    Notes: In this study we evaluated antigen-specific in vitro responses of peripheral blood lymphocytes to lipopoiysaccharide (LPS)-depleted food allergens in children who reacted to food challenge (cow's milk or hen's egg) with a deterioration of their atopic dermatitis (AD). Some ofthe children showed immediate symptoms (urticaria, bronchial asthma or gastrointestinal symptoms) as well. The proliferation of casein-stimulated lymphocytes from children reacting to cow's milk (age (0.7–5.9 years) was significantly higher (P〈0.01) than the proliferation of lymphocytes from 15 children with AD without milk allergy (age: 2.1–9.1 years). Twenty-eight T-cell clones (TCC) were established from the blood of three children sensitized to cow's milk and hen's egg who reacted to double-blind, placebo-controlled oral food challenge both with a deterioration of AD and wilh immediate symptoms. Surprisingly, 16 of 28 casein- or ovalhumin-specific TCC were CD8+. All TCC produced high amounts of IFN-γ upon stimulation with concanavalin A. In addition. 75%. of the CD4+ TCC and 44% of the CD8+ TCC secreted IL-4. Our results indicate that: (i) food-specific proliferation of blood lymphocytes can be detected in patients with clinically relevant food allergy with LPS-depleted allergens in vitro and (ii) circulating food-specific lymphocytes are CD4+ and CD8+ T cells with the capacity of producing both type 1 and type 2 cytokines.

    Notes: Abstract The interaction of the cyclic decapeptide antibiotic gramicidin-S (GrS) with the nonionic detergent octaethylene glycol mono-n-dodecyl ether was studied by NMR spectroscopy. Detergent binding led to a slightly altered average conformation in the d-Phe side chains of the peptide. The changing diamagnetic shielding of nearby protons resulted in chemical shift variations, the largest effect being observed for the d-Phe C α proton. The continuous upfield shift of this proton resonance, indicating rapid exchange of the peptide between detergent-associated and unassociated states, was employed for an evaluation of the detergent/peptide aggregation equilibria. The nonlinear binding plot thus obtained was attributed to essentially different aggregational states, depending on the detergent/peptide ratio. The almost linear dependence of the spin-lattice relaxation rate and of the hydrogen-deuterium exchange rate on the fraction of detergent-associated GrS could be reconciled with a simple model, comprising binding of detergent monomers and cooperative binding of micelles at low and high detergent/peptide molar ratios, respectively. Thus, GrS provides a useful model for a study of backbone dynamics and water penetration in detergent- and membrane-bound peptides and proteins. The results will also be discussed with reference to the interaction of GrS with biological membranes.

Aufgrund fortlaufend durchgeführter chemisch-toxikologischer Untersuchungen konnten die Carbromalkonzentration im Serum und die eliminierten Carbromalmengen angegeben werden. Die Effektivität der einzelnen Eliminationsverfahren, insbesondere der forcierten Diurese und Peritonealdialyse, lie\ sich anhand der Aufzeichnungen näher untersuchen. Die Todesursache wurde von pathologisch-anatomischer und von chemischtoxikologischer Sicht besprochen.
    Notes: Summary The course of a lethal carbromal (bromdiethylacetylurea) poisoning is described. Continuous quantitation of carbromal levels in the serum and the amount excreted was performed. The effectiveness of several methods of elimination of the poison, in particular forced diuresis and peritoneal dialysis, could be compared. The cause of death is discussed from a pathological-anatomical and chemical-toxicological point of view.

    Notes: Abstract In a polluted site in Germany not only metallic, ionic and MeHg could be detected, but also organomercurials such as methyl- ethyl- and phenyl-mercury. In addition sometimes other organomercurials could be separated but up to now not identified. Extraction of the organomercurials from soil and percolating water was performed by dithizone, differentiation and detection by High-Performance-Liquid-Chromatography and atomic fluorescence detection, respectively. Differentiation between gaseous organic and gaseous elemental mercury was done by adsorption on Carbotrap® and gold filters, thermal desorption and detection by atomic fluorescence The behaviour of 8 organomercurials in soil is described. For this a lysimeter of a new design was filled with polluted soil. The top layer of the soil was spiked with 8 organomercurials. The percolating water and the air above the soil, were analyzed. Different layers of the soil were also investigated after the experiment. By synthetic rain the movement of Hg species is low: the compounds stay in the first centimeters. Ethoxyethylmercury+, tolylmercury+, and nitromersol were not detected in the percocolate. Concentrations of phenylmercury+ and hydroxymercurybenzoicacid decreased. Methoxymethylmercury+ increased. In the head of the lysimeter, volatile Hg(0) concentration increased during 150 hours by a factor of 5, while volatile organic mercury decreased during this time by a factor of 10. The organomercurial-content in soil decreased.A transformation of organic to inorganic Hg is therefore presumed.
