    Notes: Abstract: A cDNA (M41) corresponding to a mouse myelin basic protein (MBP) mRNA with a longer 5′-untranslated region than predicted from earlier studies of MBP gene structure has been isolated and characterized. The additional 5′-untranslated region is encoded by two previously unidentified exons upstream of the major transcription start site of the gene. Using a DNA probe specific for M41-MBP mRNAs, Northern blot analysis indicated that expression of this transcript follows a developmental course in mouse brain similar to that of the majority of MBP mRNAs, but that the level of expression varies between brain and spinal cord. Expression of MBP mRNAs similar to the mouse M41-MBP also was identified in rat brain. The results suggest that the structure of the MBP gene is more complex than originally thought, containing at least two more exons. There appears to be at least one more MBP gene promoter that directs the synthesis of a subset of MBP mRNAs with a unique 5′-untranslated region.

    Notes: Garnet films of nominal composition (Y,Nd)3Ga5O12, were grown on (110) 1°-off Gd3Ga5O12 substrates for investigation of their growth-induced optical anisotropy. Optical birefringence and directions of the electric vectors of polarized rays passing through the films were measured under a polarizing microscope using a Brace–Köhler compensator. The growth-induced anisotropy of these films optically exhibited orthorhombic characteristics with the X, Y, and Z optic elasticity axes coinciding with the [001], [110], and [1¯10] directions, respectively. The crystallographic data obtained by means of single-crystal diffractometry suggested that the cubic crystal system of the garnet film was distorted, though very slightly, to an orthorhombic one with a,b, and c axes that coincided, respectively, with the [1¯10],[001], and [110] of the original cubic cell. In addition, by annealing at 1150 °C, this distortion disappeared and the crystal system reverted to cubic.

    Notes: LiNbO3 single crystals with a composition close to stoichiometry are grown by using a double crucible Czochralski method equipped with an automatic powder supply system. The photorefractive properties are compared with those of LiNbO3 crystals grown from the congruent composition melt. Two beam coupling experiments show that both nondoped and Fe-doped stoichiometric crystals exhibit stronger photorefractive effect than the Fe-doped congruent crystals.© 1997 American Institute of Physics.

    Notes: Photorefractive damage, photoconductivities, and photogalvanic currents of LiNbO3 single crystals have been investigated as a function of [Li]/[Nb] and MgO concentrations. Near stoichiometric LiNbO3 single crystal, having large [Li]/[Nb] ratios of 0.993, shows lower photorefractive damage resistance than congruent LiNbO3. However, crystals doped with a small amount of MgO (〉0.78 mol %) exhibit no measurable photorefractive damage at 532 nm to intensities of as much as 2 MW/cm2. This remarkable decrease of photorefraction that was found can be attributed to the decrease of saturated space charge field results from a combination of the increased photoconductivity and the decreased photogalvanic current of the crystal. © 2000 American Institute of Physics.

    Notes: Abstract— Developmental changes of myelin proteins in chick sciatic nerve were studied at the stage of myelination by sodium dodecylsulfate (SDS)-polyacrylamide gel electrophoresis. The myelin of adult hen peripheral nervous system (PNS) contained two glycoproteins (BR-P0 and PASII), both of which are unique to PNS myelin, in addition to the basic encephalitogenic protein, BP, which is common to CNS and PNS myelin. The other basic protein (BF-P2) found in the PNS of other species was not definitely detectable in hen PNS. At the early stages of myelination (from 14 to 18 embryonic days) the amounts of myelin proteins increased rapidly in parallel with the increase in number of layers of the myelin sheath of the PNS. At 14 embryonic days high molecular weight proteins were dominant, while myelin specific proteins were barely detectable in the PNS myelin fraction. At 18 embryonic days, however, BR-PO, BP and PASII proteins became the main protein components of the PNS myelin, whereas high molecular weight proteins decreased in quantitative importance during development. At the early stage of myelination other glycoproteins were also detectable in the PNS myelin. Radioactive fucose was actively incorporated into the two glycoproteins, BR-P0 and PASII, at the early stage of myelination in vivo. These results suggested that myelin proteins especially glycoproteins, may play an important role in PNS myelin formation.

    Notes: Abstract— 〈list xml:id="l1" style="custom"〉1Chromatin protein fractions were separated from the nuclei from brain, liver and kidney of the guinea pig. The fractions were studied by electrophoretic methods and amino acid analysis.2Brain nuclear fractions were washed with 0.15m-NaCl and nuclear acidic proteins then removed by 0.35 m-NaCl. These 0.35 m-NaCl-extracted proteins were considered to be similar to the nuclear soluble acidic proteins.3Nonhistone-1, histone and nonhistone-2 fractions were obtained from 2.0 m-NaCl-soluble chromatin fractions by lowering the salt concentration and successive extraction with acid and alkali. The nonhistone-3 fraction was also extracted from the nuclear residue by alkaline solution.4The contents and characteristics of the nonhistone fractions of the brain, especially the nonhistone-1 fraction, differed among the three tissues. The histone fractions showed no obvious difference among the three tissues. The nonhistone-1 fraction of the brain, which comprised a low percentage of total nuclear protein, contained relatively high amounts of acidic proteins.

    Notes: Abstract— Antiserum to BF protein isolated from bovine spinal roots has been used to study the distribution of the protein in other species and tissues.Significant amounts of protein could be demonstrated in bovine, pig and rabbit peripheral nerve myelin. It was, however, scarcely detectable in guinea pig peripheral nerve myelin. There was BF protein in rabbit spinal cord as well as in peripheral nerve, but little or no BF protein in the liver, kidney, muscle or brain. BF protein in bovine spinal cord was localized in the myelin. The ratio of the BF protein to the encephalitogenic protein in the spinal cord myelin was around 0.15:1.0. BF protein was extractable from peripheral nerve myelin by saline as well as by acid solutions.The circular dichroism spectrum of the BF protein in aqueous solution suggested that this protein contained a very large amount of β-structure. This structure was not considered to be the result of acid denaturation because the protein purified from the saline extract of peripheral nerve also showed a similar spectrum.

    Notes: Platelet-activating factor (PAF). as wel as PAF acetylhydrolase (PAF-AH) activity in the peripheral blood plasma of patients with psoriasis and palmoplantar pustolosis, was measured with a radio-immunoassay technique, and compared with leukotriene (LT) B4. LTC4. LTD4, and E4, (LTD4/E4). thromboxane (TX) B2 and prostaglandin (PG) E2 levels. In a normal healthy group (n= 1 2) PAF level was 25.9 ± 6.5 pg/0.1 ml plasma (mean ± standard error of the mean: SEM). and this was elevated in patients with psoriasis (68.1 ± 11.8, n= 25. P〈0.01), without a change in the PAF-AH level. LTB4 showed a similar increase (115.0 ± 21.6 pg/ml vs. 68.2 ± 11.8 pg/ml. P 〈 0.05), while TXB2, and PGE2 showed insignificant (P 〉 0.05) changes. LTC4 and LTE4/E4 were around the level of the limit of detection. Patients with palmoplantar pustulosis (n= 33) demonstrated similar, but milder and statistically insignificant, increases in PAF. LTB4. TXB2 and PGF2 levels. Modulation of the mediator levels before and after treatment was compared in 16 patients with psoriasis and 11 with palmoplantar pustulosis. PAF in psoriasis significantly decreased after treatment (70.9 ± 17.1 to 25.1 ± 5.5, P 〈 0.05) and this was moderately correlated (r= 0.298) with clinical improvement as indicated by the psoriasis area and severity index (38.5 ± 7.5 to 10.9 ± 4.2. P 〈 0.01). TXB2. (180.2 + 100.4 to 34.1 ± 13.5). PGF2 (3.7 ± 0.7 to 2.9 ± 0.5) and LTB4 (120.1 + 31.1 to 84.2 + 8.2). in psoriasis, mildly decreased without statistical significance. Patients with palmoplantar pustulosis demonstrated a similar decrease in all mediators without statistical significance. The results obtained suggest a role of PAF in psoriasis. As the priming effects of PAF have been shown, for leucocytes and endothelial cells, to enhance their inflammatory response, we assume that PAF has roles in the acute phase of osoriatic and leucotactic inflammation.
