    Notes: Abstract. Objective: To investigate the effects of adenosine receptor agonists and antagonists on 5-HT release from rat isolated pleural mast cells and on plasma protein extravasation in the skin of conscious rats.¶In vitro Methods: Rat isolated pleural mast cells were loaded with [14C] 5-HT, sensitised with mouse monoclonal anti-DNP and then challenged with human serum albumin-DNP. DNP-stimulated 5-HT release from mast cells was determined.¶In vivo Methods: Rats, loaded intravenously with [125I] human serum albumin, were injected intradermally with adenosine agonists at sites on the back. 30 min later plasma protein extravasation at each injection site was determined.¶Results: In isolated mast cells, each adenosine agonist enhanced DNP-induced 5-HT release, N6-(3-iodobenzyl)-5-(N-methyl-carboxamidoadenosine), (IB-MECA), being the most potent agonist. The adenosine A1/A2 antagonist, 8-phenyltheophylline (8-PT), had no effect on the response to IB-MECA. In contrast, 3-(4-amino-iodobenzyl)-8-[4-[[[carboxy]methyl]oxy]phenyl]-1-propylxanthine, (I-ABOPX), inhibited (pA2 6.2) the IB-MECA responses. In the skin of conscious rats, intradermal IB-MECA produced a marked plasma protein extravasation (PPE) which was mimicked by N6-2-(4-aminophenyl)-ethyladenosine (APNEA). The PPE produced by IB-MECA was not affected by either 8-PT or CGS15943A, but was virtually abolished by cyproheptadine and in rats pre-treated with Compound 48/80.¶Conclusions: These results indicate that stimulation of adenosine A3 receptors both enhances degranulation in vitro and directly produces degranulation of rat mast cells in vivo.

    Description / Table of Contents: Zusammenfassung Oft wird eine scheinbare Wärmeleitfähigkeit für inhomogene Materialien verwendet. In diesem Artikel wird gezeigt, daß es im allgemeinen nicht genügt, die instationäre Übergangswärmeleitung mit der stationären Wärmeleitfähigkeit zu beschreiben. In unserer Geometrie gibt dies eine Überschätzung der Wärmeleitung. Ein numerisches Modell für die Wärmediffusions-gleichung ist entwickelt worden, um mehrere Schätzungen der scheinbaren Wärmeleitfähigkeit zu kontrollieren. Für kurze und lange Zeiten stehen unterschiedliche analytische Beziehungen zur Verfügung.
    Notes: Abstract An apparent thermal conductivity for inhomogeneous materials is widely used. In this paper it is demonstrated that the apparent thermal conductivity for stationary heat conduction is not sufficient to describe the transient heat response of an inhomogeneous medium. In the geometry we used the heat transfer is estimated too high when the stationary thermal conductivity is employed. A numerical solution of the equation of thermal diffusion has been used to check several approximations. For short and for long times a separate approximate analytic expression can be used.

    Notes: Abstract We studied motor-unit recruitment and decruitment thresholds in the three heads of the human elbow extensor, the triceps brachii muscle (caput mediale and laterale, both mono-articular heads, and caput longum, the bi-articular head) by means of intramuscular electromyographic-recordings. Two experiments were performed: an ‘isometric’ and a ‘movement’ experiment. In the isometric experiment, subjects were asked to increase the elbow extension torque isometrically to a specific level, keep the torque at the level for 10 s, and then decrease the torque again to zero. In the movement experimental subjects moved their forearm from 90° to 110° extension against an increasing flexion torque, kept the latter position for 10 s and then moved their forearm back while the torque decreased. Results for caput longum showed that recruitment thresholds were higher than decruitment thresholds, whereas in caput mediale and laterale no difference in thresholds was found. In caput longum recruitment thresholds were found to be lower in movement conditions than in isometric conditions. The reverse effect was observed in caput mediale, whereas no difference in recruitment thresholds was found in caput laterale. Our results point to a transfer of force from mono-articular muscles in isometric conditions to bi-articular muscles in movement conditions. A similar transfer is found when recruitment and decruitment are compared. This means that the transfer is not only a property of the elbow-flexor muscles, but is a more common trait. A qualitative analysis of firing frequencies at recruitment and at decruitment in both conditions supports our findings.

    Notes: Systematic electron diffraction studies on intermetallic precipitates formed within a lightweight Mg–RE–Zn alloy (RE = La or Ce) identify these to be of structural type Mn12Th (space group I4/mmm). Analytical electron microscopy yields an overall composition of Mg12(LaxCe1−x) with x ̃0.43, with 1 at.% Zn incorporated within the lattice. Variations in characteristic X-ray emission rates, as an electron beam is rocked near zone-axis orientations, are used to form two-dimensional channelling patterns, termed X-ray incoherent channelling patterns. This channelling contrast enables a specific sublattice site that is occupied by Zn to be unambiguously identified within the Mg12RE lattice. The particular sublattice site is denoted by the Wyckoff letter f, and is one of the three different Mg sublattice sites f, i and j. Of these three sites, the Wigner–Seitz cell that is centred on the f sublattice site has the largest Mg–RE interatomic distance, and therefore the f site is expected to be favoured for accommodating the substitution of a larger Zn atom.

    Notes: Evidence is presented that transcription of most of the early genes in the Streptomyces coelicolor A3(2) phage φC31 is from a series of unusual promoters that depend on a function expressed early in the φC31 lytic cycle. Primer extension analysis on the 5′ ends of three early mRNAs, from samples prepared 10min after induction of a thermosensitive φC31 lysogen, showed that the 5′ ends all mapped close to highly similar sequences, which are proposed to be an important part of phage-specific promoters. In a shotgun cloning experiment, a fragment containing one of these sequences strongly activated transcription of the xylE reporter gene in plaques of a φC31-derived promoter-probe vector. Another of the sequences was inserted into a xylE-containing promoter-probe plasmsid vector, and promoted xylE expression only when the host was supporting the lytic cycle of φ C31. This suggested that a transcription factor needed for activity of the promoters was present only in φC31 -infected cells. Examination of published and unpublished φ C31 sequence data revealed several more sequences that closely resemble the conserved region of the characterized promoters. Most of these are found in positions close to apparent transcription start sites mapped previously by low-resolution S1 mapping. An overall consensus sequence for the conserved region suggests a general organization (though not a primary sequence) resembling that of promoters recognized in other bacteria by the σ;54 form of RNA polymerase.

    Notes: An AlGaInP-GaInP strained-layer quantum well laser structure emitting at 670 nm is pumped by a 7–20 keV scanning electron beam. At room temperature, a threshold power density as low as 57 W/mm2 is obtained, which is only 3.8× the value for diode laser operation. More than 250 points are resolved on a 5 mm bar. They are scanned at a speed of 1000 m/s at an output level of 20 mW.
