    Basement membrane components outline the tumour islands in cylindroma (1984)

    Notes: The main histological feature of cylindroma is the deposition of sheaths of a‘hyalinized’ material contiguous to the tumour cell clusters. Although ultrastructural studies of this material have revealed a basement membrane-like structure, its exact nature has remained unclear. Using immuno-staining with affinity-purified antibodies directed against distinct basement membrane components, we have shown that type IV collagen and laminin are major constituents of this zone. In addition, cell culture studies indicated that both proteins are synthesized by the tumour cells. The immunohistological data make it clear that the tumour matrix between the tumour cell islands is composed not only of basement membrane components, but also is composed of other connective tissue constituents, i.e. type I and III collagen and fibronectin.

Dieses Protein besitzt unter experimentellen Bedingungen bei Vorliegen einer xenogenen Beziehung immunogene Eigenschaften. Die Immunogenität ist jedoch gering und Spuren von Begleitproteinen wie z. B. Serumproteine können zu einer beträchtlichen zusätzlichen Immunantwort führen. Die Antikörper gegen Kollagen sind gegen mindestens drei verschiedene Klassen von antigenen Determinanten gerichtet, die an verschiedenen Stellen des Moleküls lokalisiert sind. Die wichtigsten von ihnen befinden sich an den Endbereichen und können durch Proteasen oder eine Alkalibehandlung zerstört werden. Diese antigenen Determinanten zeigen in Hinblick auf ihre Struktur beträchtliche Speciesunterschiede. Die Fragen bezüglich einer Immunantwort gegen allogenes Kollagen bzw. ob Antikörper gegen Kollagen überhaupt eine Transplantationsabsto/gbung herbeiführen können, sind vorläufig noch ungeklärt.
    Notes: Summary Tissue substitutes contain mainly collagen. This protein exhibits under experimental conditions of a xenogenic species relation immunogenic properties. The immunogenicity, however, is of a low degree and trace contaminants like serum proteins may evoke a considerable and independent immune response. The antibodies to collagen are directed to at least three distinct groups of antigenic determinants which are localized in different regions of the molecule. The most important antigenic determinants are found on the terminal sites and can be destroyed either by proteases or by an alkaline treatment. They vary considerably in structure from one species to the other. The questions whether allogenic collagen behaves as an immunogenic protein or if at all antibodies to collagen can reject transplantates still remain to be answered.

    Notes: Summary Glomerular basement membrane synthesis was measured in vivo in diabetic rats at various times after streptozotocin injection. Simultaneously the type IV collagen concentration in serum was determined by a radioimmunoassay specific for the terminal cross-linking domain, 7S collagen. Basement membrane collagen synthesis and the 7S collagen levels were markedly raised in diabetic animals 18–34 days after streptozotocin injection and showed a significant correlation. The results suggest that the amount of serum 7S collagen reflects increased basement membrane synthesis in a diabetic situation. Both basement membrane collagen synthesis and serum 7S collagen concentration also show a positive correlation with the blood glucose levels of diabetic rats. This indicates that the degree of metabolic dysregulation influences basement membrane synthesis.

Es handelt sich dabei um eine spezifische Verminderung der Immunantwort, die als partielle Toleranz interpretiert wird.
    Notes: Summary The humoral immune response towards L-asparaginase (Crasnitin®) in adult rats could be significantly reduced after 3 weeks of intravenous pretreatment with cyanogenbromide-cleaved L-asparaginase. The operation of the immune response seems specific and was interpreted in terms of induction of a partial immunological tolerance.
