    Notes: Summary A possible pathogenic mutation in the Β3-adrenergic-receptor gene (Trp64Arg) has been reported to be associated with an earlier age of onset of non-insulin-dependent diabetes mellitus (NIDDM) and clinical features of the insulin resistance syndrome in Pima Indian, Finnish and French subjects. Since marked heterogeneity has been reported in the association of mutations of candidate genes with NIDDM between Japanese and other ethnic groups, we investigated the association of Trp64Arg with NIDDM in Japanese subjects. The allele frequency of the mutation (Arg) was slightly, but not significantly, higher in NIDDM than in control subjects (70 out of 342 alleles [20.5%] vs 40 out of 248 [16.1%], respectively, p〉0.2). When our data were combined with those of Pima Indian and Finnish subjects, however, the Arg/Arg genotype was significantly associated with NIDDM as compared with the other two genotypes (p〈0.005, relative risk [RR] 2.13, 95% confidence interval [CI] 1.28–3.55). The Arg allele was also associated with NIDDM (p〈0.05, RR 1.27, 95% CI 1.06–1.52). Japanese subjects homozygous for the mutation had a significantly higher body mass index (mean ± SD∶25.5±3.9 kg/ m2) than heterozygotes (22.6±4.1, p〈0.05) and normal homozygotes (22.8±3.8, p〈0.05). NIDDM patients homozygous for the mutation tended to have an earlier age of onset of NIDDM than those with other genotypes. These data suggest that the Trp64Arg mutation not only contributes to weight gain and age-at-onset of NIDDM but is also associated with susceptibility to NIDDM.

    Notes: Summary Cystic acoustic neurinomas (ANs) are less frequent and are different from solid ANs in clinical and radiological features. We had 14 cystic ANs (13.5% of 104 cases) in the last 17 years. Computerized tomographic or magnetic resonance images allowed for the classification of these cystic ANs into three types: Type A being large single cysts with a thin tumourous wall (7 cases); type B single cysts with a thick tumourous wall (3 cases); type C multicystic (4 cases). Half of the cystic ANs were not accompanied by enlargement of the internal auditory canal, despite the largeness of the cysts. The mean size of the tumours was 29 mm in diameter. Type A cysts had a shorter clinical history than types B and C. One patient had intact hearing. In five cases, an atypical initial symptom such as facial pain, dysgeusia, facial palsy, unsteadiness or vertigo presented. The trigeminal nerve was involved in 12 cases, the facial nerve in nine. The characteristic features of cystic ANs are largeness of the tumour, a short clinical history, an atypical initial symptom, facial nerve involvement, and/or no enlargement of the internal auditory canal. In addition, the histological features are a lobular growth pattern, high nuclear atypia, and numerous macro phages.

    Notes: Abstract We studied 184 Japanese males for the tetranucleotide TAGA repeat polymorphism at the Y -27H39 locus on the Y chromosome, and discovered a new allele, tentatively named F. Direct sequencing of Y -27H39 alleles revealed that the new allele (206 nt) is larger than allele E (202 nt) by 1 repeat unit. The six alleles differ only in the number of repeats, the flanking sequences being constant. The allele frequencies are different in Japanese and Caucasians.

    Notes: Abstract Cachexia frequently occurs in the late stages of cancer, and is difficult to manage. We previously reported that interleukin-6 (IL-6) cDNA transfection into Lewis lung carcinoma (LLC-IL6) induced cachexia-like symptoms in C57BL/6 mice. This was thought to be a useful experimental model of cancer cachexia. We have examined the effects of two eicosanoids, docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA), in order to evaluate whether they could relieve cachexia. LLC-IL6-bearing animals were divided into three treatment groups receiving DHA, EPA or water as the control; 80-μl samples of these compounds (purity〉95%) were administered orally by catheter daily starting 7 days after tumor transplantation. Tumor growth curves were similar in the three groups. There were no differences in water or food intake in the three groups. However, body weight, a marker of cachexia, was significantly higher in treated mice than in the control group. Sixteen days after tumor transplantation, the mean body weight was 17.45 g (P〈0.05), 17.2 g and 16.41 g in the groups receiving DHA, EPA and water respectively. The eicosanoids did not affect serum levels of IL-6. Ubiquitination of muscle protein, a marker of proteolysis coupled to cachexia, was compared in LLC-IL6-and LLC-transplanted mice. The eicosanoids prevented the ubiquitination of approximately 180 kDa protein. These results suggest that eicosanoids may prevent the cachexia mediated by IL-6.

    Notes: Sphingomonas paucimobilis SYK-6 is able to grow on a wide variety of dimeric lignin compounds. These compounds are degraded via vanillate and syringate by a unique enzymatic system, composed of etherases, O demethylases, ring cleavage oxygenases and side chain cleaving enzymes. These unique and specific lignin modification enzymes are thought to be powerful tools for utilization of the most abundant aromatic biomass, lignin. Here, we focus on the genes and enzymes involved in β-aryl ether cleavage and biphenyl degradation. Two unique etherases are involved in the reductive cleavage of β-aryl ether. These two etherases have amino acid sequence similarity with the glutathione S-transferases, and use glutathione as a hydrogen donor. It was found that 5,5′-dehydrodivanillate, which is a typical lignin-related biphenyl structure, was transformed into 5-carboxyvanillate by the reaction sequence of O-demethylation, meta-ring cleavage, and hydrolysis, and the genes involved in the latter two reactions have been characterized. Vanillate and syringate are the most common intermediate metabolites in lignin catabolism. These compounds are initially O-demethylated and the resulting diol compounds, protocatechuate (PCA) and 3-O-methylgallate, respectively, are subjected to ring cleavage catalyzed by PCA 4,5-dioxygenase. The ring cleavage products generated are further degraded through the PCA 4,5-cleavage pathway. We have isolated and characterized genes for enzymes involved in this pathway. Disruption of a gene for 2-pyrone-4,6-dicarboxylate hydrolase (ligI) in this pathway suggested that an alternative route for 3-O-methylgallate degradation, in which ligI is not involved, would play a role in syringate catabolism. In this article, we describe the genetic and biochemical features of the S. paucimobilis SYK-6 genes involved in degradation of lignin-related compounds. A possible application of the SYK-6 lignin degradation system to produce a valuable chemical material is also described.

    Notes: Abstract The β-NMR spectra of 21F, 23Mg, 27Si and 39Ca which were produced in heavy ion collisions and implanted in various crystals have been observed. The magnetic moments of 21F and 27Si were determined to be |μ(21F)| = 3.9194 ± 0.0012 μN and |μ(27Si)| = 0.8653 ± 0.0003 μN, respectively. The electric quadrupole coupling constants were determined for the first time to be |eqQ(21F in MgF2)/h|= 9.94 ± 0.09 MHz, |eqQ(23Mg in MgF2)/h|= 1.96 ± 0.06 MHz, |eqQ(27Si in Al2O3)/h|= 1.90 ± 0.12 MHz, |eqQ(39Ca in CaCO3)/h|= 0.60 ± 0.04 MHz. From the present eqQ/h, the Q moments were deduced as |Q(21F)|= 110 ± 22 mb, |Q(23Mg)|= 114 ± 3 mb, |Q(27Si)|= 60 ± 13 mb and |Q(39Ca)|= 36± 7 mb. The present data were compared with the theoretical values obtained by the OXBASH shell model code.

    Notes: Abstract Hyperfine interactions of 8Li impurity nucleus imbedded in ferromagnetic Ni metal were studied using the β-NMR technique. Two kinds of hyperfine fields B82 were found, corresponding to two different final sites of Li atoms in the Ni lattice. The nuclear spin-lattice relaxation times T1 of 8Li in Ni were also determined for each field. Temperature dependencies of B82 and T1 were observed to deduce these values at T=0 K that can be compared with those calculated recently by Akai et al.

    Notes: Abstract The electric quadrupole interaction of 19O(Iπ=(5/2)+, T1/2=27.0 s) in TiO2 single crystal was studied in detail by means of the β-NQR to determine the electric quadrupole moments Q of short-lived β-emitting nuclei 19O and 13O(Iπ=(3/2)-, T1/2=8.6 ms). Two implantation sites were found for the implanted O nucleus and the quadrupole coupling constants of 19O at these sites were determined. We observed FT-NMR of the enriched stable isotope 17O in TiO2 and obtained the electric field gradient (EFG) at the oxygen substitutional site. With this knowledge, we have determined Q(13O)=11.0 ± 1.3 mb and Q(19O)=3.7 ± 0.4 mb. The present results are compared with the theoretical values calculated by the shell model code, OXBASH and by the Hartree–Fock calculation with the realistic potential.

    Notes: Abstract The spin manipulation technique by use of the β-NMR is further refined, based on the recent thorough studies of hyperfine interactions of 12B and 12N in Mg. By using this technique, the alignment correlation terms in the β-ray angular distribution of the A = 12 system are precisely measured, so that the axial charge is singled out to be y = 4.66 ± 0.06 (stat.) ± 0.13 (syst.) which shows clear mesonic enhancement over the impulse value.
