    Description / Table of Contents: Zusammenfassung Zur Bestimmung von Haselnußprotein in Haselnüssen und verschiedenen Schokoladenwaren wie Schokolade, Haselnußmus und Nuß-Nougat-Creme wurde eine Elektro-Immunobestimmung nach Laurell ausgearbeitet. Dabei wurde ein optimales Entfettungs- und Extraktionsverfahren ermittelt, in dem die Eignung verschiedener Lösungsmittel und Puffersysteme geprüft wurde. Als schonendstes Entfettungsmittel haben sich Aceton und als beste Puffer Tris-Tricine und Tris-Borat erwiesen. Harnstoff setzt die Ausbeute an intaktem Protein je nach Molarität bis auf 15% herab. Die Eichkurve des isolierten Standards (Corylin) umfaßt bei sparsamster Verwendung des Antiserums einen Bereich von 0,01–0,1 %, mit jeweils einem statistisch ermittelten Fehler von ±6,8% (3-s-Wert); bei Handelsprodukten liegt er je nach Art zwischen ±7% bis ±9%. Die Wiederfindungsrate lag bei allen Produkten über 95% ± 8%. Die Identität der gefundenen Proteine gegenüber dem Standard konnte mit der Geldiffusion nach Ouchterlony und mit einer modifizierten Laurell-Technik gesichert werden. Die erhaltenen Ausbeuten an intaktem Protein gehen mit steigendem Röstgrad zurück. Doch selbst bei einer Rösttemperatur von 150 °C und 30 min läßt sich noch intaktes Haselnuß-protein nachweisen.
    Notes: Summary An electro-immuno-assay was worked out for the determination of hazelnut protein in some products like chocolate, hazelnut purée and nut-nougat-cremes. During the investigations, an optimal procedure for defatting and extraction was identified. The most suitable defatting agent for nut cremes was acetone, the most suitable buffer combination tris-tricine resp. tris-borate. Urea depending on its molarity, decreased the amounts of protein in the extracts, down to 15% of the original value. The standard curve of the isolated hazelnut protein (corylin) when using the lowest amount of antiserum 0.1 ml per 15 g agarose per plate extents from 0.01–0.1%. The rate of error (3 s,n=10, double estimation) was found to be ±6.8%, for products of commerce ±7–9%. The recovery in all products was greater than 95% ± 8%. The identity of proteins as compared with the standard protein was evaluated by the double-gel diffusion method of Ouchterlony and the modified electro-immuno-assay of Laurell. With increasing roast temperatures, the values of intact protein decrease. These results will be shown in the subsequent paper.

    Description / Table of Contents: Zusammenfassung Mit Hilfe der trägerfreien Elektrophorese nach Hannig gelingt es, zuvor nicht antigene Maisproteine so schonend zu isolieren und anzureichern, daß eine erfolgreiche Immunisierung durchge führt werden kann. Es wird dabei angenommen, daß es zu einer Abtrennung von die Immunisierung störenden Stoffen kommt. Mit den erhaltenen Antisera lassen sich bei Bier verwendete Rohfruchtproteine in immunologischen Methoden nachweisen.
    Notes: Summary By means of free flow electrophoresis according to Hannig it is possible to isolate and enrich previously not antigenic proteins ofZea mays in a very careful manner, so that afterwards a successful immunisation can be carried out. It is assumed that the separation procedure of the electrophoresis eliminates some interfering substances from the protein extract e.g. carbohydrates. With the thus obtained antiserum added raw fruit proteins from corn in beer sud could be detected in the final product by immunological method.

    Description / Table of Contents: Zusammenfassung Ein Zusatz von Kuhmilch zu Schaf-und/oder Ziegenmilch bzw. -käse kann nach Carbamylierung der Proteine durch Gegenstromelektrophorese auf Agarosegelen im Bereich von 0,1 bis 1 % als untere Grenze bei Milch bzw. von 0,1 bis 2% Kuhmilchkäse als untere Grenze bei Käse nachgewiesen werden. Hierzu wird ein spezifisches Antiserum gegen Rinder-Immunglobulin vom Typ G eingesetzt. Nur bei einer wesentlichen Überschreitung der üblichen Pasteurisationsbedingungen für Milch oder bei sehr starker Proteolyse während der Käsereifung kann ein sicherer Nachweis nicht gewährleistet werden.
    Notes: Summary The addition of cow milk to sheep or goat milk respectively — sheep or goat cheese can upon carbamylation of the proteins, be detected by counter current electrophoresis on agarose gel. Lower limits of detection are 0.1–1% in milk and 0.1–2% in cheese. To accomplish this an antiserum specific to bovine immunoglobulin type G is used. Only, when the usual pasteurisation conditions for milk have been substantially exceeded, or upon very strong proteolysis during ripening of the cheese, certain detection cannot be guaranteed.

    Notes: Abstract An antiserum was prepared for the determination of glibenclamide and for the estimation of other commercially available sulfonylureas. Rabbits were immunized with a glibenclamide-BSA conjugate. Tritiated glibenclamide was used as the tracer. The assay was performed in the presence of 8-anilinonaphthalenesulfonic acid to displace glibenclamide bound to serum protein, and free and antibody bound tracer were separated by dextran-coated charcoal. For glibenclamide determination in serum and plasma the limit of detection was 3 ng/ml. Sensitivity calculated for the whole determination range was 102 cpm for a 10 % concentration difference. Specificity studies showed a cross-reaction of less than 0.1 % for glibenclamide metabolite M1 and 9 % for metabolite M2. Other sulfonylurea drugs display cross-reactivities from 0.1% (chlorpropamide) to 190% (gliquidone). Both intra-assay and inter-assay imprecision were below 10 %. Accuracy was established by comparison of the present method with HPLC. The assay was applied to the specific determination of glibenclamide in clinical trials and for diagnosing factitious hypoglycemia caused by sulfonylureas.

    Notes: The identification of turkey meat in food products has been accomplished using specific antisera against turkey troponin T. This protein (Mr 37 000) was extracted from fresh muscle tissue and isolated by a procedure that included free-flow electrophoresis as final fractionation step. Preparations of troponin T with purities of more than 95 % were applied as antigen in the immunization of rabbits. Several standard immunological techniques were used to determine additions of turkey meat in food products heated to different temperatures (including roasting temperature). That way, it was possible to also detect additions of chicken meat on the basis of a partial identity between troponins T from turkey and chicken muscle.
