    Notes: Abstract To determine how and when the satellite cells are stimulated to replicate in muscle regeneration, the rat soleus muscle was examined chronologically after bupivacaine-induced myonecrosis. Bromodeoxyuridine and desmin-positive mononuclear cells, indicating the start of satellite cell replication, were seen 25 h after bupivacaine treatment when macrophages had already invaded the sarcoplasm of necrotic fiber. These findings suggest that muscle regeneration starts as early as the time at which macrophages begin to scavenge necrotic material. Proliferating myoblasts increased in number, reaching a maximum at 49 h after myonecrosis, and decreased in number 3 days after the myoblasts fused with each other to form myotubes. The satellite cell proliferation after bupivacaine-induced myonecrosis began at almost the same time as in crush injury, and earlier than after muscle transplantation using whole intact or minced muscle fragments. The earlier begining and more rapid regenerating process probably resulted from the preservation of intact satellite cells, blood vessels and peripheral nerves in the bupivacaine-induced myonecrosis.

    Notes: Abstract The mechanism of regulation of intracellular pH (pHi) in dispersed acini from the rat mandibular salivary gland has been studied with a microfluorimetric imaging method and the pH probe 2′,7′-bis(2-carboxyethyl)-5(and −6)-carboxyfluorescein. The pHi in the TRIS/HEPES-buffered standard solution was 7.29±0.01. Addition of 1 μmol/l acetylcholine (ACh) or ionomycin caused a sustained increase in the pHi. These agents decreased pHi in the absence of external Na+ or in the presence of amiloride. The rate of pHi recovery from an acid load after NH 4 + prepulse was a linear function of pHi and increased as pHi became more acidic. Addition of ACh shifted the relationship towards a more alkaline pHi range. The increase in pHi induced by ACh or ionomycin was not inhibited by the protein kinase C inhibitors staurosporine (10 nM) and 1-(5-isoquinolinesulfonyl)-1-methylpiperazine (50 μmol/l). Addition of 0.1–1 μmol/l phorbol 12-myristate 13-acetate (TPA) had little effect on pHi within 10 min; however, exposure to TPA for 120 min resulted in a significant rise in pHi. In Ca2+-free solution with 50 μmol/l 8-(diethylamino)-octyl-3,4,5-trimethoxybenzoate, the ACh-induced rise in both pHi and cytosolic Ca2+ concentration was suppressed. ACh and ionomycin caused an increment of amiloride-sensitive acid output into the extracellular fluid, while 20 μmol/l 1-oleoyl-2-acetylglycerol had little effect on it. It was concluded that (a) stimulation with ACh activated the Na+/H+ antiport in the plasma membrane, (b) ACh also stimulated the intracellular acid production but acid extrusion by the Na+/H+ antiport prevented the cell from intracellular acidification, and (c) the major route of signal transduction for the ACh-induced activation of the Na+/H+ antiport was independent of protein kinase C but was dependent on the rise in cytosolic Ca2+ concentration. The implication of the cytosolic acidification and cell volume change in pHi regulation is discussed.

    Notes: Abstract Using double-barreled Cl−-sensitive microelectrodes, intracellular Cl− activity (A Cl i ) in the mouse lacrimal acinar cells in vitro was determined in both resting and secretory phases. In the resting stateA Cl i was 31 mmol/l which was 1.4 times higher than that predicted for the passive distribution according to the membrane potential (V m) of −41 mV. Addition of acetylcholine (ACh, 1μM) hyperpolarizedV m to −63 mV and decreasedA Cl i to 20 mmol/l which was still twice the equilibrium activity. A-23178 produced similar changes inV m andA Cl i to those induced by ACh. It was concluded that Cl− was actively accumulated in the acinar cells and, in the secretory phase, Cl− efflux was enhanced by the increased driving force and Ca2+-mediated increase in the Cl− permeability across the cell membrane.

    Notes: Summary The effect of column temperature, especially at low temperatures, on the separation of fullerenes on monomeric and polymeric octadecyl silica (ODS) bonded phases has been studied. Decreasing the column temperature induces an increase in selectivity. The best temperature for the separation of fullerenes was determined for both types of ODS phase with n-hexane eluent. The selectivity for higher fullerenes on monomeric phases becomes similar to that on polymeric phases to low temperature. It has been found that as the carbon content of monomeric phases is increased, the selectivity also becomes similar to polymeric phases.

The abundance spectra of alkali metal clusters showed distinct maxima and steps atn=3, 7, 9, 13 and 19 for Li, and atn=3, 5, 11, 13 and 19 for Na. Mass spectra of Si, Ge and Sn clusters were very similar each other, showing intensity drops aftern=4 and 6 (and alson=10 for Ge) for singly charged clusters. The magic numbers observed are discussed in terms of stability of charged clusters.

    Notes: Abstract Sodium cluster ions Na+ n withn ranging up to 25 have been observed from a liquid sodium ion source by using a magnetic mass analyzer. Ion intensity as a function of cluster size showed distinct steps and local maxima atn=3, 5, 11, 13 and 19 (magic numbers), and a pronounced odd-even alternation. The features in the ion abundance curve are attributed to the relative stability of cluster ions. The observed magic numbers are only partially explained by the electronic shell model, indicating need to include a consideration of atomic structure in a cluster.

    Notes: Abstract Cluster ions of alloys (Li-Na, Li-Mg) have been produced by a liquid metal ion source (LMIS), and analyzed by mass spectrometry. For the Li-Na system, bimetallic clusters with various compositions were formed, and dominant bimetallic species were Na2Li+, NaLi+, NaLi 2 + and NaLi 8 + with this sequence of ion intensity. These clusters are systems containing 2 or 8 valence electrons except for NaLi+. For the Li-Mg, observed bimetallic clusters were limited to only three species (MgLi+, MgLi 2 + and Mg2Li+), but unexpectedly small multiply charged homonuclear clusters, Mg 2 2+ and Mg 3 2+ , were observed.

    Notes: Summary The temperature effect on the retention of fullerenes in the range 80°C to −70°C in liquid chromatography (LC) has been examined using octadecylsilica stationary phases (ODS). It has been found that the maximum retention temperature lies around −10°C with a highly carbon loaded ODS phase. Solid state CPMAS NMR measurements on the stationary phases indicated that the relaxation time at the 30 ppm methylene signal changes with the temperature and has a minimum relaxation time at the temperature which closely matches the maximum retention temperature observed in chromatography. The interpretation of both NMR spectroscopic and LC chromatographic data are discussed.
