    Notes: Abstract Experiments that demonstrate quantitatively the importance of laser absorption dynamics for ultraviolet laser ablation of organic materials are presented. Laser pulse transmission measurements have been performed on 0.1 μm spin-coated polyimide films at three ultraviolet wavelengths (193 nm, 248 nm, and 355 nm) over the fluence range 10−3 −10 J/cm2. Target transmission is observed to increase with increasing fluence by a factor of ∼5 at 193 nm, and a factor of ∼10 at 248 nm. In contrast, transmission decreases by approximately one half during 355 nm target irradiation. These results are analyzed theoretically with a two-level model of chromophore absorption. This theory is also applied to reported pulsed UV-laser polyimide ablation data. It is shown that an accurate description of the fluence-dependent film absorption leads to a prediction of the etch depth versus pulse fluence relationship in good agreement with experimental data.

    Notes: Abstract We examined the abundance and spatial distribution of major phylogenetic groups of the domain Bacteria in hindguts of the Australian lower termite Mastotermes darwiniensis by using in situ hybridization with group-specific, fluorescently labeled, rRNA-targeted oligonucleotide probes. Between 32.0 ± 7.2% and 52.3 ± 8.2% of the DAPI-stained cells in different hindgut fractions were detected with probe EUB338, specific for members of the domain Bacteria. About 85% of the prokaryotic cells were associated with the flagellates of the thin-walled anterior region (P3a) and the thick wall of the posterior region (P3b/P4) of the hindgut, as shown by DAPI staining. At most, half of the EUB338-detected cells hybridized with one of the other probes that targeted a smaller assemblage within the bacterial domain. In most fractions, cells were found in varying numbers with probe ALF1b, which targeted members of the α-Proteobacteria, whereas substantial amounts of sulfate-reducing bacteria, gram-positive bacteria with a high DNA G+C content and members of the Cytophaga-Flavobacterium cluster of the Cytophaga-Flavobacterium-Bacteroides (CFB) phylum could be detected only in the wall fraction of P3b/P4. This clearly indicates that the hindgut microhabitats differ in the composition of their microbial community. In situ hybridization of cryosections through the hindgut showed only low numbers of bacteria attached to the P3a wall. In contrast, the wall of P3b was densely colonized by rod- and coccus-shaped bacteria, which could be assigned to the Cytophaga-Flavobacterium cluster of the CFB phylum and to the group of gram-positive bacteria with a high DNA G+C content, respectively. Oxygen concentration profiles determined with microelectrodes revealed steep oxygen gradients both in P3a and P3b. Oxygen was consumed within 100 μm below the gut surface, and anoxic conditions prevailed in the central portions of both gut regions, indicating that oxygen consumption in the hindgut does not depend on the presence of a biofilm on the hindgut wall.

    Notes: Abstract In situ hybridization with rRNA-targeted, fluorescent (Cy3-labeled) oligonucleotide probes was used to analyze bacterial community structure in ethanol- or paraformaldehyde-fixed bulk soil after homogenization of soil samples in 0.1% pyrophosphate by mild ultrasonic treatment. In ethanol-fixed samples 37 ± 7%, and in paraformaldehyde 41 ± 8% of the 4′, 6-diamidino-2-phenylindole(DAPI)-stained cells were detected with the bacterial probe Eub338. The yield could not be increased by enzymatic and/or chemical pretreatments known to enhance the permeability of bacterial cells for probes. However, during storage in ethanol for 7 months, the detectability of bacteria increased in both ethanol- and paraformaldehyde-fixed samples to up to 47 ± 8% due to an increase in the detection yield of members of the α-subdivision of Proteobacteria from 2 ± 1% to 10 ± 3%. Approximately half of the bacteria detected by probe Eub338 could be affiliated to major phylogenetic groups such as the α-, β-, γ-, and δ-subdivisions of Proteobacteria, gram-positive bacteria with a high G+C DNA content, bacteria of the Cytophaga-Flavobacterium cluster of the CFB phylum, and the planctomycetes. The analysis revealed that bacteria of the α- and δ-subdivision of Proteobacteria and the planctomycetes were predominant. Here, members of the α-subdivision of Proteobacteria accounted for approximately 10 ± 3% of DAPI-stained cells, which corresponded to 44 ± 16 × 108 cells (g soil, dry wt.)–1, while members of the δ-subdivision of Proteobacteria made up 4 ± 2% of DAPI-stained cells [17 ± 9 × 108 cells (g soil, dry wt.)–1]. A large population of bacteria in bulk soil was represented by the planctomycetes, which accounted for 7 ± 3% of DAPI-stained cells [32 ± 12 × 108 cells (g soil, dry wt.)–1]. The detection of planctomycetes in soil confirms previous reports on the occurrence of planctomycetes in soil and indicates a yet unknown ecological significance of this group, which to date has never been isolated from terrestrial environments.

    Description / Table of Contents: Summary Purpose: The aim of the study was to correlate delayed contrast enhancement of dysfunctional regions of the myocardium after injection of Gd-DTPA with the improvement of regional contractility 3 months after revascularization. Material and methods: Eleven patients with coronary artery disease and wall motion abnormalities underwent MR imaging before and 3 months after revascularization therapy (PTCA or CABG). Short-axis images were acquired using a cine gradient echo sequence. After revascularization, a representative slice was analyzed 14±1 min after injection of 0.1 mmol/kg Gd-DTPA using a T1-weighted turbo fast low-angle shot sequence. Improved systolic wall thickening 3 months after revascularization served as criterion of viability and was correlated with delayed contrast enhancement patterns. Results: After revascularization, 6 patients showed complete recovery of the dysfunctional area, 1 patient partial recovery, and 4 patients remained unchanged. All 4 patients with persisting wall motion abnormalities, 1/6 patients with recovery and the patient with partial recovery revealed a delayed enhancement. Conclusions: Three months after revascularization, delayed enhancement of dysfunctional myocardium is evident in patients without regional wall motion improvement, while the lack of delayed enhancement correlates with improvement of regional contractility.
    Notes: Zusammenfassung Zielsetzung: Die Korrelation der Signalintensität motilitätsgestörter Myokardareale in Spätaufnahmen nach der Gabe von Gd-DTPA mit der Erholung der regionalen Funktion nach Revaskularisierung sollte untersucht werden. Material und Methode: 11 Patienten mit Koronararterienstenosen und Wandbewegungsstörungen wurden mit einer CINE-GRE-Sequenz in der kurzen Herzachse vor und 3 Monate nach Revaskularisierung (PTCA oder Bypassoperation) untersucht. Bei der Kontrolle wurde eine repräsentative Schicht im motilitätsgestörten Areal 14±1 min nach der Gabe von 0,1 mmol/kg Gd-DTPA mit einer T1-w-TurboFLASH-Sequenz untersucht. Eine regionale Kontraktilitätsverbesserung wurde als vermehrte systolische Wanddickenzunahme nach Revaskularisierung definiert und mit der Signalintensität in Spätaufnahmen korreliert. Ergebnisse: Bei 6 Patienten erholte sich nach Revaskularisierung das gesamte motilitätsgestörte Areal, bei 1 Patienten partiell, bei 4 Patienten blieben die Wandbewegungsstörungen unverändert. Bei 4/4 Patienten mit persistierenden Wandbewegungsstörungen, bei 1/6 Patienten mit erholtem Myokard sowie bei dem Patienten mit dem partiell erholten Areal war ein spätes Enhancement nachweisbar. Schlussfolgerungen: Ein Enhancement von Myokardarealen in Spätaufnahmen nach der Gabe von Gd-DTPA 3 Monate nach der Revaskularisierung ist bei persistierenden Wandbegungsstörungen zu beobachten, wogegen das Fehlen eines späten Enhancements mit einer Verbesserung der regionalen Wandfunktion korreliert.

    Description / Table of Contents: Summary Purpose: Heart valve disease combined with left ventricular hypertrophy leads to derangements in cardiac energy metabolism, which can be detected non-invasively by 31P-MR-spectroscopy. The purpose of the present study was to examine whether the derangements in cardiac metabolism are reversible after surgical valve replacement. Patients and methods: 10 healthy volunteers and 10 patients with aortic stenosis (pressure gradients 〉60 mmHg) were included. For assessment of energy metabolism, 31P-MR spectra were obtained with a double oblique 3D-CSI technique (voxel size 25 cm3). In 5 of 10 patients, follow-up examination was performed 3 months after surgical valve replacement (SVR). Left ventricular (LV) function was analyzed by cine MRI. Results: Before SVR the myocardial phosphocreatine to adenosinetriphosphate (PCr-ATP) ratio was significantly (p=0.0002) reduced to 0.80±0.25 in patients compared to 1.65±0.21 in volunteers. 3 months after SVR, LV mass had significantly (p=0.04) decreased from 238±33 g to 206±47 g. At the same time a significant (p=0.04) increase of the PCr-ATP ratio from 0.80±0.25 to 1.28±0.22 was observed. A slight, but not significant, reduction of the phosphodiester ATP ratio was observed before SVR, with a trend towards normalization after SVR. Conclusions: After SVR, the deranged energy metabolism shows a trend towards normalization. Further follow-up is necessary to determine whether complete normalization of the energetic derangement can be observed over longer periods of time following SVR.
    Notes: Zusammenfassung Zielsetzung: Mit einer linksventrikulären Hypertrophie einhergehende Herzvitien führen zu Veränderungen des kardialen Energiestoffwechsels, die mit der 31P-MR-Spektroskopie nichtinvasiv erfasst werden können. Ziel der vorliegenden Studie war es, zu untersuchen, ob nach chirurgischem Klappenersatz eine Normalisierung des Energiestoffwechsels beobachtet werden kann. Patienten und Methode: 10 gesunde Probanden und 10 Patienten mit Aortenklappenstenose (Druckgradient 〉60 mmHg) wurden untersucht. Zur Erfassung des kardialen Energiestoffwechsels wurde eine doppelt angulierbare 3D-CSI-Technik verwendet (Voxelgröße 25 ccm). 3 Monate nach Aortenklappenersatz (AKE) erfolgte bei 5 Patienten eine Nachkontrolle. Bei den Patienten wurden zusätzlich linksventrikuläre (LV) Funktionsparameter mittels cine-MRI-Technik analysiert. Ergebnisse: Vor Klappenersatz war bei den Patienten das Phosphokreatin-Adenosintriphosphat(PCr-ATP)-Verhältnis mit 0,80±0,25 signifikant im Vergleich zu Gesunden (1,65±0,21; p=0,0002) erniedrigt. 3 Monate nach AKE konnte eine signifikante (p=0,04) Verringerung der LV-Masse von 238±33 g auf 206±47 g nachgewiesen werden. Die LV-Ejektionsfraktion änderte sich jedoch nicht wesentlich (von 61±11% auf 65±7%; p=0,22). Zur selben Zeit zeigte sich ein signifikanter (p=0,04) Anstieg des PCr-ATP-Verhältnis von 0,80±0,25 auf 1,28±0,22 an. Für das Phosphodiester-ATP-Verhältnis zeigte sich eine geringe, jedoch nicht signifikante Reduzierung vor AKE, mit einer Tendenz zum Wiederanstieg nach AKE. Schlussfolgerungen: Nach Klappenersatz ist eine Tendenz zur Normalisierung des pathologisch veränderten Energiestoffwechsels zu beobachten. Weitere Nachkontrollen sind notwendig, um zu klären, ob es zu einem späteren Zeitpunkt zu einer vollständigen Normalisierung des Energiestoffwechsels kommt.
