    Notes: The green fraction of humic acids (HAs), Pg, was fractionated by gel chromatography on Sephadex G-50. Repeated chromatography of the crude Pg obtained by the first chromatography of HA yielded a sharp peak (G1) and two broad bands (G2) of purified Pg. The recovery of Pg in G1 and G2 as percentages of the total Pg content of the HA were 16.2 and 14.5%, respectively. Distinct peaks were detected at 615, 570, 450 and 280 nm in the UV-visible spectra of both G1 and G2 in alkali solutions. Estimates of weight-average molecular weight were 9.7 * 103 for G1 and 1.23 * 104 for G2. In the 1H-NMR spectra the percentage of hydroxyl groups of 4,9-dihydroxy-perylene-3, 10-quinone (DHPQ) nuclei, which are known to be the chromophore of Pg, was 4.1 and 4.5% of the total H, respectively. Based on these values and the H/C ratios, DHPQ-C was estimated as 28% of total C. 13C-NMR spectra obtained using inverse-gated decoupling yielded 40–41% aromatic C, which suggested the presence of aromatic rings other than DHPQ. All of the IR, 13C- and 1H-NMR spectra indicated that the two purified Pg samples largely differed from each other in their contents of alkyl and polysaccharide components.

    Notes: The best pulverization of emulsions that can be achieved using conventional mixing blades is in the micrometer realm. Now, with our new ‘thin-film spin system’ high-speed mixer, it is possible to attain pulverization in the nanometer realm. The particle size distribution can now be controlled to achieve an almost single distribution state. Particles can be pulverized without being severed, preventing secondary agglomeration after processing. This new mixing system also solves many of the problems common to conventional pulverization processes.

    Notes: Abstract New neutron-deficient isotopes,197Rn,197mRn, and200Fr have been produced and identified on the basis of genetic correlations in the166Er(36Ar,5n)197Rn (Elab=186, 200 MeV), and169Tm(36Ar,5n)200Fr (Elab=186 MeV) reactions. The evaporation residues were separated from the beam by using a gas-filled recoil separator and implanted onto a position-sensitive solid-state detector. The α-decay energies (half-lives) of197Rn,197mRn and200Fr have been determined to be 7261±30 keV (51 −15 +35 ms), 7370±30 keV (18 −5 +9 ms), and 7500±3O keV, (570 −140 +270 ms), respectively.

    Notes: Abstract We present an unusual case of clear cell carcinoma arising from endometriosis in a 43-year-old woman. During laparotomy, a right adnexal tumor was found to be a sigmoidal subserosal tumor. Concomitant abnormal findings were endometriosis externa and enlarged ovaries. Intraoperative cytology suggested an adenocarcinoma; however, because of the unusual nature of the tumor and because the patient had not been fully informed about colostomy, further surgery was postponed. During the postoperative follow-up, the tumor grew and repeat aspiration cytology also suggested an adenocarcinoma. The patient again underwent laparotomy, and a Miles operation, total abdominal hysterectomy, bilateral salpingo-oophorectomy, regional lymphadenectomy and descending colostomy were performed. The patient completed two courses of multidrug chemotherapy (cisplatin, 70 mg/m2; epirubicin, 50 mg/m2; cyclophosphamide, 500 mg/m2) postoperatively, and she remains without recurrence 7 months after the second laparotomy.

    Notes: Abstract The matrix metalloproteinases (MMPs) MMP-2 and MMP-9 (gelatinases) have been suggested as serving an important role in cleaving the basement membrane structure. Tissue inhibitors of metalloproteinases TIMPs (particularly TIMP-1) are known to inhibit MMPs. Based on this background, we raised monoclonal antibodies against human gelatinase (MMP-9) and human recombinant TIMP (TIMP-1), and immunostained these two components in skin from patients with squamous cell carcinoma (SCC), Bowen’s disease (BD) and keratoacanthoma (KA). MMP-9 showed positive staining mainly in the granular layer of normal epidermis. In some cases of SCC and BD, MMP-9 showed positive staining in the dysplastic lesions even in the basal layer. TIMP showed a thorough positivity in normal epidermis. Unstained regions with this antibody were observed in SCC and BD. These results suggest that an altered staining pattern for MMP-9 and TIMP may be closely related to the malignant transformation of SCC and BD.

    Notes: Abstract We report lysinuric protein intolerance (LPI) and systemic lupus erythematosus (SLE) in a 14-year-old girl. The co-existence of LPI and SLE in the same patient has been reported before and suggests that SLE is directly connected with LPI. Conclusion Possibly in LPI the metabolic derangement can cause immunological abnormalities. Therefore, when multisystem involvement is observed in LPI patients, SLE should be considered as a rare complication.

    Notes: Summary.  We previously reported that infectivities of human and other nonhuman lentiviruses including human immunodeficiency virus type 1 (HIV-1) are activated by desialylation of the virion surface [H. Hu et al., J Virol 70: 7 462–7 470 (1996)]. The present study was designed to determine whether neuraminidase (NA) is useful for isolation of HIV-1 from patients. Peripheral blood mononuclear cells (PBMC) or CD4+ cells isolated from the PBMC of infected individuals were cocultured with PBMC or CD4+ cells from uninfected healthy donors, and the efficiency and frequency of virus isolation in the presence of NA were compared with those by a routine conventional procedure. In a total of 41 isolation trials from 28 individuals, the presence of NA markedly increased the frequency of isolation. Furthermore, both the day when virus was first detected and the day when the virus titer was the highest in the cultures were significantly earlier in the presence of NA than in the absence of NA. The deduced amino acid sequences of the V2 and V3 regions of gp120 were identical or very similar between the isolates obtained in the presence or absence of NA, suggesting that both isolation procedures selected a similar population. NA was thus found to facilitate HIV-1 isolation and its use is recommended particularly when isolation is negative by the conventional procedures.

    Notes: Summary.  The C-terminal cytoplasmic tail of chemokine receptors is important for their internalization upon ligand binding. We generated several deletion mutants of the C-terminal cytoplasmic tail of CXCR-4, a co-receptor for T cell line tropic strains of human immunodeficiency virus type 1 (HIV-1), to know whether or not co-receptor internalization is associated with HIV-1 entry. Our data showed that the removal of C-terminal 15 amino acid residues of the cytoplasmic tail from CXCR-4 completely abolished its internalization, but did not affect the co-receptor activity at all. Co-receptor activity was fully retained even when all 45 amino acid residues in the C-terminal cytoplasmic tail had been deleted. These data indicated that no cytoplasmic tail nor internalization of CXCR-4 is required for its co-receptor activity for HIV-1 entry.
