    Description / Table of Contents: Summary The study of the DNA replication pattern in a case of a 47,XYY-karyotype revealed a marked asynchrony of both presumable Y chromosomes. This finding is most likely an expression either of a functional difference or even of a different origin. In this latter context the derivation of the extra chromosome from a deleted supernumerary X-chromosom is discussed at least for part of the XYY-cases.
    Notes: Zusammenfassung Die autoradiographische Analyse des DNS-Replikationsmusters bei einem Mann mit „XYY“-Karyotypus zeigte einen qualitativ deutlich erfaßbaren Unterschied beider Y-Chromosomen. Dieser Befund wird als Ausdruck einer besonderen Zustands- oder Funktionsform des überzähligen Chromosoms gegenüber dem „autochthonen“ Y-Chromosom angesehen. Zumindest für einen Teil der Fälle eines „XYY“-Status wird darüber hinaus auch die Möglichkeit erörtert, daß das überzählige Y-ähnliche Chromosom dem kurzen Arm eines deletierten überzähligen X-Chromosoms entspricht.

    Description / Table of Contents: Summary The clinical, histological and cytogenetic findings of 9 patients 19–28 years after Thorotrast administration are presented. The patients complaints were fatigue and epigastric distress. One patient had lymphosarcomatosis. In 4 patients the only hematologic finding was a low red blood cell count. In 6 patients a needle biopsy of the liver was performed. In all of them Thorotrast was detectable on microscopic and autoradiographic examination. Only two patients had cellular infiltration and fibrosis of the liver. In all of 4 bone marrow biopsies Thorotrast was detectable. Twice a significant increase of fatty marrow was seen. Lymphocyte cultures from peripheral blood showed abnormal chromosomes in all of the 9 patients, while there were no chromosomal abnormalities in bone marrow cells among the 4 patients examined. Possible explanations for this difference between peripheral and bone marrow cells are discussed.
    Notes: Zusammenfassung Es werden die Befunde von 9 Patienten mit Thorotrastschaden dargestellt. Die Latenzzeit betrug 19–28 Jahre. Subjektive Beschwerden bestanden in allgemeiner Müdigkeit und Abgeschlagenheit sowie in Oberbauchbeschwerden. Ein Patient hatte eine Lymphosarkomatose. Außer einer Anämie wurden bei 4 weiteren Patienten keine Blutbildveränderungen beobachtet. In allen Leberpunktaten der 6 untersuchten Patienten war histologisch und autoradiographisch Thorotrast nachweisbar. Ausgeprägte entzündliche Veränderungen mit Fibrose lagen nur bei 2 Patienten vor, bei 2 weiteren Patienten geringgradige Leberschäden. Bei 4 untersuchten Knochenmarkbiopsien wurde Thorotrast in allen Schnitten gefunden. Dabei wurde zweimal eine deutliche Fettmarkvermehrung beobachtet. Lymphocytenkulturen aus Venenblut ergaben bei allen 9 Patienten deutliche Chromosomenschäden, während bei 4 dahingehend untersuchten Patienten in den Knochenmarkzellen keine Chromosomenanomalien nachweisbar waren. Die möglichen Ursachen dieses unterschiedlichen Verhaltens werden erörtert.

    Description / Table of Contents: Zusammenfassung Die Vermutung, daß die Insektivoren in besonderem Maße Strukturvariationen der Chromosomen im Prozeß der Evolution der Arten widerspiegeln, war Ausgangspunkt dieser Arbeit. Die Chromosomenuntersuchungen bei Erinaceus europaeus (Westigel) und E. roumanicus (Ostigel) sowie bei Hemiechinus auritus und H. megalous zeigten, daß alle vier Formen die gleiche Chromosomenzahl von 48 besitzen. H. auritus und H. megalotis haben einen identischen Karyotyp und das gleiche DNS-Replikationsmuster. Der Befund kann als Beleg für die Annahme gelten, daß beide einer gemeinsamen Art zugehören und H. megalotis nur eine Großform von H. auritus darstellt. Beide Arten der europäischen Igel (Erinaceus) besitzen, anders als Hemiechinus, große, spät replizierende und in der Meiose heteropyknotische Segmente, die als autosomales Heterochromatin aufgefaßt werden. Die sehr auffälligen Unterschiede der Karyotypen betreffen überwiegend diese heterochromatischen Segmente des Genoms, die möglicherweise besonders günstige Voraussetzungen für strukturelle Umschichtungen im Laufe der Evolution boten. Vermutlich stellt das autosomale Heterochromatin einen genetisch inaktiven Anteil des Chromosomenkomplements dar. Seine Umlagerung im Laufe der Evolution ließ in cytologischer Hinsicht grobe Variationen des Genoms zu, ohne daß der genetisch aktive euchromatische Bestandteil des Genoms in gleichem Ausmaße betroffen wurde. Die beiden europäischen Igelarten sind dafür ein deutliches Beispiel.
    Notes: Abstract The chromosome variations found in the two European forms of Erinaceus have been studied by the use of 3H-Thymidine autoradiography in karyotype analyses and of meiotic preparations. The observations in Erinaceus are compared with the results of two forms of Hemiechinus. — The Western and the Eastern European hedgehogs (Erinaceus) have 48 chromosomes. Marked karyotypic differences corroborate the view that they represent different species, i.e. E. europaeus and E. roumanicus. Both possess large amounts of late labelling autosomal heterochromatin which corresponds to heteropyknotic non-pairing segments in meiosis and to large chromocenters in the interphase nuclei. — Obviously, the prominent variations of the chromosomes in E. europaeus and E. roumanicus concern the heterochromatic autosomal segments which probably represent genetically inactivated material. It is proposed that the gross karyotypic differences result from translocations, and possibly pericentric inversions, of heterochromatin and, therefore, are of lesser genetic importance. The genetically more effective changes in speciation, however, must mainly be caused by minor rearrangements or gene mutations. — H. auritus and H. megalotis have identical karyotypes (also with 48 chromosomes) and similar DNA-replication patterns. Larger heterochromatic segments are absent from mitotic and meiotic chromosomes. The lack of karyotype differences is in line with biometric data leading to consider H. megalotis as a subspecies of H. auritus. — It is conceivable that heterochromatinization as cytologic mechanism of karyotype evolution is not restricted to the European hedgehogs (Erinaceus). Other mammalian species displaying similar features must, however, be found in order to prove a more general importance of this principle.

    Description / Table of Contents: Zusammenfassung Bereits 42 Tage nach Diäthylnitrosamin-Gabe an Ratten wurden gutartige Hepatome beobachtet. Der Einbau von3H-Phenylalanin wurde dann autoradiographisch und radiochemisch untersucht. Dieakute Gabe von Cycloheximid (CHX) ruft eine starke einschränkung des3H-Phenylalanin-Einbaus in den normalen und den cancerisierten Leberzellen (gutartige Hepatome und basophile Knoten) hervor. Diechronische CHX-Gabe hat ebenfalls eine hemmende Wirkung auf den3H-Phenylalanin-Einbau der normalen Leberzellen, aber kaum einen Einfluß auf die Inkorporation der Aminosäure in den cancerisierten Leberzellen. Die autoradiographischen Ergebnisse zeigen, daß durch CHX der Eiweiß-Stoffwechsel des Kernes stärker beeinflußt wird als der des Cytoplasma. Dies gilt für die normalen wie für die cancerisierten Leberzellen.
    Notes: Summary After forty-two days of treatment with diethylnitrosamine, the first hepatomas appeared. The incorporation of3H-phenylalanine was measured autoradiographically and radiochemically. The results showed that the acute administration of CHX produced a very striking inhibition of the3H-phenylalanine incorporation by the normal and neoplastic liver cells (benign hepatoma and basophilic nodules). Chronic treatment with CHX also inhibited the uptake of3H-phenylalanine by the normal liver cells; however, the incorporation by the neoplastic cells was scarcely disturbed. The autoradiographic results showed that the protein synthesis of the nucleus was more affected than that of the cytoplasma; the effect was evident in both normal and neoplastic liver cells.

    Notes: Summary Normal tissue as well as various benign and malignant lesions of the breast were histochemically examined for the presence of the Thomsen-Friedenreich (TF)-antigen. Fluorescein- or 3H-labelled peanut agglutinin was used for this purpose, a lectin that is known to have a high affinity for the TF-antigen. The occurrence of this TF-antigen seemed in all cases, even in the carcinoma lobulare in situ that is regarded as being derived from myoepithelial cells by some authors, to be associated with a secretory condition. Its presence (free and neuraminicacid covered) in normal, hyperplastic and malignant breast tissue, however, cannot be considered a specific tumour associated antigen as has been previously assumed. Furthermore the investigations have shown that the intensity of fluorescence for peanut agglutinin (PNA)-receptors was generally stronger in differentiated carcinomas than in undifferentiated carcinomas of the breast. The histochemical findings are discussed with regard to diagnostical and immunotherapeutical aspects.

    Notes: Summary Serum levels of antibodies against the Thomsen-Friedenreich (TF) antigen were determined for a group of patients with malignancies of the breast, as well as a control group of healthy persons and patients with no breast disease. Despite recent claims of anti-TF levels being severely depressed in patients with breast cancer, we were unable to show any significant differences between the two groups. Standardised anti-TF containing sera or solutions of the TF specific agglutinin from peanuts (PNA) were absorbed with membrane preparations from normal and cancerous breast tissue, but again no differences in absorption were noted. Histological examination, by contrast, showed both free and sialic acid-covered receptors to be present on both normal and malignant breast epithelium. Based on these findings it was concluded that determination of serum anti-TF is an unsatisfactory method of diagnosing breast cancer and that the TF antigen cannot be considered to be carcinoma associated.
