Notizen: Calcium salts of high-oleic sunflower oil (〉 86% oleic acid) were prepared and fed at 0% and 7.5% of diet dry matter weight to 8 Holstein cows in early- to mid-lactation in a cross-over experimental design. Oleic acid content of milk fat increased from 26% to over 40% and cholesterol-raising saturates were reduced from 41% to 33%. Latin American white cheese, queso blanco, was produced from modified and normal milk. No differences in firmness were found between cheeses using penetration-compression testing with an Instron machine. Sensory difference tests indicated no significant differences between cheeses. Latin American white cheeses made with high oleic milk are similar to traditional cheese.

Notizen: Patients with renal disease often develop accelerated atherosclerosis resulting in more frequent cardiovascular complications compared to the general population. It has been suggested that peroxidation of low density lipoprotein (LDL) contributes to the pathogenesis of atherosclerosis and glomerulosclerosis. The present study aimed to investigate the phosphatidylcholine hydroperoxide (PC-OOH) content of LDL in patients with chronic renal failure (CRF) and patients on haemodialysis (HD). Plasma was obtained from healthy volunteers (n = 20; age 45.8 ± 16.9 years), CRF patients (n = 23; age 49.4 ± 18.1 years) and patients on HD (n = 19; age 59.1 ± 10.3 years). The lipid profile was studied in all patients. The LDL was isolated by sequential ultracentrifugation. The oxidized PC-OOH content of LDL was measured by high performance liquid chromatography with chemiluminescence detection (HPLC-CL). Compared to the controls, patients with proteinuria had significantly higher plasma concentrations for total cholesterol, LDL-cholesterol and apoB, whereas patients on HD had significantly higher levels of triglycerides and reduced HDL and apoA1 plasma concentrations. Significantly elevated levels of PC-OOH were found in HD patients (22.56 ± 13.40 pmol/mg LDL-cholesterol) and CRF patients (13.12 ± 15.15 pmol/mg LDL-cholesterol) compared to the controls (0.56 ± 0.38 pmol/mg LDL-cholesterol; P 〈 0.001). In HD patients the PC-OOH concentration was higher compared to CRF patients (P 〈 0.05). It was concluded that lipid hydroperoxides increase in renal disease and in patients on HD. These elevated lipid hydroperoxides may contribute to the increased risk for atherosclerosis in these patients.

Notizen: Abstract The prevalence and clinical and laboratory associations of IgM, IgG and IgA rheumatoid factors (RF) were determined in 352 patients with systemic lupus erythematosus (SLE). IgM, IgG, and IgA class RF were detected in 17.9%, 20.5%, and 20.5% of the sera, respectively. RF were associated with sicca syndrome, hypergammaglobulinemia, high titer of antinuclear antibodies, anemia, SSA- and SSB-antibodies, and with the presence of HLA-DR3. RF correlated negatively with nephritis and livedo racemosa. Moreover, we observed an association of RF and parameters of inflammatory activity such as elevated erythrocyte sedimentation rate (ESR) and leukopenia. Analysis of immunoglobulin classes revealed that laboratory parameters of inflammatory activity, SSA- and SSB-antibodies and HLA-DR3 correlated with IgA RF only. IgA RF define a subgroup of SLE patients characterized by distinct autoimmune phenomena and high disease activity in the absence of nephritis.

Notizen: Abstract The Wilms' tumor gene (WT1) encodes a transcriptional regulator involved in growth and differentiation of various tissue types. A continuous over-expression of WT1 was found in leukemic blasts, thus suggesting an oncogenic function. Solid cancer entities have also been described as expressing WT1. We systematically analyzed WT1 expression in small-cell and non-small-cell lung cancer, colon cancer and glioblastoma patients and in the respective tumor cell lines. Using reverse transcription/polymerase chain reaction, we found WT1 expression in glioblastoma (5 of 8), lung (5 of 11), and colon cancer (5 of 15) cell lines. While WT1 was expressed in only 1 of 12 lung cancer and 1 of 5 glioblastoma specimens, it was not detected in colon cancer or macroscopically tumor-free colon and lung tissue. In addition, HT29 colon cancer cells showed a loss of WT1 expression when grown to confluence or induced to differentiate by sodium butyrate. From this evidence, testing for WT1 expression is not clinically relevant for colon cancer, lung cancer, or glioblastoma patients. WT1 expression in cancer cell lines can probably be attributed to optimized in vitro growth conditions.

Beschreibung / Inhaltsverzeichnis: Abstract Background. In patients with diabetes mellitus an elevated level of plasma prorenin (PP) may be associated with proliferative diabetic retinopathy. Although retinopathy of prematurity (ROP) is also characterized by retinal vasoproliferation, no study on PP in ROP appears to have been carried out. This study investigated PP prospectively in preterm infants with high risk of ROP. Patients and methods. In 304 preterm infants (gestational age 24–36 weeks, mean ±SD 29.8±2.6 weeks; birth weight 570–1750 g, 1180±294 g) PP was examined prospectively between 3 and 14 weeks postnatally. Renin and total renin (after cryoactivation) were determined by radioimmunoassay. Total renin minus renin is the PP level; PP was correlated with the presence of ROP, stage of ROP, gestational age, birth weight, and postnatal age. Results. There was no significant difference between mean PP in 112 preterm infants with ROP (682±666 ng/l) and that in 192 preterm infants without ROP (622±454 ng/l). There was no correlation between PP and the stage of ROP, gestational age, or birth weight. Mean PP decreased with increasing postnatal age (postnatal age 3–4 weeks: 906±587 ng/l; 7–8 weeks: 585±423 ng/l; 13–14 weeks: 326±205 ng/l). Conclusion. This study found no significant difference in PP between preterm infants with and those without ROP. Thus PP is not a valid predictor or indicator of ROP. However, the study showed a hitherto unknown correlation between PP and postnatal age in preterm infants.
Notizen: Zusammenfassung Hintergrund.

Notizen: Abstract This review describes differing profiles of vascular risk factors in different types of dementia. Although vascular risk factors are related to various types of strokes, their independent effect on the occurrence of poststroke dementia appears to be small. Various risk factors have been identified for microangiopathy-related cerebral abnormalities, such as white matter changes and lacunae, which are the core lesions for the development of a vascular dementia syndrome without stroke symptoms. Most consistently, arterial hypertension and diabetes mellitus have been found to be associated with such brain abnormalities. Diastolic blood pressure seems to be of particular importance as recent investigations demonstrate that this factor is related to the course of multiple lacunar strokes and the progression of white matter disease. Epidemiological studies report that various vascular risk factors including arterial hypertension, diabetes mellitus, and atrial fibrillation may also be associated with Alzheimer’s disease. There is also evidence of a direct relationship between Alzheimer’s disease and general atherosclerosis. Further investigations are needed to determine whether these associations are due to the weakness of diagnostic criteria, or whether vascular risk factors indeed modulate the clinical expression of primary degenerative dementia. Common susceptibility genes leading to shared risk factors may be one of the reasons for a higher coincidence of Alzheimer’s disease and vascular dementia than can be expected by chance. A modulatory effect of vascular risk factors in the development of primary degenerative dementia may extend treatment options.

Notizen: Abstract. A general formalism of the so-called non-adiabatic quantum molecular dynamics is presented, which applies to atomic many-body systems in external laser fields. The theory treats the nuclear dynamics and electronic transitions simultaneously and self-consistently in a mixed classical-quantum approach. The equations of motion are derived from a general action principle by combining time-dependent density functional theory in basis-set-expansion with classical molecular dynamics. Structure and properties of the resulting equations of motion as well as the energy and momentum balance equations in this formalism are discussed in detail. For the model system H2 +, the classical-quantum equations of motion are solved. The results agree very well with that of full quantum mechanical calculations. Moreover, from the present calculation a transparent physical interpretation of the mechanisms of energy absorption and dissociation dynamics can be obtained. Future applications and approximations of the formalism presented here are briefly outlined.
