procedure, a heptamer and nonamer recombinational signal sequence-specific DNA-binding protein(s) was examined in the nuclear extracts from lymphoid and nonlymphoid cell lines. Nuclear extracts were subjected to SDS-polyacrylamide gel electrophoresis, and transferred by electroblotting to nitrocellulose filters. Then the filters were hybridized to *P-labelled synthetic double-stranded heptamer-23bp-nonamer or nonamer- l2bp-heptamer recombinational signal sequence probes. A relatively large amount of a DNA-binding protein(s) of M, 115,000 for both probes was detected in the nuclear extracts from immature B and immature T cell lines. No DNA-binding proteins were detected in a myeloma cell line. Interestingly, this DNA-binding protein(s) might be able to recognize both heptamer and nonamer. Recombinational signal sequence-specific DNA-binding activity of the protein(s) and the presence of the protein(s) in a stage-specific manner strongly suggest that the protein(s) of M, 115,000 detected here may play an important role in the recombination of Ig and TCR genes.

    Notes: Lipopolysaccharide (LPS), an endotoxin produced by Gram-negative bacteria, inhibits gonadotrophin secretion and ovulation in female mammals. The present study was undertaken to examine whether γ-aminobutyric acid (GABA)-synthesising neurones in the medial preoptic area (mPOA) may be involved in the inhibition by LPS of progesterone-induced luteinising hormone (LH) surge in ovariectomised oestrogen-primed rats. An intravenous injection of LPS (10 µg) at noon significantly increased the content of glutamic acid decarboxylase 67 (GAD67), a GABA-synthesising enzyme, in brain tissues including the mPOA. LPS treatment also significantly increased the number of detectable GAD67-immunoreactive (ir) as well as GABA-ir cells in the mPOA, compared to control rats. The same LPS challenge completely eliminated the steroid-induced LH surge, which was normally induced in control rats in the afternoon. A local injection of muscimol (100 ng), a GABAA receptor agonist, into the mPOA also significantly attenuated the LH surge, mimicking the effect of LPS. These results suggest that LPS stimulates GABA synthesis in preoptic neurones, which in turn inhibit the LH surge in female rats. The results also support the hypothesis that diminution of the GABAergic suppressive activity in the mPOA permits the LH surge to be induced.

    Notes: Abstract The significance of the development of polyglucosan bodies (PBs) in the CNS is incompletely understood. We present the clinicopathological features of three autopsy cases with numerous PBs other than the common corpora amylacea or Lafora bodies. The first patient had pleomorphic PBs in the neuronal processes of pallidum and substantia nigra which, thus, are consistent with Bielschowsky bodies. Bielschowsky bodies involved also the hypothalamus and tegmentum of midbrain and medulla. The present case was the first not associated with any clinical symptoms. The second patient also had incidental Bielschowsky bodies in the external pallidum, substantia nigra, and pallidothalamic, pallidonigral and nigrostriatal tracts. Additionally, unique clusters of small PBs appeared in the cerebral cortex, putamen, pallidum, and caudate nucleus. Immunostaining suggested that these small clustered PBs were located in the cytoplasm and processes of astrocytes. Ultrastructurally, these clustered PBs were in agreement with previous descriptions of PBs. The third patient had adult polyglucosan body disease. Most PBs in the white matter were corpora amylacea situated in astrocytic processes or axons. In the gray matter, many pleomorphic PBs resembling Bielschowsky bodies occurred in neuronal processes. In the peripheral nervous system, a few PBs were seen in myelinated axons. The following conclusions may be drawn from this study: (1) each type of PBs develops in distinct cell types of the human brain in variable distribution; (2) Bielschowsky bodies may not manifest clinically; (3) PBs other than corpora amylacea or Lafora bodies may be distributed in localized or diffuse patterns; (4) in the localized pattern (patients 1 and 2), PBs occur as Bielschowsky bodies or clustered PBs, and tend to involve certain systems (pallidum, striatum, and substantia nigra); and (5) in the diffuse pattern (patient 3), PBs develop as corpora amylacea and Bielschowsky-like bodies of adult polyglucosan body disease.

    Notes: Summary Abnormal granular structures, which stained positively with periodic acid-Schiff (PAS-positive granular structures; PGS), were observed in the brain of senescence accelerated mouse (SAM). They were small, round to ovoid, homogenous structures measuring up to 5 μm in diameter and usually grouped in clusters. PGS were localized in the hippocampus, piriform cortices, olfactory tubercle, nucleus of the trapezoid body, and cerebellar cortices. Quantitative analysis revealed that PGS remarkably increased in the hippocampus of SAM-P/8, a substrain of SAM, with advancing age, although a few PGS also appeared in the aged control mice, SAM-R/1 and DDD. Their histochemical nature, morphological features and distribution pattern were different from those of corpora amylacea and other similar bodies. A close anatomical relationship between PGS and glial fibrillary acidic protein-positive astrocytes was inferred from immunohistochemical studies. PGS is considered to be one of the morphological manifestations of senescence in mice brains, and are found to occur more numerously in the brains of learning or memory deficit mice, SAM-P/8.

    Notes: Summary The effects of nutrition on the timing of reproductive initiation of a short-day annual plant Xanthium canadense (cocklebur) were examined with the following hypotheses in mind: If the plant always follows an optimal growth schedule, low-nutrient plants will initiate reproductive growth earlier than high-nutrient plants. On the other hand, if the plant flowers in response to photoperiodic stimuli, both plants will initiate reproductive growth on the same day. The sand-culture experiment showed that high-nutrient plants flowered earlier than the low-nutrient plants, leading to rejection of the first hypothesis. The predicted optimal flowering time is 2 days later than the actual flowering time in high-nutrient plants and 10 days earlier in low-nutrient plants. These deviations from the optimal times reduced the reproductive yield by 0.1% and 2.3%, respectively. The ratio of the final reproductive yield to the vegetative mass at flower initiation was 1.10 in high-nutrient plants and 0.63 in low-nutrient plants. Since the expected ratio for the optimal growth schedule is 1.0, high-nutrient plants followed the opitmal growth schedule more closely than the low-nutrient plants. Cocklebur is a fast-growing annual which is common in relatively nutrient-rich environments. This study suggests that cocklebur adapts itself to such environments through its photoperiodic response.

    Notes: Abstract Five saponites with various substitution degrees of the constituent OH by fluorine were subjected to complex formation with α-naphthylamine to examine the effect of substitution on the stabilities of the resulting complexes. Saponite with a larger substitution degree intercalated a lesser quantity of a-naphthylamine and was broken down more easily in the reaction process. The complexes were subsequently heated in nitrogen to examine their thermostabilities. The thermostability of saponite was enhanced with increasing degree of substitution. All saponite, especially those without substitution, enhanced their thermostabilities by intercalating with α-naphthylamine. The reasons for these phenomena are briefly discussed.

    Notes: Abstract Samples of montmorillonite/α-naphthylamine complex (MNC), saponite/α-naphthylamine complex (SNC) and their mixtures were heated at temperatures below 1473 K under a nitrogen atmosphere to examine macroscopic structural change and crystalline phases deposited after pyrolysis. SNC resulted in a dense structure after pyrolysis. Heating of samples of MNC/SNC mixtures gave a porous structure which became dense as the proportion of SNC increased. No porosity measured by the mercury porosimeter was detectable in samples pyrolysed from MNC containing more than 25% SNC. After pyrolysis, the MNC/SNC mixtures gave crystalline phases which were never formed in the pyrolysed pure MNC and SNC. The formation of these crystalline phases was explained by reference to the chemical compositions of the mixtures and the phase diagram.

    Notes: Abstract A total of 18 enzymes (encoded by 20 loci) fromParagonimus heterotremus collected at a local area in Saraburi, Thailand, were electrophoretically compared with those from the three Japanese speciesP. ohirai, P. miyazakii andP. westermani using horizontal starchgel electrophoresis. Relative genetic distances between these species were quantified. The ThaiP. heterotremus was found to be most closely related toP. miyazaki and rather distantly related toP. westermani. On the other hand, among the three Japanese species, the closest relationship was observed betweenP. miyazakii andP. ohirai.
