    Notes: Abstract The expression of human placental-type glutathione S-transferase (GST-π) was investigated in human cutaneous squamous cell carcinoma (SCC) and normal skin using Northern blot and immunohistochemical analysis. In Northern blot examination, the expression of GST-π transcript was recognized in all instances, and SCC showed a significantly higher expression of GST-π than normal skin. In immunohistochemical examination, GST-π was stained well in the cytoplasm of all cells of the stratum granulosum, many cells of the stratum spinosum and a few cells of the stratum basale in normal skin. Some cells of the stratum spinosum and almost all cells of the stratum basale showed only a weakly positive or almost negative reaction for GST-π. No nuclear staining of GST-π was obvious in normal epidermal cells. In SCC, many cells showed strong positivity for GST-π in the cytoplasm, and some were obviously accompanied by nuclear staining of GST-π. These findings suggest that GST-π exists mainly in many cells in the upper layers of the normal epidermis and that GST-π is involved in the process of carcinogenesis.

    Notes: Abstract The distribution of lipocortin I, a steroid-induced inhibitory protein of phospholipase A2, was examined in normal and psoriatic human skin. Using immunoblotting analysis with specific antibody against human lipocortin I purified from human placenta, lipocortin I was detected as a 37 kDa protein in cultured epidermal cells, whole skin and epidermis. In the dermis and stratum corneum, lipocortin I was only weakly detectable by Western blotting. In contrast to normal skin, much less lipocortin I was detected by Western blotting analysis in psoriatic skin. Using immunoperoxidase immunohistochemical analysis, lipocortin I was demonstrated in the cytoplasm of keratinocytes in the upper and middle layers of the epidermis and in some infiltrating cells in the dermis in normal skin. In involved psoriatic skin, by contrast, lipocortin I was almost undetectable in the epidermis, although it was demonstrated in some infiltrating cells in the dermis. No immunostaining of lipocortin I was observed in the stratum corneum of normal or psoriatic skin. These results, together with the finding that phospholipase A2 activity is higher in psoriatic epidermis than in normal epidermis, suggest that lipocortin I plays an important role in the regulation of differentiation and proliferation of epidermal keratinocytes.

    Notes: Summary Ling Zhi-8 (LZ-8), a novel and recently discovered immunomodulatory protein having in vivo immunosuppressive activity, was tested for in vivo effect against Type 1 (insulin-dependent) diabetes mellitus in the non-obese diabetic mouse, the disease having immunologically mediated aetiology in this animal. LZ-8 had mitogenic activity in vitro towards spleen cells of the non-obese diabetic mice as previously shown towards those of DBA/2 mice. Intraperitoneal administration of LZ-8 twice weekly into the mice (10.3–12.6 mg/kg body weight) from 4 weeks of age prevented insulitis and an almost normal number of insulin producing cells were observed. Extreme insulitis and reduction of the number of insulin producing cells were observed in the pancreata of the untreated non-obese diabetic mouse. No cumulative incidence of diabetes mellitus was observed in the LZ-8 treated group, while cumulative incidences of 70% and 60% were observed in an untreated group followed up to 42 weeks of age when the incidence of diabetes was defined as a plasma glucose level of greater than 11 mmol/l and as a urine glucose level of greater than 2 +, respectively. T cell subset population analysis was performed to further investigate the action of LZ-8 on the non-obese diabetic mouse which revealed that LZ-8 treatment increased in L3T4+/Lyt-2+ ratio.

These polymorphic sites also seem to be present in other Asian macaques. The present study of the polymorphism at theBamHI site 3′ to the β-globin gene in Asian macaques supports, at the nuclear DNA level, the idea that thefascicularis group includingM. fuscata, M. mulatta, M. cyclopis, andM. fascicularis is different from other Asian macaque groups.

    Notes: New resist systems based on acid-catalyzed, electrophilic aromatic substitution are described. These new resists show high sensitivity to deep UV and E-beam radiation with values approaching 2 mJ/cm2 and 2 μC/cm2, respectively. The resists are based on a three component system consisting of poly(4-hydroxystyrene), a polyfunctional, low molecular weight, latent electrophile, and a photoactive onium salt used as an acid generator. Irradiation of the resist film produces a latent image of acid dispersed in the matrix. During the postbaking step the photo-generated acid reacts with the latent polyfunctional electrophile and releases a reactive carbocationic intermediate with concomitant liberation of acetic acid. The carbocationic intermediate then reacts with neighboring phenolic moieties in a crosslinking reaction. The substitution reaction liberates a proton, making the process catalytic in nature, thus incorporating the concept of chemical amplification. These highly sensitive materials can be used as nonswelling negative multipurpose resists that function in deep-UV, x-ray or E-beam modes. © 1993 John Wiley & Sons, Inc.
