    Notes: Objective— To observe if a combination of a depot GnRH agonist and cyclical hormone replacement therapy decreases menstrual b od loss.Design— open, observational study comparing the objective assessment of menstrual blood loss before, during and after 3 months treatment.Subjects— 20 women with a subjective complaint of heavy menstrual loss in whom no cause could be discovered.Interventions— Each woman received 3 months of depot goserelin (Zoladex) combined with cyclical hormone replacement therapy (Cyclo-Progynova, 1 mg). Menstrual loss and symptoms were assessed before, throughout and after the study.Main outcome measures— Changes in objective and subjective assessments of menstrual blood loss and the acceptability of the treatment.Results— The median pretreatment menstrual loss was 68 ml (range 23–397). Only 8 (40%) of the patients had a loss exceeding 80 ml per period. The median blood loss was 30 ml, 16 ml, and 17 ml in the three treatment cycles (P 〈 0.001 Wilcoxon rank sum for the third cycle). There was a significant decrease in the median length of menstruation (P 〈 0.001) and the number of towels or tampons (P 〈 0.01) used per period in the third treatment cycle. There was a significant decrease (P 〈 0.005) in the number of women complaining of dysmenorrhoea, premenstrual symptoms, flooding and the passage of clots. Seventeen patients experienced hot flushes. Eighteen of the 20 patients were completely satisfied with the treatment and would have been happy to continue with it for longer than 12 months.Conclusions— The combination of a depot gonadotrophin releasing hormone agonist and cyclical hormone replacement therapy is a successful and acceptable treatment of dysfunctional uterine bleeding.

    Notes: Gold powder was mixed with YBa2Cu3O7−x superconductors for the purpose of increasing the critical current density (Jc). The critical transition temperature at zero resistance (Tzeroc) gradually decreased by the Au addition. However, the depression of Tzeroc was as low as 2 K for the YBa2Cu3O7−x -Au composite with Au mixing up to 40 wt.%. A maximum Jc of 307 A/cm2 was obtained for the Au 5-wt. % mixed composite. The Jc was about four times as high as that for YBa2Cu3O7−x without Au mixing. In addition to the Jc enhancement, the weak-link problem was appreciably improved by the Au addition.

    Notes: We have initiated studies to determine whether the antibody and T-lymphocyte proliferative responses to lysozyme and its antigenic sites is genetically controlled in mice. Mice of the H-2f, H-2k and H-2p were high responders, while haplotypes H-2b, H-2d, H-2r and H-2s were low responders. Studies with recombinants indicated that the immune response is controlled by two H-2I region loci, one being in the I-A subregion and the other may be in the I-C subregions.

    Notes: This study examined the serum IgG and IgM responses against Porphyromonas gingivalis and 3 serotypes of Actinobacillus actinomycetemcomitans, and the correlations of these responses with age and homologous infection. A total of 90 individuals were included in this study: 40 subjects with gingivitis, 40 periodontally healthy subjects, and 10 adult periodontitis subjects. The subjects in the gingivitis and periodontally healthy groups were divided into 4 stages based on their physiological age: early childhood, school age, puberty, and adult. In the gingivitis group, there was a positive correlation between increase in age and increase in serum IgG antibody levels against P. gingivalis until puberty. However, no statistically significant difference was found between the puberty stage and the adult stage. The average level of IgG antibodies against A. actinomycetemcomitans in the school age gingivitis group was significantly higher than that in the early childhood gingivitis group for all serotypes (p 〈 0.01). In serotype c, IgG antibody levels in the school age gingivitis group were significantly higher than in the early childhood gingivitis group or the adult gingivitis group (p 〈 0.01). With both P. gingivalis and A. actinomycetemcomitans, positive correlations between elevated IgG level and infections by these microorganisms were found in the puberty gingivitis and adult periodontitis groups.

    Notes: Abstract. The present study was performed to evaluate the clinical, microbiological and immunological aspects in the early stages of recurrent periodontal disease. After clinical monitoring of pockets with recent evidence of disease recurrence, microbiological samples for cultural analysis, serum and gingival crevicular fluid (GCF) samples were taken for IgG antibody analysis from 14 sites, 7 in 6 recurrent periodontitis patients and 7 in 7 periodontally healthy control subjects. IgG responses of serum antibody to 8 gram-negative bacterial strains were compared with those of GCF sampled from the recurrent site. The results clearly demonstrated the predominance of Bacteroides gingivalis in most subgingival plaque samples during the early stages of disease recurrence; the mean proportions of B. gingivalis were significantly different from those of the healthy sites (p〈0.05). 4 out of 6 serum samples showed the elevated antibody responses to B. gingivalis 381: and this was closely correlated to homologous infection by this micro-organism in recurrent sites. Elevated serum antibody responses were also noted to Eikenella corrodens 1073 and Actinobacillus actinomycetemcomitans Y4. However, no relationship with homologous infection was found for A. actinomycetemcomitans. 3 out of 6 GCF samples had greater antibody titers than the serum, suggesting local antibody synthesis by the gingival cells in the recurrent pockets. The present study showed that B. gingivalis might play an important role in the pathogenesis of disease recurrence in its early stages.

    Notes: In vitro antigenic reactivity of chemically synthesized monosaccharide analogues of nonreducing sugar moiety of lipid A with 4 monoclonal antibodies against Salmonella minnesota R595 lipid A was studied by the inhibition test of enzyme-linked immunosorbent assay (ELISA). In the assays with 2 monoclonal antibodies (mAb 5G and mAb36G), which have been suggested to recognize the Salmonella type lipid A structure, no antigenic reactivity of the tested analogues was observed. However, several monosaccharide analogues exhibited antigenic reactivities with other monoclonal antibodies (mAb161M and mAb19M), especially some structural differences of hydrophobic and/or hydrophilic parts among monosaccharide analogues were recognized by mAb161M.

    Notes: Abstract The specificity of monoclonal antibodies directed against the Salmonella minnesota R595 lipid A and the structural requirements of lipid A epitopes were studied with chemically synthesized lipid A analogues by enzyme-linked immunosorbent assay and its inhibition test. The results suggest that lipid A has specific and common epitopes, in which the specificities are derived from the chemical and conformational structures of the backbone and/or acyl groups.

    Notes: Summary Autoantibody-secreting hybrid cell lines were obtained by fusion of spleen cells from unimmunized (NZB×NZW) F1 mice with the HAT-sensitive mouse myeloma cell line SP2/0-Ag14. Eight hybridoma cell lines producing autoantibodies to mouse thymocytes were cloned and the resultant antibodies were partially characterized. All eight monoclonal antibodies lysed mouse thymocytes in the presence of rabbit complement. The anti-thymocyte cytotoxic antibody activities were absorbed with thymocytes, lymph node cells, unfractionated spleen cells, and splenic T cells; but not with bone marrow cells, splenic B cells, or homogenates of mouse kidney, liver, or striated muscle cells. In addition, the cytotoxic activities of culture supernatants from seven of the eight hybrid clones were absorbed with mouse brain tissue homogenates. Isotyping of the monoclonal antibodies revealed that five were IgM and three were IgG2a. Mouse thymocytes sensitized with each of the eight monoclonal antibodies in vitro became highly susceptible to phagocytosis by syngeneic macrophages. The monoclonal antithymocyte antibodies, thus, appear to be similar to the naturally occurring, (NZB×NZW) F1 thymocytotoxic autoantibodies (NTA) described by Shirai et al. [20–23].
