    Notes: Abstract A chitinase gene (chiA) from Pseudomonas sp. YHS-A2 was cloned into Escherichia coli using pUC19. The nucleotide sequence determination revealed a single open reading frame of chiA comprised of 1902 nucleotide base pairs and 633 deduced amino acids with a molecular weight of 67,452 Da. Amino acid sequence alignment showed that ChiA contains two putative chitin-binding domains and a single catalytic domain. Two proline-threonine repeat regions, which are linkers between catalytic and substrate-binding domains in some cellulases and xylanases, were also found. From E. coli, ChiA was purified 12.8-fold relative to the periplasmic fraction. The Michaelis constant and maximum initial velocity for p-nitrophenyl-N,N′-diacetylchitobiose were 1.06 mM and 44.4 μmol/h per mg protein, respectively. The purified ChiA binds not only to colloidal chitin but also to other substrates (avicel, chitosan, and xylan), but the binding affinity of avicel, chitosan, and xylan is around 10 times lower than that of colloidal chitin. The reaction of ChiA with colloidal chitin and chitooligosaccharides (trimer-hexamer) produced an end product of N,N′-diacetylchitobiose, indicating that ChiA is a chitobiosidase.

    Notes: An experimental H2O2/browning inhibitor wash treatment and its effect on mushroom structure and composition were studied. Experimentally washed mushrooms (Agaricus bisporus) were compared with conventionally washed mushrooms and untreated controls. Examination by scanning electron microscopy showed damage to hyphae producing a matted appearance at the pileus surface with both experimental and conventional washed samples. Mushrooms after the experimental wash had an elevated sodium content from the sodium erythorbate browning inhibitor but contained no H2O2 residue. In pileus tissue, soluble phenol levels were higher and the content of free amino acids was lower in mushrooms after the experimental wash. No other notable compositional differences or adverse effects of treatment on quality were found.

    Notes: We report optical characteristics of linear graded InxGa1−xAs (XIn=0–0.58) buffer layers grown on GaAs by low-pressure metalorganic chemical vapor deposition. Two types of wirelike surface structures were observed from the layers grown at two different temperatures. Low-temperature photoluminescence (PL) and double-crystal x-ray diffractometric measurements indicate that the PL energy and the relaxation of the graded layers were strongly dependent on the top surface structure. InGaAs cap layers were grown on top of the graded buffer layers with a variation of indium composition. A strong PL signal was observed from the top region of the graded layer grown with a lattice-matched cap layer. It suggests that the top region of the grade, similar to a graded well structure, is compressively strained but is of high structural quality without dislocations. © 1996 American Institute of Physics.

    Notes: Background Aloe vera has been used as a family medicine for promoting wound healing, but it is not known which component of the plant is effective for this purpose. Objectives To isolate and characterize the component effective in wound healing. Methods Chromatography, electrophoresis and spectroscopic methods were used. The cell-proliferation activity of each component isolated was measured by a [3H]thymidine uptake assay. The cell-proliferation activity of the effective component was tested on a three-dimensional raft culture (cell culture technique by which artificial epidermis is made from keratinocytes). The effect of the active component on cell migration and wound healing was observed on a monolayer of human keratinocytes and in hairless mice. Results A glycoprotein fraction was isolated and named G1G1M1DI2. It showed a single band on sodium dodecyl sulphate–polyacrylamide gel electrophoresis, with an apparent molecular weight of about 5·5 kDa. It exhibited significant [3H]thymidine uptake in squamous cell carcinoma cells. The effect of G1G1M1DI2 on cell migration was confirmed by accelerated wound healing on a monolayer of human keratinocytes. When this fraction was tested on a raft culture, it stimulated the formation of epidermal tissue. Furthermore, proliferation markers (epidermal growth factor receptor, fibronectin receptor, fibronectin, keratin 5/14 and keratin 1/10) were markedly expressed at the immunohistochemical level. The glycoprotein fraction enhanced wound healing in hairless mice by day 8 after injury, with significant cell proliferation. Conclusions It is considered that this glycoprotein fraction is involved in the wound-healing effect of aloe vera via cell proliferation and migration.

    Notes: Background:  Glutamate activates the N-methyl-d-aspartate (NMDA) receptors and this receptor is involved in the proliferation and migration of various tumour cells in vitro. However, the relationship between NMDA receptor expression and clinical parameters in cancer patients is unclear. Therefore, NMDA receptor 1 (NMDAR1) expression along with its clinical significance was examined in patients with oral squamous cell carcinoma (OSCC).Methods:  Eighty-one tumour specimens from OSCC patients were used to determine the NMDAR1 expression level by immunohistochemical staining. The control was obtained from a matched normal adjacent mucosa. The cases were considered to be positive if reactivity was displayed in 〉25% of the cells.Results:  The NMDAR1 reactivity was positive in 50 of 81 cases, while it was negative in the control. NMDAR1 expression was significantly associated with a lymph node metastasis (P = 0.008), the tumour size (P 〈 0.001), and the cancer stage (P = 0.034). The patients whose tumours expressed NMDAR1 had a significantly poorer survival than the patients who were NMDAR1-negative (log-rank = 6.45, d.f. = 1, P = 0.011).Conclusions:  The NMDAR1 overexpression was significantly associated with the prognosis-related factors. Therefore, it might be one of the prognostic markers of OSCC.

    Notes: Abstract A modified elliptic low-pass filter function with progressively diminishing ripples in both the passband and the stopband is proposed and analyzed. The modified elliptic function is realizable by the passive doubly-terminated ladder network for the order n even or odd, thus lending themselves amenable to high-quality low-sensitivity active RC or switched capacitor filters through the simulation techniques. Besides the passive ladder network realization, the modified elliptic function has improved characteristics both in the frequency domain and the time domain as compared with the classical elliptic function. In addition, the modified function exhibits better magnitude sensitivity properties than the classical function.
