    Notes: Abstract We have analyzed the allele frequency distribution at the highly polymorphic variable number of tandem repeat (VNTR) locus D1S80 (pMCT118) in seven ethnic populations (namely, New Guinea Highlanders of Papua New Guinea, Dogrib Indians of Canada, Pehuenche Indians of Chile, American and Western Samoans, Kacharis of Northeast India, and German Caucasians) using the polymerase chain reaction (PCR) technique. In the pooled sample of 443 unrelated individuals 20 segregating alleles were detected. A trimodal pattern of allelic distribution is present in the majority of populations and is indicative of the evolutionary antiquity of the polymorphism at this locus. In spite of the observed high degree of polymorphism (expected heterozygosity 56%–86%), with a single exception — the marginally significant P value (0.04) of the exact test in American Samoans — the genotype distributions in all populations conform to their respective Hardy-Weinberg expectations. Summary statistics indicate that, in general, the allele frequency distribution at this locus may be approximated by the infinite allele model. The data also demonstrate that alleles that are shared by all populations have the highest average frequency within populations. Furthermore, the kinship bioassay analysis demonstrates that the extensive variation observed at the D1S80 locus is at the interindividual within population level, which dwarfs any interpopulation allele frequency variation, consistent with the population dynamics of hypervariable polymorphisms. These characteristics of the D1S80 locus make it a very useful marker for population genetic research, genetic linkage studies, forensic identification of individuals, and for determination of biological relatedness of individuals.

    Notes: Summary Substructured populations exhibit an overall deficiency of heterozygosity whose proportional magnitude depends on the nature of substructuring, i.e., the number of subpopulations (s), their time of divergence (t) from the ancestral population, and the rate of gene flow amongst them (m). Since apparent heterozygote deficiency could be caused by many factors other than population substructuring, one must examine the nature of substructuring that could produce the observed extent of heterozygote deficiency, in order to infer the substructuring from an observed heterozygote deficiency. Using the equivalence of proportional heterozygote deficiency and the coefficient of gene differentiation (G ST), we can generate isolines of G ST as functions of s, t (in units of 2N e generations, N e being the effective population size) and m. Analytical results suggest that large G ST values cannot be reached by substructuring alone, unless the number of subpopulations are large and they remain isolated over a long period of time. Application of the theory to population data on six variable number of tandem repeats (VNTR) loci in US Caucasians and US Blacks demonstrates that the observed heterozygote deficiencies at these loci cannot be explained by substructuring within these populations alone. This is so because such large values of G ST (3%–10%) would require an absence of gene exchange between the subpopulations and a divergence time from each other of at least 25000 years ago, neither of which is compatible with the demography and ethnohistory of US Caucasians and Blacks. In contrast, the inability to detect extreme-sized alleles and/or incomplete resolution of nearly similar-sized alleles following Southern gel electrophoresis could easily explain the observed heterozygote deficiencies. The implications of these results are discussed in the context of the forensic use of DNA-typing data, and justify the employment of population genetic principles in forensic genetics.

    Notes: The effect of second-phase particles on the grain refinement of AZ61 and AZ91 Mg-Al-Znalloys with different volume fractions of β-Mg17Al12 phase particles during equal channel angularextrusion (ECAE) has been investigated. The microstructure were observed by optical microscopy(OM), scanning electron microscopy (SEM) and transmission electron microscopy (TEM), whichrevealed that grain refinement was enhanced by second phase particles at initial stage of ECAE. Andfiner grains with the high angle grain boundaries (HAGBs) and disperse second-phase particles couldbe obtained in AZ61 and AZ91 after 8 passes of ECAE

    Notes: In-plane bending is an advanced plastic processing technology which controls the unevencompressing plastic deformation of strip metal harmoniously and forces the strip metal to producein-plane bending and to form an open circle. In-plane bending is divided into incremental andcontinuous process according to the loading manner. While much attention has been paid to thecontinuous in-plane bending techniques and achievements have been made at home and abroad, onlylimited experimental investigation has so for been done on incremental in-plane bending by overseasscholars. In this paper, a 3D finite element simulation based on the rigid-plastic principle is performedfor incremental in-plane bending of pure aluminum sheet with width of 25 ~ 27.5 mm, thickness of1.5 ~ 3 mm. Influences of technical parameters such as punch inclined angle αp, punch indentation s,thickness of strip t0, width of strip w0 and strip pitch p on the strip bending radius Rin are studied. Theachievement of this study enriches the uneven deformation theory. It lays a theoretical foundation forgeneralization and industrialization of incremental in-plane bending and offers a new idea to theresearch and development of the advanced plastic processing technologies

    Notes: The effect of second phase on the mechanical properties of AZ61 and AZ91 Mg-Al-Znmagnesium alloy processed by equal-channel angular extrusion (ECAE) has been investigated. Theresults revealed that cracks initiated at and propagated through the α-Mg/β-Mg17Al12 interfaces andgrain boundaries of Mg-Al-Zn alloys during tensile deformation. For AZ61, the elongation increasedwith the increase of ECAE pass number and the decrease of grain size. However, the elongation ofAZ91 with more second phase particles decreased with the increase of ECAE pass number and thedecrease of grain size

    Notes: An NAD+-dependent succinic semialdehyde dehydrogenase from bovine brain was inactivated by pyridoxal-5′- phosphate. Spectral evidence is presented to indicate that the inactivation proceeds through formation of a Schiff's base with amino groups of the enzyme. After NaBH4 reduction of the pyridoxal-5′-phosphate inactivated enzyme, it was observed that 3.8 mol phosphopyridoxyl residues were incorporated/enzyme tetramer. The coenzyme, NAD+, protected the enzyme against inactivation by pyridoxal-5′-phosphate. The absorption spectrum of the reduced and dialyzed pyridoxal-5′-phosphate-inactivated enzyme showed a characteristic peak at 325 nm, which was absent in the spectrum of the native enzyme. The fluorescence spectrum of the pyridoxyl enzyme differs completely from that of the native enzyme. After tryptic digestion of the enzyme modified with pyridoxal-5′-phosphate followed by [3H]NaBH4 reduction, a radioactive peptide absorbing at 210 nm was isolated by reverse-phase HPLC. The sequences of the peptide containing the phosphopyridoxyllysine were clearly identical to sequences of other mammalian succinic semialdehyde dehydrogenase brain species including human. It is suggested that the catalytic function of succinic semialdehyde dehydrogenase is modulated by binding of pyridoxal-5′-phosphate to specific Lys347 residue at or near the coenzyme-binding site of the protein.
