Summary After injection of a priming dose glucose-U-C14 was given by a constant intravenous infusion over a period of 3 hours to nine persons without metabolic disorder in the fasting state. Several times during the infusion arterial blood was drawn and the specific activity of plasma glucose was estimated after removal of the acid substances by use of a strongly basic ion exchange resin. Radioactivity of plasma triglycerides was determined after extraction of arterial plasma by the method ofFolch and separation of the different lipid fractions by thin layer chromatography. The incorporation rate of plasma glucose-carbon into plasma triglycerides was calculated from specific activity of plasma glucose and the increase of radioactivity in plasma triglycerides. In the persons examined the mean incorporation rate of plasma glucose-carbon into plasma triglycerides was 18.4 µg/min/1000 ml plasma. The total radioactivity was detected in the glyceride-glycerol. In all the other lipid fractions of the plasma (glyceride-fatty acids, free fatty acids, phospholipids, free and esterified cholesterol) no radioactivity could be detected after the 3 hour infusion of glucose-U-C14. Total plasma triglyceride influx was measured by means of constant infusion of palmitic acid-9,10-H3. Glucose-U-C14 was given simultaneously. Using this double label method the percentage of glycerol, which is derived from plasma glucose and has been incorporated into plasma triglycerides, in total triglyceride influx could be calculated. Under the conditions mentioned above, this percentage was 20–71%.

Zusammenfassung An 9 stoffwechselgesunden Personen wurde Glucose-U-C14 nach Verabreichung einer „Priming Dose“ in konstanter Geschwindigkeit über einen Zeitraum von 3 Std intravenös infundiert. Während der Infusion wurde zu verschiedenen Zeitpunkten arterielles Blut abgenommen und die spezifische Aktivität der Plasmaglucose bestimmt; die Radioaktivität der Plasmaglucose wurde nach Entfernung der sauren Substanzen mittels eines stark basischen Ionenaustauschers gemessen. Vor Bestimmung der Radioaktivität in den Plasma-TG wurde arterielles Plasma nachFolch extrahiert und hierauf die einzelnen Lipidfraktionen dünnschicht-chromatographisch getrennt. Aus der spezifischen Aktivität der Plasmaglucose und dem Anstieg der Radioaktivität in den Plasma-TG wurde die Einbaurate von Plasmaglucose-Kohlenstoff in Plasma-TG berechnet. Sie betrug bei den untersuchten Personen im Mittel $$\bar x$$ =18,4 γ/min/1000 ml Plasma,s=6,23. Die gesamte Radioaktivität befand sich unter den gegebenen Versuchsbedingungen im Glycerol-Anteil der Triglyceride. In den übrigen Lipidfraktionen des Plasmas (freie Fettsäuren, Phospholipide, freies und verestertes Cholesterin) konnte während der dreistündigen Infusion von Glucose-U-C14 keine Radioaktivität nachgewiesen werden. Durch Bestimmung des Plasma-TG-Influx mittels konstanter Infusion von Palmitinsäure-9,10-H3, die gleichzeitig mit der Infusion der markierten Glucose verabreicht wurde, konnte berechnet werden, wie groß der Anteil des aus Blutglucose stammenden Glycerols am gesamten Glycerol ist, das in Plasma-TG eingebaut wird: Bei den untersuchten Personen wurden unter den gegebenen Bedingungen zwischen 20 und 71% des in Plasma-TG eingebauten Glycerols durch die Plasmaglucose geliefert.
