    Notizen: Wxa and Wxb are the most common alleles of the Wx gene in Asian cultivated rice. The difference between them is responsible for differences in the levels of the Wx gene product in the endosperm, as well as in amylose content, which is an important determinant of the quality of edible rice. Since Indica rice mostly carries the Wxa allele, we introduced the Wxb allele into Indica-type rice (IR36) from a Japonica-type rice (T65) by repeated backcrossing. In the near-isogenic line (IR36 Wxb), the level of the Wx gene product was reduced and, as expected, the level of amylose. However, IR36Wxb had a lower amylose content than the recurrent parent of T65 with the Wxb allele. The results suggest that the Indica (IR36) background might lower the amylose content more than the Japonica (T65) background when the Wx allele is the same. The possible importance of modifiers that regulate expression of the Wx gene is also discussed in relation to improvements in the grain quality of rice.

    Notizen: summary  The present study was designed to examine the hypothesis that the motor system controlling phonation is functionally coupled with the motor system controlling posture of the head and neck, as previously suggested (U. Haddar, T.J. Steiner, E.C. Grant and F.C. Rose Human Movement Science 2: 35 and Language and Speech 26: 117). Head tilt and trunk drift in the antero-posterior and right–left directions were measured simultaneously for 20 s before and during three types of speech sounds (/pa/, /ta/ and /ka/) as well during a maximum jaw open-close task in 14 healthy young subjects. The major findings obtained in all of the four tasks were: (i) the trunk drift measured in both directions was negligible in comparison with the head tilt; (ii) the head tilt in the antero-posterior direction was much larger than in the right–left direction and (iii) the head tilt during performance of the four tasks consisted of ‘initial’ and ‘sustained’ phases. In the initial phase, the head tilted posteriorly in association with the start of individual tasks. In the sustained phase, the head tilted either anteriorly or posteriorly when the task progressed. The magnitude of the net head tilt in the sustained phase negatively correlated with that of the initial head tilt. These findings indicate that phonation, in the form of jaw open-close, accompanies head movement in two consecutive phases.

    Notizen: Abstract Aims/hypothesis. Streptozotocin (STZ), a chemically reactive analogue of N-acetylglucosamine, induces necrosis of the beta cells, resulting in diabetes mellitus. Glucose-induced insulin resistance is mediated by increased activity of the hexosamine pathway. We aimed to examine the regulation of O-GlcNAc transferase expression and activity in the normal and streptozotocin diabetic pancreas.¶Methods. Rats were made diabetic by an injection of streptozotocin (65 mg/kg). The expression of O-GlcNAc transferase protein was examined by immunoblot analysis. Activity of O-GlcNAc transferase was assayed by the incorporation of [3H]GlcNAc into the synthetic peptide. Localization of O-GlcNAc transferase was done by immunohistochemistry. The change of O-GlcNAc modification of proteins was examined by immunoblot analysis.¶Results. In the STZ-induced diabetic pancreas, a severe loss of beta cells was observed, whereas alpha cells had increased in number. The diabetic pancreas showed an increase in the expression of O-GlcNAc transferase at the protein level and the O-GlcNAc transferase activity in it was increased significantly (p 〈 0.05). An increase in the immunostaining intensity in the cytoplasm of islet beta cells was also observed in the diabetic pancreas, whereas exocrine cells and islet cells other than beta cells showed little change in immunostaining intensity. The pancreas of STZ-diabetic rats showed a 3.1-fold increase in total cellular O-GlcNAc-modified proteins.¶Conclusion/interpretation. These findings indicate that O-GlcNAc transferase plays an important part in the modulation of O-GlcNAc concentrations in the pancreas and suggest that the increase in O-GlcNAc modification of the proteins correlates closely with diabetes.     Notizen: Abstract We report a 63-year-old man who had a rare bilateral thalamic glioma. He complained of difficulty with calculations and had mental deterioration. T1-weighted images revealed bilateral thalamic swelling with homogeneous low signal and no contrast enhancement. The tumour, showing decrease of N-acetylaspartate and the presence of lactate on magnetic resonance spectroscopy, was diagnosed as an astrocytoma by stereotactic biopsy.

    Notizen: Abstract  In almond, gametophytic self-incompatibility is controlled by a single multiallelic locus (S-locus). In styles, the products of S-alleles are ribonucleases, the S-RNases. Cultivated almond in California have four predominant S-alleles (S a, S b, S c, S d). We previously reported the cDNA cloning of three of these alleles, namely S b, S c and S d. In this paper we report the cloning and DNA sequence analysis of the S a allele. The Sa-RNase displays approximately 55% similarity at the amino-acid level with other almond S-RNases (Sb, Sc, and Sd) and this similarity was lower than that observed among the Sb, Sc and Sd-RNases. Using the cDNA sequence, a PCR-based identification system using genomic DNA was developed for each of the S-RNase alleles. Five almond cultivars with known self-incompatibility (SI) geno-types were analyzed. Common sequences among four S-alleles were used to create four primers, which, when used as sets, amplify DNA bands of unique size that corresponded to each of the four almond S-alleles; S a (602 bp), S b (1083 bp), S c (221 bp) and S d (343 bp). All PCR products obtained from genomic DNA isolated from the five almond cultivars were cloned and their DNA sequence obtained. The nucleotide sequence of these genomic DNA fragments matched the corresponding S-allele cDNA sequence in every case. The amplified products obtained for the S a- and S b-alleles were both longer than that expected for the coding region, revealing the presence of an intron of 84 bp in the S a-allele and 556 bp in the S b-allele. Both introns are present within the site of the hypervariable region common in S-RNases from the Rosaceae family and which may be important for S specificity. The exon portions of the genomic DNA sequences were completely consistent with the cDNA sequence of the corresponding S-allele. A useful application of these primers would be to identify the S-genotype of progeny in a breeding program, new varieties in an almond nursery, or new grower selections at the seedling stage.

    Notizen: Abstract Genes that encode products containing a NAC domain, such as NO APICAL MERISTEM (NAM) in petunia, CUP-SHAPED COTYLEDON2 (CUC2) and NAP in Arabidopsis thaliana, have crucial functions in plant development. We describe here molecular aspects of the OsNAC genes that encode proteins with NAC domains in rice (Oryza sativa L.). Sequence analysis revealed that the NAC genes in plants can be divided into several subfamilies, such as the NAM, ATAF, and OsNAC3 subfamilies. In rice, OsNAC1 and OsNAC2 are classified in the NAM subfamily, which includes NAM and CUC2, while OsNAC5 and OsNAC6 fall into the ATAF subfamily. In addition to the members of these subfamilies, the rice genome contains the NAC genes OsNAC3, OsNAC4 (both in the OsNAC3 subfamily), OsNAC7, and OsNAC8. These results and Southern analysis indicate that the OsNAC genes constitute a large gene family in the rice genome. Each OsNAC gene is expressed in a specific pattern in different organs, suggesting that this family has diverse and important roles in rice development.

    Notizen: Abstract. We have studied inclusive muon events using all the data collected by the TOPAZ detector at $\sqrt{s}=58$ GeV with an integrated luminosity of 273pb $^{-1}$ . From 1328 inclusive muon events, we measured the ratio $R_{q\bar{q}}$ of the cross section for $q\bar{q}$ production to the total hadronic cross section and forward-backward asymmetry $A_{FB}^{q}$ for b and c quarks. The obtained results are $R_{b\bar{b}}=0.13\pm0.02(stat)\pm0.01(syst)$ , $R_{c\bar c}=0.36\pm0.05(stat)\pm0.05(syst)$ , $A_{FB}^{b}=-0.20\pm0.16(stat)\pm0.01(syst)$ and $A_{FB}^ c=-0.17\pm0.14(stat)\pm0.02(syst)$ , in fair agreement with a prediction of the standard model.
