    Notes: The recombinant bacillus Calmette–Guérin (rBCG) secretion system utilizing an extracellular α antigen of Mycobacterium kansasii (α-K) was characterized biochemically and immunologically. The human immunodeficiency virus type1 (HIV-1) p17gag B cell epitope fused to α-K was secreted in extremely large amounts. At least three mice out of seven inoculated with rBCG generated high titres of antibody to the epitope. The long-lasting antibody production persisted more than 14 months.

    Notes: Abstract We examined the different vasodilatory effects of the K+ channel openers levcromakalim and 5-amino-N-[2-(2-chlorophenyl)ethyl]-N′-cyano-3-pyridinecarboxamidine (KRN4884), and the Ca2+ channel blocker nifedipine in the rat aorta. KRN4884 (10–10–10–5 M) and nifedipine (10–10–10–5 M) produced concentration-dependent relaxation in the rat aorta precontracted by 25 mM KCl. The K+ channel blocker glibenclamide (1 μM) inhibited the relaxation induced by KRN4884 but did not influence nifedipine-induced relaxation. KRN4884 had almost no effect on contraction induced by 80 mM KCl, whereas nifedipine completely relaxed the muscle precontracted by 80 mM KCl, whereas nifedipine completely relaxed the muscle precontracted by 80 mM KCl. These results indicate that KRN4884 is a K+ channel opener. We investigated the relaxant effects of KRN4884 (10–10–10–5 M), levcromakalim (10–9–10–5 M) and nifedipine (10–9–10–5 M) on A23187 (1 μM)-induced contraction. KRN4884 and levcromakalim had a potent relaxant effect but nifedipine only a weak effect on the smooth muscle contracted by A23187. Glibenclamide (1 μM) inhibited the relaxation induced by KRN4884 and levcromakalim, but did not influence the nifedipine-induced relaxation. KRN4884 (1 μM) produced a larger relaxation of A23187-induced contraction but had little effect on the increase in intracellular [Ca2+] induced by A23187. These results suggest that KRN4884 is a specific K+ channel opener and its vasodilating mechanisms involve not only deactivation of Ca2+ channels but also a decrease in the Ca2+ sensitivity of contractile elements.

    Notes: Abstract A MultiPhoton-Ionization Time-Of-Flight Mass Spectrometry (MPI/TOFMS) apparatus was developed for real-time measurement of the uranium isotopic ratio in nascent UF5 formed by the 266 nm photolysis of effusive UF6 ( 〈 300 K, ≈ 1.3 × 10−4 Pa). The UF5 was selectively and efficiently multiphoton ionized by 532 nm radiation at appreciably low fluences ( 〈 10 J/cm2). The main ions observed, U+ and U2+, were subsequently analyzed with a TOFMS with mass resolution of 1190 to separate235U n+ and238U n+ completely. The isotopic ratio measurements showed good precision resulting from the excellent agreement which was observed between the isotopic ratios in UF5 products and those in a parent UF6 sample. These results suggested that the MPI/TOFMS method can be applied to the real-time analysis of separation factors in the molecular laser isotope separation of uranium by ionization of UF5 following the infrared photodissociation of UF6.

    Notes: Abstract The time-dependent change in the concentration of UF6 monomers populated in the ground state was monitored in a supersonic Laval nozzle flow with an infrared diode-laser spectrometer in which the frequency of the laser beam was fixed at theυ = 1 ← 0 transition (627.7 cm−1) of theν 3 vibrational mode of the238UF6 monomer. The concentration of UF6 monomers in the ground state increased immediately after a single shot from a Raman laser tuned to a vibrational mode of UF6 clusters (614.8 cm−1) was applied to the gas in the nozzle. Subsequently, this concentration leveled off and slowly returned to the previous level. These results indicate that the population of UF6 monomers in the ground state increased as a result of the predissociation of UF6 clusters vibrationally excited with Raman laser radiation. It is demonstrated that one can utilize this procedure for vibrational predissociation of UF6 clusters as a technique to increase the concentration of UF6 monomers in an irradiation zone for molecular laser isotope separation of uranium.

    Notes: Abstract In infrared multiphoton dissociation of supercooled UF6 irradiated with multifrequency para-H2 Raman laser beams, separation factors and contrast ratios were measured by selective multiphoton ionization of the photoproduct UF5 at 532 nm followed by time-of-flight mass spectrometric analysis. The relationship between separation factor and contrast ratio is discussed theoretically and quantitatively investigated in the experiments. From this relationship, we obtain the irradiation conditions of the laser-beam fluences for attaining a high separation factor and estimate the value of the intrinsic separation factor in multifrequency dissociation of UF6.

    Notes: Abstract In the brain of the intact crayfish, three pairs of non-spiking giant interneurons (G1, G2, G3; NGIs) scarcely responded to substrate tilt about the longitudinal axis of the body either in the dark or in the presence of an overhead light. However, when the statolith was removed, these NGIs responded with depolarizing and hyperpolarizing potentials respectively to upward movements of the ipsilateral legs (2nd–5th pereiopods) and upward movements of the contralateral legs produced by substrate tilt. The relationships between the polarity of the potential and the direction of movement in the contralateral legs were opposite to those in the ipsilateral legs. The amplitude of the responses was proportional to the frequency (0.5-0.05 Hz) and amplitude of tilting. When the legs were moved unilaterally, the NGIs responded with depolarizing and hyperpolarizing potentials to upward movements of the ipsilateral legs and to upward movements of the contralateral legs, respectively. When the legs were moved bilaterally in the same direction by upward or downward movement of the substrate, the NGIs scarcely responded to the leg movements. A hypothetical model is presented to account for the pathways of sensory inputs to the NGIs and the role of NGIs in compensatory oculomotor system.

    Notes: Abstract Immunolocalization of matrix metalloproteinases (MMPs) and tissue inhibitors of matrix metalloproteinases (TIMPs) in periarticular tissues of β2-microglobulin amyloidosis patients was investigated. MMP-1 (interstitial collagenase) the most strongly expressed of the MMPs, was localized in the synovial lining cells, mesenchymal cells in granulation tissue and nodular amyloid deposits, and chondrocytes within areas of cartilage erosion. Expression of MMP-1 was correlated with the degree of macrophage infiltration and synovial cell hyperplasia, but it was not correlated with the degree of amyloid deposition or haemodialysis period. Expression of MMP-1 appeared more intense than that of TIMP-1 and TIMP-2 in highly inflammatory cases. MMP-2 was mildly expressed in the interstitial fibroblasts and MMP-3 was faintly stained in the extracellular matrix of the synovial membrane. MMP-9 (gelatinase B) was found to be strongly positive in the osteoclasts which increased in the progressing osteolytic lesion from the destructive arthropathy. These results suggest involvement of MMPs in inflammation with an imbalance between expression of MMPs and TIMPs being closely related to pathogenesis of the destructive arthropathy.

    Notes: Abstract. Background: To clarify the radiographic characteristics of intractable gastric ulcers with H2-receptor antagonists. Methods: The radiographic findings at the time of starting treatment were compared between 42 patients with gastric ulcers that did not heal within eight weeks of starting treatment with H2-receptor antagonists (the intractable group) and 58 patients whose ulcers healed within the eight-week period (the tractable group). Results: The following radiographic findings in the intractable group were observed at a significantly higher incidence than those in the tractable group and included: an ulcer located on the angle, linear ulcers, a greater depth, an uneven mound surrounding an ulcer, prominent folds' convergence, an overhanging gastric mucosa, an irregular ulcer base, a shortening of the lesser curvature and a U-shaped deformity of the angle. A multiple logistic regression analysis showed that the following three factors had a significant and independent delaying effect on healing: a U-shaped deformity of the angle, an uneven mound surrounding an ulcer and prominent folds' convergence. The relative risk of these factors were 12.7, 14.4 and 12.5, respectively. Conclusions: Intractable gastric ulcer with H2-receptor antagonists can be predicted based on the characteristic radiographic findings at the start of treatment.

    Notes: Summary The complete nucleotide sequence of the genome of a rakkyo strain of tobacco mosaic virus (TMV-R), which exhibits distinct host range differences from the common strain of TMV, was determined. The overall nucleotide sequence homology with TMV-U1 (a common strain of TMV) is 94.2%. The amino acid sequence homologies of the four encoded proteins (180K, 130K, 30K, coat protein) are from 95.9% to 98.0% compared with TMV-U1. To facilitate the analysis of the novel host range of TMV-R, a full-length clone of the genome containing a bacteriophage T7 RNA polymerase promoter was assembled from two cDNA clones and designated pRF3. In vitro transcripts derived from pRF3 were highly infectious. The infections of RF3, wild-type TMV-R, and U3/12-4 (derived from pU3/12-4, an infectious clone of TMV-U1) were compared onNicotiana tabacum cv. Bright Yellow (BY) plants. No systemic mosaic symptoms were observed on plants inoculated with RF3 and TMV-R, while BY plants inoculated with U3/12-4 developed distinct mosaic symptoms on the upper leaves 8–9 days post-inoculation. The green fluorescent protein (GFP) gene was introduced into pRF3 and pU3/12-4 by replacing the coat protein gene to get two GFP expressing chimeric virus clones: pR-GFP or pU1-GFP. Transcripts from pU1-GFP produced strong fluorescence when inoculated onto BY leaves, while those from pR-GFP produced only very faint fluorescence.
