    Notes: Summary Glomerular basement membrane synthesis was measured in vivo in diabetic rats at various times after streptozotocin injection. Simultaneously the type IV collagen concentration in serum was determined by a radioimmunoassay specific for the terminal cross-linking domain, 7S collagen. Basement membrane collagen synthesis and the 7S collagen levels were markedly raised in diabetic animals 18–34 days after streptozotocin injection and showed a significant correlation. The results suggest that the amount of serum 7S collagen reflects increased basement membrane synthesis in a diabetic situation. Both basement membrane collagen synthesis and serum 7S collagen concentration also show a positive correlation with the blood glucose levels of diabetic rats. This indicates that the degree of metabolic dysregulation influences basement membrane synthesis.

    Notes: Abstract. The calcium-binding extracellular matrix protein BM-40 was obtained as a mouse cDNA product from a stably transfected kidney cell clone. Electrophoresis and N-terminal sequence analysis demonstrated absence of the proteolytic processing previously observed for a mouse tumour-derived BM-40. Yet the two forms of BM-40 were very similar in their CD spectra, their calcium-dependent change in α helix content and their immunological epitopes. In surface plasmon resonance assays, recombinant mouse BM-40 showed distinct binding to the triple-helical domains of collagens I, II, III, IV and V with Kd=1–4 μM but no binding to collagen VI. These interactions were abolished in the presence of EDTA. Tissue-derived mouse BM-40, however, bound collagens I and IV with Kd=0.1–0.2 μM. Activation of collagen binding to give a similar Kd could be achieved for recombinant mouse BM-40 by treatment with the matrix metalloproteinase collagenase-3. The major cleavage site was located in helix C of the extracellular calcium-binding module of BM-40 and other less prominent cleavages occurred close to the N-terminus. The sensitive helix C site was just one residue away from that sensitive to endogenous tissue proteolysis, suggesting that cleavage could be a physiological mechanism to modulate collagen binding.

    Description / Table of Contents: Summary Patients with rheumatoid arthritis were investigated serologically with the antiglobulin-consumption test and surveyed clinically over a period of three years. In a group of 226 cases with rheumatoid arthritis, showing a mild or less severe course of disease, 66 patients (29,2%) possessed antibodies to collagen. In a group of 50 patients with a severe and quickly progressing form of rheumatoid arthritis 44 patients (88,0%) showed collagen antibodies. In this group 16 cases showed also antinuclear antibodies, however, with lower titers as observed usually in lupus erythematosus. The demonstration of collagen antibodies, and in addition eventually also of antinuclear antibodies is regarded as an indication of an unfavourable prognosis for rheumatoid arthritis. — Cases with lupus erythematosus, Felty's syndrome collagen disorders and controls were investigated in addition serologically with different antigens (collagen, thymus-nuclei, heart homogenate, blood vessel homogenate, leucocytes). The serologic results and the symptoms of these patients were compared with the observations obtained in cases with rheumatoid arthritis.
    Notes: Zusammenfassung Ein größeres, klinisch genau kontrolliertes und durch 3 Jahre beobachtetes Krankengut mit primär chronischer Polyarthritis wurde mit dem Antiglobulin-Konsumptionstest auf das Auftreten von Kollagenantikörpern untersucht. Von 226 Fällen mit leichterem Krankheitsverlauf zeigten 66 Patienten (29,2%) Kollagenantikörper. Von 50 Patienten mit einem schweren und sich sehr rasch verschlechternden Krankheitsverlauf zeigten 44 Fälle (88,0%) Kollagenantikörper. Gleichzeitig waren bei 16 Fällen dieser Patientengruppe Zellkernantikörper nachweisbar, allerdings mit schwächeren Titerwerten als es bei Lupus erythematodes zu beobachten ist. Der positive Nachweis von Kollagenantikörpern, allenfalls mit gleichzeitigem Auftreten von Zellkernantikörpern wird als Hinweis auf eine ungünstige Prognose bewertet. — Weiter wurden Fälle mit Lupus erythematodes, Felty-Syndrom, Kollagenose bzw. Kontrollfälle mit einem Antigenspektrum (Kollagen, Thymuskerne, Herzhomogenat, Gefäßhomogenat, Leukocyten) serologisch untersucht. Das klinische Bild und die serologischen Befunde bei diesen Patienten wurde mit dem Ergebnis der Untersuchungen bei primär chronischer Polyarthritis verglichen.

    Description / Table of Contents: Zusammenfassung In Untersuchungen an erwachsenen Ratten konnte nachgewiesen werden, daß die humorale Immunantwort gegen das Antigen L-Asparaginase, das in Form des Präparates Crasnitin® verabreicht wurde, nach einer dreiwöchigen intravenösen Vorbehandlung mit Bromcyan-gespaltener L-Asparaginase signifikant herabgesetzt ist. Es handelt sich dabei um eine spezifische Verminderung der Immunantwort, die als partielle Toleranz interpretiert wird.
    Notes: Summary The humoral immune response towards L-asparaginase (Crasnitin®) in adult rats could be significantly reduced after 3 weeks of intravenous pretreatment with cyanogenbromide-cleaved L-asparaginase. The operation of the immune response seems specific and was interpreted in terms of induction of a partial immunological tolerance.
